Feruloylated Arabinoxylans Are Oxidatively Cross- Linked by Extracellular Maize Peroxidase but Not by Horseradish Peroxidase

Covalent cross-linking of soluble extracellular arabinoxylans in living maize cultures, which models the cross- linking of wall-bound arabinoxylans, is due to oxidation of feruloyl esters to oligoferuloyl esters and ethers. The oxidizing system responsible could be H2O2/peroxidase, O2/laccase, or reactive oxygen species acting non-enzymically, To distinguish these possibilities, we studied arabinoxylan cross-linking in vivo and in vitro. In living cultures, exogenous, soluble, extracellular, feruloylated [pentosyl-3H]arabinoxylans underwent cross-linking, beginning abruptly 8 d after sub-culture. Crosslinking was suppressed by iodide, an H2O2 scavenger, indicating dependence on endogenous H2O2. However, exogenous H2O2 did not cause precocious cross-linking, despite the constant presence of endogenous peroxidases, suggesting that younger cultures contained natural cross-linking inhibitors. Dialysed culture-filtrates cross-linked [^3H]arabinoxylans in vitro only if H2O2 was also added, indicating a peroxidase requirement. This cross-linking was highly ionic-strength-dependent. The peroxidases responsible were heat-labile, although relatively heat-stable peroxidases （assayed on o-dianisidine） were also present. Surprisingly, added horseradish peroxidase, even after heat-denaturation, blocked the arabinoxylancross-linking action of maize peroxidases, suggesting that the horseradish protein was a competing substrate for [^3H]arabinoxylan coupling. In conclusion, we show for the first time that cross-linking of extracellular arabinoxylan in living maize cultures is an action of apoplastic peroxidases, some of whose unusual properties we report.

The moderating effect of physical activity on the relationship between bullying and mental health among sexual and gender minority youth

Background'.Central arterial stiffness is a cardiovascular risk factor that can be readily affected through engagement in physical exercise training,with resistance and aerobic exercise having disparate *** the growing popularity of high-intensity cross-training(HICT),little is currently known about the effects of this mixed modality exercise stimulus on arterial ***,the purpose of this study was to characterize the arterial stiffness of habitual HICT participants *** active and sedentary controls using a cross-sectional ***'.A total of 30 participants were recruited:10 middle-aged long-term participants of HICT(CrossFit)and 20 age,sex,and height matched controls(10 recreationally active,10 sedentary).Central and peripheral pulse wave velocities were measured for the carotid-femoral and femoral-dorsalis pedis arterial *** fitness(maximal oxygen uptake,VChmax)was measured and typical exercise participation rates were self-reported for each ***'.HICT participants manifested central pulse wave velocity(PWV)(5.3±1.0 m/s,mean±SD)and VChmax(43±6 mL/kg/min)values nearly identical to active *** active groups had significantly better values than sedentary controls(7.1±1.0 m/s,p≤0.001;and 32±7 mL/kg/min,p=0.01).No differences were observed in peripheral PWV between ***'.Habitual participation in HICT exercise was not associated with increased central nor peripheral arterial ***-term HICT participants presented with similar fitness and arterial stiffness as compared with participants who practiced traditional aerobic *** to sedentary living,HICT may offer musculoskeletal and cardiovascular health benefits without negatively impacting arterial stiffness.

Genotypes and viral variants in chronic hepatitis B: A review of epidemiology and clinical relevance

The Hepatitis B Virus(HBV) has a worldwide distribu-tion and is endemic in many populations. It is constantly evolving and 10 genotypic strains have been identified with varying prevalences in different geographic regions. Numerous stable mutations in the core gene and in the su-rface gene of the HBV have also been identified in untreated HBV populations. The genotypes and viral variants have been associated with certain clinical featu-res of HBV related liver disease and Hepatocellu-lar carcinoma. For example Genotype C is associated withlater hepatitis B e antigen(HBe Ag) seroconversion, and more advanced liver disease. Genotype A is associated with a greater risk of progression to chronicity in adu-lt acqu-ired HBV infections. Genotype D is particu-larly associated with the precore mu-tation and HBe Ag negative chronic hepatitis B(CHB). The genotypes prevalent in parts of West Africa, Central and Sou-th America, E, F and H respectively, are less well stu-died. Viral variants especially the Basal Core Promotor mutation is associated with increased risk of fibrosis and cancer of the liver. Althou-gh not cu-rrently part of rou-tine clinical care, evalu-ation of genotype and viral variants may provide u-sefu-l adj.-nctive information in predicting risk abou-t liver related morbidity in patients with CHB.

