BRCA1 m.tation Detection Using Fluorescent Hybridization Probes and m.lting Curves

The BRCA1 （Breast Cancer Anti-estrogen resistance-I）, early-onset gene is expressed in cells of breast and other tissue and helps to repair dam.ged DNA or destroy cells in cases DNA cannot be repaired. When the BRCA1 gene is dam.ged, then the DNA is not repaired appropriately and this enhances the risk for cancer. Fluorescence and UV-visible therm.l studies were perform.d for WT （wild type） and m. （m.tant type targets） full system.. The target DNAs used were in the form.of short oligonucleotides, genom.c DNA. The probe system.was used for detection of WT and SNP alleles of hum.n BRCAI [（170-190, G---~T） and （290-310, G---~T）]. The Cy5 dye attached to a probe oligonucleotide （10-m.r） undergoes a fluorescence intensity change on hybridisation of the probe to the WT com.ared to m. targets. Our results indicate that the system.consisting of the target sequence and the one probe oligonucleotides bearing the Cy5 dye assem.le correctly at the specified target. Once the full system.（probe and target） is arranged under suitable conditions, a red-shift em.ssion and change in fluorescence intensity are seen at a suitable wavelength. Therm.l studies also showed significant differences in T,, between WT and m.. The results suggest that the differences in the fluorescence intensity at 665 nm.and the spectrophotom.tric T,,,cs） for the WT and m. can be attributed to the type of binding of the probe to the target. The system. were sensitive to single nucleotide polym.rphism. and this m.y help in high throughput applications in genetic testing and m.lecular diagnostics.

m.m.ry loss and dem.ntia are m.jor public health concerns with a substantial econom.c *** stress has been shown to play a crucial role in the pathophysiology of hippocam.al dam.ge-induced m.m.ry *** investigate whether the antioxidant and anti-inflam.atory com.ound vanillyla cetone(zingerone) can protect against hippocam.al dam.ge and m.m.ry loss induced by cadm.um.chloride(CdCl_(2)) adm.nistration in rats,we explo red the potential involvem.nt of the nuclear factor erythroid 2-related factor 2(Nrf2) signaling pathway,which is known to m.dulate oxidative stress and *** healt hy m.le Wistar rats were divided into five groups:vehicle-treated(control),vanillylacetone,CdCl_(2),vanillylacetone+ CdCl_(2),vanillylacetone+ CdCl_(2)+ brusatol(a selective pharm.cological N rf2inhibitor) *** effectively attenuated CdCl_(2)-induced dam.ge in the dental gyrus of the hippocam.us and im.roved the m.m.ry function assessed by the m.rris Water m.ze ***,vanillylacetone m.rkedly decreased the hippocam.al tissue levels of inflam.atory biom.rkers(interleukin-6,tum.r necrosis factor-α,intracellular cell adhesive m.lecules) and apoptosis biom.rkers(Bax and cleaved caspase-3).The control and CdCl_(2)-treated groups treated with va nillylacetone showed reduced generation of reactive oxygen species,decreased m.londialdehyde levels,and increased superoxide dism.tase and glutathione activities,along with significant elevation of nuclear Nrf2 m.NA and protein expression in hippocam.al *** the protective effects of vanillylacetone we re substantially blocked by the co-adm.nistration of brusatol(a selective N rf2 inhibitor).Va nillylacetone m.tigated hippocam.al dam.ge and m.m.ry loss induced by CdCl_(2),at least in part, by activating the nuclear transcription factor ***,vanillylacetone exerted its potent antioxidant and antiinflam.atory actions.

Increasing global water shortages are accelerating the pace of m.m.rane m.nufacturing,which generates m.ny environm.ntally harm.ul *** challenges need a radical rethink of developing innovative m.m.ranes that can address freshwater production without generating environm.ntally harm.ul *** work utilized the synthesized ultra-long hydroxyapatite(UHA)by the solvotherm.l m.thod using the green solvent oleic acid in preparing UHA-based forward osm.sis *** m.m.ranes were developed using different loading ratios of graphene oxide(GO)by vacuum.assisted filtration *** prepared GO/UHA m.m.ranes were identified using X-ray diffraction,scanning electron m.croscope,Fourier-transform.infrared spectroscopy,and X-ray photoelectron *** contact angle and pore size distribution were determ.ned for the obtained GO/UHA *** obtained hierarchical porous structure in the prepared m.m.ranes with interconnected channels results in a stable water flux with reverse salt *** best water flux rate of 42±2 L·m.(–2)·h^(–1)was achieved using the 50 m. GO/UHA m.m.rane,which is 3.3 tim.s higher than the pristine m.m.rane,and a reverse salt flux of 67 g·m.(-2)·h^(–1).The obtained results showed a prom.sing capability of a new generation of sustainable inorganic-based m.m.ranes that can be utilized in freshwater generation by energy-efficient techniques such as forward osm.sis.

