Antihypertensive efficacy of extract of Hedera helix in high salt-induced hypertensive Sprague-Dawley rats

Objective: To explore the antihypertensive effect of extracts from the leaves of Hedera helix(H. helix) on normotensive and hypertensive rats in-vivo followed by vasodilatory studies ***: The crude methanolic extract was prepared and the activity directed fractionation was carried out. Spectrophotometric analysis of total phenolic and flavonoid content was also done. HPLC analysis was performed for the detection of hederacoside C. In-vivo blood pressure study was carried out in normotensive and high salt-induced hypertensive SpragueDawley rats. Isolated aortic tissues from rat and rabbit were used for in-vitro studies. The effects were recorded and analyzed through PowerL ab data acquisition system. Results: Crude extract of H. helix(1-30 mg/kg) decreased blood pressure to greater extent in high salt-induced hypertensive rats in-vivo compared to the normotensive [Max. fall(58.59±0.02) mm Hg vs.(67.53±3.07) mmH g]. The n-hexane, chloroform, ethyl acetate and aqueous fractions were also checked. These fractions were more effective in hypertensive rats. Aqueous fraction was more potent and n-hexane the least. In isolated rat aortic rings precontracted with phenylephrine, crude extract induced endothelium-dependent effect. The endothelium-dependent component of vasodilatory effect was ablated with L-NAME, and denudation of endothelium. The aqueous fraction was most potent vasodilator. In aortic rings from hypertensive rats, extract and fractions produced partial endothelium-independent effect which was not affected by pretreatment with L-NAME, indicating endothelium dysfunction in the hypertensive rats and suggesting additional vasodilatory mechanisms. In rabbit aorta, the extract and fractions also inhibited phenylephrine and high K^+-induced precontractions, and shifted Ca^(++) concentration–response curves. Conclusions: Our findings indicate that extract and fractions of H. helix are antihypertensive remedies, which is the outcome of vasodilatory effect.

Biocontrol of Rhizoctonia solani K1 by Iturin A Producer Bacillus subtilis RB14 Seed Treatment in Tomato Plants

Bacillus subtilis RB14 was used as an antagonist against fungal pathogen Rhizoctonia solani K1 to control damping-off diseases in tomato plants. Tomato seeds were treated with B. subtilis RB14 culture. The concentration of bacterial cells for the treatment was about 108 cfu/ml. Treated tomato seeds showed 99% germination index similar to the untreated seeds. Scanning Electron Microscopic observations showed a clear evidence of the presence of B. subtilis RB14 on tomato seed surface. Clear inhibition zone was observed using treated seed in dual plate assay against R. solani K1. B. subtilis RB14 treated seed showed 80% reduction in disease incidence during in vivo plant experiments. B. subtilis RB14 produces lipopeptide antifungal antibiotic iturin A which could suppress R. solani K1. The phenomenon was supported by our observation where we found significant amount of iturin A from the root zone soil of the seed treated plants.

目的：这项研究是对既往通过目前所提出的一种新的治疗方法对重度宫腔粘连治疗后,观察其治疗效果的一个回顾性的研究。方法：根据1988年美国生育协会提出的宫腔粘连分类标准,选择的60例分为Ⅲ级的重度宫腔粘连患者。60例患者均在中南大学湘雅三医院妇科经宫腔镜直视下利用剪刀或针状电极分离粘连术后,宫腔内留置双通道Foleys导尿管、透明质酸凝胶及宫型避孕环。在第一次的宫腔粘连分离术的1个月后行宫腔镜检术及粘连松解术。2个月后再次行宫腔镜检,再次评定粘连等级。其中有54例患者经过2次宫腔镜手术,34例经过3次宫腔镜检,17例经过4次宫腔镜检,11例经过5次宫腔镜检。有1例患者在2次宫腔镜术后没有继续随访治疗。其效果评定是根据每个患者在首次宫腔镜下粘连接松解术后3个月,与第一次手术时宫腔粘连情况作比较。结果：60例患者宫腔粘连等级,在首次粘连松解术后第一个月明显降低。粘连等级的降低说明对于重度粘连患者采用术后宫腔内留置Foleys导尿管、透明质酸钠凝胶及避孕环有治疗意义。所有患者在经过完整的治疗后,其中2例为中度粘连,1例为轻度粘连,56例恢复为正常宫腔。56例恢复正常的患者中,其中1例只经过5次宫腔镜复查,20例经过2次复查,31例经过3-5次的复查。结论：本课题的研究表明,60例闭经或月经量明显减少的重度宫腔粘连患者,在首次宫腔粘连松解术后一个月再次行宫腔镜检,可以有效的减少粘连的复发。术后的宫腔镜复查对于降低宫腔粘连患者的粘连等级有重要的价值,并且不会增加盆腔感染的风险。

Production of iturin A through glass column reactor (GCR) from soybean curd residue (okara) by <i>Bacillus subtilis</i>RB14-CS under solid state fermentation (SSF)

The present study was conducted with an aim to scale up the production of iturin A using soybean curd residue (okara). Iturin A was produced by indigenous bacterial strain Bacillus subtilis RB14-CS through glass column reactor (GCR) under solid state fermentation (SSF) was characterized. The enhanced iturin A production was observed with respect to enhanced substrate bed height when SSF was conducted in Erlenmeyer flask. To check the effect of substrate bed height on iturin A production under SSF of okara, GCR was introduced. Substrate bed height of 15 cm was suitable for iturin A production which was about 2700 mg/kg wet substrate. The observed iturin A production by the aerobic bacteria Bacillus subtilis in nearly anaerobic condition in such high substrate bed for SSF is a wonderful finding for development of SSF system in future.