Portal vein thrombosis and arterioportal shunts:Effects on tumor response after chemoembolization of hepatocellular carcinoma

AIM: To evaluate the effect of portal vein thrombosis and arterioportal shunts on local tumor response in advanced cases of unresectable hepatocellular carcinoma treated by transarterial chemoembolization. METHODS: A retrospective study included 39 patients (mean age: 66.4 years, range: 45-79 years, SD: 7) with unresectable hepatocellular carcinoma (HCC) who were treated with repetitive transarterial chemoembolization (TACE) in the period between March 2006 and October 2009. The effect of portal vein thrombosis (PVT) (in 19 out of 39 patients), the presence of arterioportal shunt (APS) (in 7 out of 39), the underlying liver pathology,Child-Pugh score, initial tumor volume, number of tumors and tumor margin definition on imaging were correlated with the local tumor response after TACE. The initial and end therapy local tumor responses were evaluated according to the response evaluation criteria in solid tumors (RECIST) and magnetic resonance imaging volumetric measurements. RESULTS: The treatment protocols were well tolerated by all patients with no maj.r complications. Local tumor response for all patients according to RECIST criteria were partial response in one patient (2.6%), stable disease in 34 patients (87.1%), and progressive disease in 4 patients (10.2%). The MR volumetric measurements showed that the PVT, APS, underlying liver pathology and tumor margin definition were statistically significant prognostic factors for the local tumor response (P = 0.018, P = 0.008, P = 0.034 and P = 0.001, respectively). The overall 6-, 12- and 18-mo survival rates from the initial TACE were 79.5%, 37.5% and 21%, respectively. CONCLUSION: TACE may be exploited safely for palliative tumor control in patients with advanced unresectable HCC; however, tumor response is significantly affected by the presence or absence of PVT and APS.

Compost Improves Urban Soil and Water Quality

Construction in urban zones compacts the soil, which hinders root growth and infiltration and may increase erosion and degrade water quality. The purpose of our study was to determine whether planting prairie grasses and adding compost to urban soils should partially mitigate these concerns. We simulated construction activities by removing the topsoil, and compacting the soil by repeatedly driving over it with a tractor. Additionally, treatments included three compost application methods (compost and aeration, rototill and compost, surface compost). Plots were subj.cted to simulated rainfall applied using overhead sprinklers at a rate of 65 or 72 mm·h-1. Bulk density was significantly reduced in surface soil where compost had been added. Compost plus prairie grasses resulted in significantly faster infiltration (63 vs. 52 mm·h-1), slower runoff (4 vs. 25 mm·h-2), less soil loss (25 vs. 119 kg·Ha-1·h-1), and reduced loss of ortho P (57 vs. 410 g·Ha-1) compared to plots planted with bluegrass (Poa pratensis L.). A 5 to 7.5 cm thick compost blanket reduced time to runoff (60 min) compared to no compost addition (9 min). Topsoil addition without compost did not develop dark surface soil. Compost additions are recommended to reduce negative effects of urban compaction. Over time, the depth of compost additions decreased, necessitating further additions.

Cyclophosphamide-associated enteritis: A rare association with severe enteritis

Cyclophosphamide is a potent cytotoxic agent used in many clinical settings. The main risks of cyclophosphamide therapy include hematological disorders, infertility, hemorrhagic cystitis and malignancies. Gastrointestinal side effects reported to date are often non-specific and not severe. We present the first case of a fatal small bowel enteritis and pan-colitis which appears to be associated with cyclophosphamide. We aim to raise the readers' awareness of this significant adverse event to facilitate clinical suspicion and early recognition in potential future cases.

Cost-effectiveness of Crohn's disease post-operative care

AIM: To define the cost-effectiveness of strategies, including endoscopy and immunosuppression, to prevent endoscopic recurrence of Crohn’s disease following intestinal ***: In the “POCER” study patients undergoing intestinal resection were treated with post-operative drug therapy. Two thirds were randomized to active care (6 mo colonoscopy and drug intensification for endoscopic recurrence) and one third to drug therapy without early endoscopy. Colonoscopy at 18 mo and faecal calprotectin (FC) measurement were used to assess disease recurrence. Administrative data, chart review and patient questionnaires were collected prospectively over 18 ***: Sixty patients (active care n = 43, standard care n = 17) were included from one health service. Median total health care cost was $6440 per patient. Active care cost $4824 more than standard care over 18 mo. Medication accounted for 78% of total cost, of which 90% was for adalimumab. Median health care cost was higher for those with endoscopic recurrence compared to those in remission [$26347 (IQR 25045-27485) vs $2729 (IQR 1182-5215), P < 0.001]. FC to select patients for colonoscopy could reduce cost by $1010 per patient over 18 mo. Active care was associated with 18% decreased endoscopic recurrence, costing $861 for each recurrence ***: Post-operative management strategies are associated with high cost, primarily medication related. Calprotectin use reduces costs. The long term cost-benefit of these strategies remains to be evaluated.
