AIm To investigate the association of tumor necrosis factor alpha(TNFα)-G308 A polymorphism with different liver pathological changes in treatment-na?ve Egyptian patients infected with hepatitis C virus(HCV) genotype *** This study included 180 subjects,composed of 120 treatment-na?ve chronic HCV patients with different fibrosis grades(F0-F4) and 60 healthy controls. The TNFα-G308 A region was amplified by PCR and the different genotypes were detected by restriction fragment length polymorphism analysis. The TNFα protein was detected by enzyme-linked immunosorbent assay. The influence of different TNFα-G308 A genotypes on TNFα expression and liver disease progression were statistically analyzed. The OR and 95%CI were calculated to assess the relative risk *** Current data showed that the TNFα-G308 A SNP frequency was significantly different between controls and HCV infected patients(P = 0.001). Both the AA genotype and A allele were significantly higher in late fibrosis patients(F2-F4,n = 60) than in early fibrosis patients(F0-F1,n = 60)(P = 0.05,0.04 respectively). moreover,the GA or AA genotypes increased the TNFα serum level greater than the GG genotype(P = 0.002). The results showed a clear association between severe liver pathological conditions(inflammation,steatosis and fibrosis) and(GA + AA) genotypes(P = 0.035,0.03,0.04 respectively). The stepwise logistic regression analysis showed that the TNFα genotypes(GA + AA) were significantly associated with liver inflammation(OR = 3.776,95%CI: 1.399-10.194,P = 0.009),severe steatosis(OR = 4.49,95%CI: 1.441-14.0,P = 0.010) and fibrosis progression(OR = 2.84,95%CI: 1.080-7.472,P = 0.034). Also,the A allele was an independent risk factor for liver inflammation(P = 0.003),steatosis(P = 0.003) and fibrosis(P = 0.014). CONCLUSION TNFα SNP at nucleotide-308 represents an important genetic marker that can be used for the prognosis of different liver pathological changes in HCV infected

The need for the bacterial proteinase is rapidly growing,urging to catch a lowcost medium for the microbial fermentation,nanoparticles can play a vital role in this *** proteinase of Talaromyces purpureogenus was produced on the tubers of Helianthus tuberosus that also operated as solid support for the fermentation *** interface amongst nitrogen sources(NH_(4)Cl and yeast extract)was investigated,applying the statistical modeling of central composite design under solid-state *** optimum medium for proteinase secretion was stimulated by 979.82 mg NH_(4)Cl and 437.68 mg yeast extract per 100 g substrate,yielding 108.15 U/g *** Plackett-Burman experimental design,the nanoparticles Co,Ni and Fe were assessed as inducers for proteinase *** nanoparticles(5 ppm)were the greatest in both proteinase production by the fungus as well as an inducer of the proteolysis process by the enzyme when using faba bean straw as a proteinaceous substrate in the reaction mixture,liberating the extreme quantity of amino acids,compared with the lack of the *** findings suggest the incorporation of Co nanoparticles in both the proteinase fabrication process and during the degradation of proteinaceous materials induce proteinase *** approach could be extended to modulate the productivity and activity of similar biomolecules.

microbial infections cause complicated health influences along with bad economic *** the present investigation,three dominant seaweeds namely,Amphiroa anceps,Corallina officinalis and Sargassum filipendula were collected from different Egyptian sites at the Red Sea and mediterranean Sea during autumn *** extracts of the three algae were screened for their antibacterial activity against three pathogenic bacteria Salmonella typhiimurium,Staphylococcus aureus and Escherichia coli,in addition to in vitro antiviral activity against Rotavirus(RV),and Coxsackie virus B3(CVB3)that cause severe diseases in *** extract of ***,*** and *** inhibit *** cells by 57.1%,85.7%,and 91.4%,*** highest level of concentration of the *** extract(100μg/mL)inhibits 100%of Staphylococcus aureus cells followed by *** and *** extract which inhibit 82.5%and 75%of ***,the highest concentration of *** extract inhibits *** by 80%.The extract of *** exhibited a high antiviral effect against RV infection with TI=22 and virus titers lessened by 2.75 log TCID_(50) followed by extractions of *** with TI=18.3 and virus titers reduced by 2.5 log TCID_(50).Against CVB3 infection,the extract of *** causes the highest antiviral activity with TI=15 and reduce the viral titers by 2.5 log TCID_(50),followed by extractions of *** with TI=8.8 and inhibition of virus titers by 1.75 log TCID_(50).Extract of *** displayed the lowest antiviral effects against RV and CVB3 infection with TI=2.4 and 1.4,*** obtained results clarified that the extract of three marine seaweeds maintains a potent antimicrobial activity,making them a future promising source of new antimicrobial drugs.