目的探讨宫腔镜在临床上鉴别诊断宫角妊娠和输卵管间质部妊娠中的应用价值。方法总结分析2007年5月～2010年5月该院收治的临床拟诊宫角妊娠或输卵管间质部妊娠、但超声检查不能明确诊断的66例患者临床资料。结果 66例患者中,经宫腔镜检查协助明确诊断为输卵管间质部妊娠25例、宫角妊娠41例;宫角妊娠均在宫腔镜下手术。输卵管间质部妊娠25例中,23例在腹腔镜下手术切除病灶和患侧输卵管,2例因为技术和条件限制行开腹病灶切除及患侧输卵管切除。所有患者术后均痊愈出院。结论在临床上,当超声检查难以鉴别宫角妊娠还是输卵管间质部妊娠时,宫腔镜检查不仅可协助明确诊断,并可同时治疗宫角妊娠;且具有简单、安全、微创等优势。

Nutritional, Phytochemicals and Antioxidant Properties of Some Popular Pulse Varieties of Bangladesh

In this study, we estimate the proximate compositions, phytochemicals (polyphenol, flavonoids, flavonol, tannin, protein, carbohydrate, reducing sugar, and β carotene), antioxidant activities, vitamins, minerals, and heavy metals of the four pulses (mung, anchor, chickpea, lentils) and compare among them to find out more nutritious pulse samples. Mung was found to contain the highest amount of polyphenol (98.02 ± 1.74 mg GAE/100 g) and tannin (447.98 ± 9.96 mg TE/100 g) and anchor (771.35 ± 3.76 mg CE/100 g) was rich in flavonoids as compared to other two pulse samples. Mung was also rich in ash, carbohydrate, vitamin B1, copper and anchor was rich in crude fiber, protein, reducing sugar, and vitamin B2 content. The highest amount of phytochemicals contained in mung and anchor corresponded to its highest antioxidant activity in analyzed antioxidant assays respectively. Other two pulses included in this study were found to contain good source of vitamins, minerals and other nutrients.

Investigation of Antibacterial, Cytotoxic and Antioxidant Properties of the Mangrove Plant Xylocarpus mekongensis

The plant (Xylocarpus mekongensis) of the Sundarbans mangrove origin was evaluated for its antibacterial, cytotoxic and antioxidant properties using methanolic and chloroformic leaf, stem and bark extracts, respectively. The methanolic extracts contained higher amount of total phenolics, flavonoids, tannins than the chloroformic extracts and the result was in correlation with their ferric reducing power ability as well. However, the chloroformic bark extract contained more potent DPPH free radical scavenging activity than others. Antibacterial activity of the extracts was determined against both Gram-positive (Micrococcus and Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa, Vibrio cholerae, Salmonella typhimurium and Salmonella paratyphi) by disc diffusion assay and their zone of inhibitions (ZOI) were measured. Moreover, their minimum inhibitory concentrations (MIC) were determined by tube dilution method. Chloroformic bark and stem extracts showed strong inhibition to growth of P. aeruginosa (ZOI = 19 mm and MIC = 150 μg/ml) and S. aureus (ZOI = 19.5 mm and MIC = 250 μg/ml), respectively. All six extracts were subjected to brine shrimp lethality bioassay for possible measure of cytotoxicity. Concentration dependent increment in percentage mortality of brine Shrimp nauplii produced by the extracts indicated the presence of cytotoxic principles in these extractives. Therefore, Xylocarpus mekongensis showed antioxidant, antibacterial and cytotoxic activities.

Objectives: We aimed to explore the diagnostic value of circulating miRNAs(miR-149, miR-208 b, miR-378, miR-424, miR-499 and miR-765) in AMI patients, and protective role of miR-208 b or miR-378 or metoprolol during H/R injury in H9c2 cell with their underlying mechanisms. Methods: 205 AMI patients and 85 healthy subjects, twelve week-old C57BL/6 mice AMI and H9c2 cells were included in this study. Total RNA was isolated from plasma and cardiac cell with TRIzol reagent. Circulating miRNAs levels were measured by quantitative real-time polymerase chain reaction. Results: We found that miR-208 b, miR-499 and miR-765 levels were significantly up-regulated by 9.6 fold, 5.2 fold, and 5.1 fold in AMI patients compared with healthy subjects(p<0.001). Plasma miR-149, miR-378 and miR-424 levels were markedly downregulated by 5.5 fold, 4.9 fold and 5 fold in AMI patients compared with controls(p<0.001). Plasma levels of these miRNAs were returned towards normal at 96 h after PCI(p<0.001). We also found that plasma miR-208 b, miR-499 and miR-765 levels were significantly elevated by 11.5 fold, 3.9 fold, and 4.7 fold, while miR-149, miR-378 and miR-424 levels were noticeably decreased by 3.7 fold, 4.1 fold and 3.8 fold in Mice AMI compared with controls(p<0.001). The ROC curves of plasma miR-208 b, miR-499, miR-765, miR-149, miR-378 and miR-424 represented a strong discrimination between AMI patients and healthy subjects, with an area under curve(AUC) of 0.974, 0.971, 0.965, 0.963 0.969 and 0.973, respectively. Conclusions: Our findings suggested that miR-208 b, miR-499, miR-765, miR-149, miR-378, miR-424 may be used as a novel and sensitive diagnostic biomarker for AMI patients. Inhibition of miR-208 b and overexpression of miR-378 were effectively protected cardiomyocyte damage during H/R injury through their potential targets NLK and TRAF6, represents a new therapeutic approach for ischemic heart disease.