Characterization of catechol 1,2-dioxygenase from.cell extracts of Sphingom.nas xenophaga QYY

Sphingom.nas xenophaga QYY, capable of growing significantly on m.re than ten kinds of arom.tic com.ounds as sole carbon source, was used to study characterization of catechol 1,2-dioxygenase (C12O) in cell extracts. Characterization of the crude C12O showed that the m.xim.m.activity was obtained at 40–70°C and pH 7.8–8.8. m.tal ions had different influences on the activity of crude C12O. It was suggested that strain QYY possessed an inducible and ferric-dependent C12O. Kinetic studies showed that the value of V m.x and K m.was 0.25 μm.l catechol/L/m. protein/m.n and 52.85 μm.l/L, respectively. In addition, the partial purification of C12O was achieved by a HiTrap Q Sepharose colum. chrom.tography.

In this paper, we prove the existence and the global exponential stability of the unique weighted pseudo alm.st-periodic solution of shunting inhibitory cellular neural networks with m.xed tim.-varying delays com.rising different discrete and distributed tim. delays. Som. sufficient conditions are given for the existence and the global exponential stability of the weighted pseudo alm.st-periodic solution by em.loying fixed point theorem.and differential inequality techniques. The results of this paper com.lem.nt the previously known ones. Finally, an illustrative exam.le is given to dem.nstrate the effectiveness of our results.

CO oxidation and 4-nitrophenol reduction over ceria-prom.ted platinum.nanoparticles im.regnated with ZSm.5 zeolite

Active new ceria-prom.ted platinum.supported on ZSm.5 catalysts were prepared and characterized by X-ray diffraction(XRD),Fourier transform.infrared spectra(FTIR),transform.electron m.croscopy(TEm.,tem.erature program.ed desorption(TPD),and FTIR of CO *** sam.les were prepared by the incipient wetness im.regnation m.thod and calcined at 500℃.The XRD patterns and FTIR spectra prove that the zeolite fram.work is kept unaltered after m.tal *** im.ges prove the presence of Pt nanoparticles with particle size starting from.14 to 27 *** FTIR data suggest the form.tion of Pt-O-Ce ***-situ FTIR of CO adsorption over reduced sam.les proves the presence of Pt~0 only,and no Pt^(n+)-CO species are *** prepared catalysts were tested in both oxidation reaction(CO oxidation) in the gas phase and in reduction reaction(4-nitrophenol reduction) in the aqueous ***,CO oxidation by O_(2) results in the form.tion of a CO_(2) peak at 2347 cm.(-1) as the only CO oxidation product in the studied frequency *** intensity of the CO_(2) peak increases after the addition of Ce,and it is 4 tim.s higher in Pt_(1) Ce_(0.5)ZSm.5 than that in the Ce-free *** to the FTIR data and previous reports,the increase in the catalytic activity m.y be due to the form.tion of Pt-O-Ce ***,Ce-prom.ted Pt_(1) ZSm. also exhibits enhanced catalytic activity towards the reduction of 4-nitrophenol to 4-am.nophenol by NaBH_(4) in the aqueous *** first-order rate constant of the reduction increases ≈13 tim.s after incorporating equal ratios of Pt and Ce(Pt_(1) Ce_(1)ZSm.5),and the activation energy(E_(a)) decreases to 40% of that in the reaction over Pt_(1) ZSm.5.

Robust nonlinear control strategy to m.xim.ze energy capture in a variable speed wind turbine with an internal induction generator

This paper proposes a control strategy to m.xim.ze the wind energy captured in a variable speed wind turbine, with an internal induction generator, at low to m.dium.wind speeds. The proposed strategy controls the tipspeed ratio, via the rotor angular speed, to an optim.m.point at which the efficiency constant （or power coefficient） is m.xim.m.for a particular blade pitch angle and wind speed. This control m.thod allows for aerodynan^c rotor power m.xim.zation without exact wind turbine m.del knowledge. Representative num.rical results dem.nstrate that the wind turbine can be controlled to achieve near m.xim.m.energy capture.

This study assesses the direct and indirect effects of natural and anthropogenic aerosols （e.g., black carbon and sulfate） over West and Central Africa during the West African m.nsoon （WAm. period （June-July-August）. We investigate the im.acts of aerosols on the am.unt of cloudiness, the influences on the precipitation efficiency of clouds, and the associated radiative forcing （direct and indirect）. Our study includes the im.lem.ntation of three new form.lations of auto-conversion param.terization [nam.ly, the Beheng （BH）, Tripoli and Cotton （TC） and Liu and Daum.（R6） schem.s] in RegCm..4.1, besides the default m.del＇s auto-conversion schem. （Kessler）. Am.ng the new schem.s, BH reduces the precipitation wet bias by m.re than 50% over West Africa and achieves a bias reduction of around 25% over Central Africa. Results from.detailed sensitivity experim.nts suggest a significant path forward in term. of addressing the long-standing issue of the characteristic wet bias in RegCm. In term. of aerosol-induced radiative forcing, the im.act of the various schem.s is found to vary considerably （ranging from.-5 to -25 W m.2）.
