Transcriptome sequencing reveals novel biomarkers and immune cell infiltration in esophageal tumorigenesis

BACKGROUND Esophageal squamous cell carcinoma(ESCC)is one of the most common malignancies worldwide,and its development comprises a multistep process from intraepithelial neoplasia(IN)to carcinoma(CA).However,the critical regulators and underlying molecular mechanisms remain largely unknown.AIM To explore the genes and infiltrating immune cells in the microenvironment that are associated with the multistage progression of ESCC to facilitate diagnosis and early intervention.METHODS A mouse model mimicking the multistage development of ESCC was established by providing warter containing 4-nitroquinoline 1-oxide(4NQO)to C57BL/6 mice.Moreover,we established a control group without 4NQO treatment of mice.Then,transcriptome sequencing was performed for esophageal tissues from patients with different pathological statuses,including low-grade IN(LGIN),high-grade IN(HGIN),and CA,and controlled normal tissue(NOR)samples.Differentially expressed genes(DEGs)were identified in the LGIN,HGIN,and CA groups,and the biological functions of the DEGs were analyzed via Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses.The CIBERSORT algorithm was used to detect the pattern of immune cell infilt-ration.Immunohistochemistry(IHC)was also conducted to validate our results.Finally,the Luminex multiplex cytokine analysis was utilized to measure the serum cytokine levels in the mice.RESULTS Compared with those in the NOR group,a total of 681541,and 840 DEGs were obtained in the LGIN,HGIN,and CA groups,respectively.Using the intersection of the three sets of DEGs,we identified 86 genes as key genes involved in the development of ESCC.Enrichment analysis revealed that these genes were enriched mainly in the keratinization,epidermal cell differentiation,and interleukin(IL)-17 signaling pathways.CIBERSORT analysis revealed that,compared with those in the NOR group,M0 and M1 macrophages in the 4NQO group showed stronger infiltration,which was validated by IHC.Serum cytokine analysis revealed that,compared with those i

Machine learning-based radiomics score improves prognostic prediction accuracy of stage II/III gastric cancer: A multi-cohort study

BACKGROUND Although accurately evaluating the overall survival(OS)of gastric cancer patients remains difficult,radiomics is considered an important option for studying pro-gnosis.AIM To develop a robust and unbiased biomarker for predicting OS using machine learning and computed tomography(CT)image radiomics.METHODS This study included 181 stage II/III gastric cancer patients,141 from Lichuan People's Hospital,and 40 from the Cancer Imaging Archive(TCIA).Primary tumors in the preoperative unenhanced CT images were outlined as regions of interest(ROI),and approximately 1700 radiomics features were extracted from each ROI.The skeletal muscle index(SMI)and skeletal muscle density(SMD)were measured using CT images from the lower margin of the third lumbar vertebra.Using the least absolute shrinkage and selection operator regression with 5-fold cross-validation,36 radiomics features were identified as important predictors,and the OS-associated CT image radiomics score(OACRS)was cal-culated for each patient using these important predictors.RESULTS Patients with a high OACRS had a poorer prognosis than those with a low OACRS score(P<0.05)and those in the TCIA cohort.Univariate and multivariate analyses revealed that OACRS was a risk factor[RR=3.023(1.896-4.365),P<0.001]independent of SMI,SMD,and pathological features.Moreover,OACRS outperformed SMI and SMD and could improve OS prediction(P<0.05).CONCLUSION A novel biomarker based on machine learning and radiomics was developed that exhibited exceptional OS discrimination potential.

Background:Knee osteoarthritis(KOA)characterized by degeneration of knee cartilage and subsequent bone hyperplasia is a prevalent joint condition primarily affecting aging adults.The pathophysiology of KOA remains poorly understood,as it involves complex mechanisms that result in the same outcome.Consequently,researchers are interested in studying KOA and require appropriate animal models for basic research.Chinese herbal compounds,which consist of multiple herbs with diverse pharmacological properties,possess characteristics such as multicomponent,multipathway,and multitarget effects.The potential benefits in the treatment of KOA continue to attract attention.Purpose:This study aims to provide a comprehensive overview of the advantages,limitations,and specific considerations in selecting different species and methods for KOA animal models.This will help researchers make informed decisions when choosing an animal model.Methods:Online academic databases(e.g.,PubMed,Google Scholar,Web of Science,and CNKI)were searched using the search terms“knee osteoarthritis,”“animal models,”“traditional Chinese medicine,”and their combinations,primarily including KOA studies published from 2010 to 2023.Results:Based on literature retrieval,this review provides a comprehensive overview of the methods of establishing KOA animal models;introduces the current status of advantages and disadvantages of various animal models,including mice,rats,rabbits,dogs,and sheep/goats;and presents the current status of methods used to establish KOA animal models.Conclusion:This study provides a review of the animal models used in recent KOA research,discusses the common modeling methods,and emphasizes the role of traditional Chinese medicine compounds in the treatment of KOA.