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is considered the causative pathogen of coronavirus disease 2019(COVID-19)and has become an international danger to human *** respiratory transmission and symptoms are still the essential manifestations of COVID-19,the digestive system could be an unconventional or supplementary route for COVID-19 to be transmitted and manifested,most likely due to the presence of angiotensin-converting enzyme 2(ACE2)in the gastrointestinal *** addition,SARS-CoV-2 can trigger hepatic injury via direct binding to the ACE2 receptor in cholangiocytes,antibodydependent enhancement of infection,systemic inflammatory response syndrome,inflammatory cytokine storms,ischemia/reperfusion injury,and adverse events of treatment *** symptoms,including anorexia,nausea,vomiting,and diarrhea,which are unusual in patients with COVID-19,and some digestive signs may occur without other respiratory ***,SARS-CoV-2 can be found in infected patients’stool,demonstrating the likelihood of transmission through the fecal-oral *** addition,liver function should be monitored during COVID-19,particularly in more severe *** review summarizes the evidence for extra-pulmonary manifestations,mechanisms,and management of COVID-19,particularly those related to the gastrointestinal tract and liver.

Detecting COVID-19 cases as early as possible became a critical issue that must be addressed to avoid the pandemic’s additional spread and early provide the appropriate treatment to the affected *** study aimed to develop a COVID-19 diagnosis and prediction(AImDP)model that could identify patients with COVID-19 and distinguish it from other viral pneumonia signs detected in chest computed tomography(CT)*** proposed system uses convolutional neural networks(CNNs)as a deep learning technology to process hundreds of CT chest scan images and speeds up COVID-19 case prediction to facilitate its *** employed the whale optimization algorithm(WOA)to select the most relevant patient signs.A set of experiments validated AImDP *** demonstrated the superiority of AImDP in terms of the area under the curve-receiver operating characteristic(AUC-ROC)curve,positive predictive value(PPV),negative predictive rate(NPR)and negative predictive value(NPV).AImDP was applied to a dataset of hundreds of real data and CT images,and it was found to achieve 96%AUC for diagnosing COVID-19 and 98%for overall *** results showed the promising performance of AImDP for diagnosing COVID-19 when compared to other recent diagnosing and predicting models.

AIm To elucidate how high diet-induced endoplasmic reticulum-stress upregulates thioredoxin interacting protein expression in müller cells leading to retinal inflammation. mETHODS male C57Bl/J mice were fed either normal diet or 60% high fat diet for 4-8 wk. During the 4 wk study, mice received phenyl-butyric acid(PBA); endoplasmic reticulum-stress inhibitor; for 2 wk. Insulin resistance was assessed by oral glucose tolerance. Effects of palmitate-bovine serum albumin(BSA)(400 μmol/L) were examined in retinal müller glial cell line and primary müller cells isolated from wild type and thioredoxin interacting protein knock-out mice. Expression of thioredoxin interacting protein, endoplasmic reticulum-stress markers, mi R-17-5p m RNA, as well as nucleotide-binding oligomerization domain-like receptor protein(NLRP3) and IL1β protein was *** High fat diet for 8 wk induced obesity and insulin resistance evident by increases in body weight and impaired glucose tolerance. The introduction of the multi well plate miniaturisation technology with its associated automated dispensers, readers and integrated systems coupled with advances in life sciences has a propelling effect on the rate at which new potential drug molecules are discovered. The translation of these discoveries to real outcome now demands parallel approaches which allow large numbers of process options to be rapidly assessed. The engineering challenges in achieving this provide the motivation for the proposed work. In this work we used computational fluid dynamics(CFD) analysis to study flow conditions in a gas liquid contactor which has the potential to be used as a fermenter on a multi well format. The bioreactor had a working volume of 6 5 mL with the major dimensions equal to those of a single well of a 24 well plate. The 6 5 mL bioreactor was mechanically agitated and aerated by a single sparger placed beneath the bottom im.eller. Detailed numerical procedure for solving the governing flow equations is given. The CFD results are combined with population balance equations to establish the size of the bubbles and their distribution in the bioreactor. Power curves with and without aeration are provided based on the simulated results.