Ultra-high-rate Bi anode encapsulated in 3D lignin-derived carbon framework for sodium-ion hybrid capacitors

Bismuth(Bi),as an alloy-based anode material,has attracted much atte ntion in the developme nt of sodiumion hybrid capacitors(SIHCs)due to its high theoretical capacity.However,the volume expansion of the Bi-based anode during the sodiation/desodiation process results in limited rate capability.In the present work,a porous Bi-based composite was constructed by a one-step hydrothermal method,and Bi was encapsulated in ligninderived nitrogen-doped porous carbon(Bi@LNPC)after carbonization.The obtained Bi nanoparticles could effectively adapt to the strain and shorten the diffusion distance of Na^(+).In addition,porous carbon skeleton provides a rigid conductive network for electronic transportation.Therefore,the assembled sodium-ion half-cell with Bi@LNPC anode shows ultra-high-rate capability.When the current density was enhanced from 0.1 to 50 A·g^(-1),the specific capacity decreased slightly from 351.5 to 342.8 mAh·g^(-1).Even at an extremely high current density of 200 A·g^(-1),it retains 81.3%capacity retention when compared to a current density of 1 A·g^(-1).The SIHCs assembled by Bi@LNPC show a high energy density of 63 Wh·kg^(-1).This work provides an effective method for developing high-rate Bi anode materials for sodium-ion hybrid capacitors(SIHCs)and sodium-ion batteries(SIBs).

Enhancing the Interaction of Carbon Nanotubes by Metal-Organic Decomposition with Improved Mechanical Strength and Ultra-Broadband EMI Shielding Performance

The remarkable properties of carbon nanotubes(CNTs)have led to promising applications in the field of electromagnetic inter-ference(EMI)shielding.However,for macroscopic CNT assemblies,such as CNT film,achieving high electrical and mechanical properties remains challenging,which heavily depends on the tube-tube interac-tions of CNTs.Herein,we develop a novel strategy based on metal-organic decomposition(MOD)to fabricate a flexible silver-carbon nanotube(Ag-CNT)film.The Ag particles are introduced in situ into the CNT film through annealing of MOD,leading to enhanced tube-tube interactions.As a result,the electrical conductivity of Ag-CNT film is up to 6.82×10^(5) S m^(-1),and the EMI shielding effectiveness of Ag-CNT film with a thickness of~7.8μm exceeds 66 dB in the ultra-broad frequency range(3-40 GHz).The tensile strength and Young’s modulus of Ag-CNT film increase from 30.09±3.14 to 76.06±6.20 MPa(~253%)and from 1.12±0.33 to 8.90±0.97 GPa(~795%),respectively.Moreover,the Ag-CNT film exhibits excellent near-field shield-ing performance,which can effectively block wireless transmission.This innovative approach provides an effective route to further apply macroscopic CNT assemblies to future portable and wearable electronic devices.

Constipation is common in the diseases of the digestive system in clinics.With the change in diet structure and the increase in life pressure,the prevalence rate increases year by year.In traditional Chinese medicine(TCM),the location of the disease of constipation is in the large intestine,which is related to the dysfunction of lung,spleen,liver,kidney and other viscera.Its pathogenesis is conductive dysfunction of large intestine.Based on the theory,Shouhui Tongbian Capsule(SHTB)is composed of eight traditional Chinese medicines,including Polygoni multiflori Radix(Heshouwu in Chinese),Aloe(Luhui in Chinese),Cassiae Semen(Juemingzi in Chinese),Ginseng Radix et Rhizoma(Renshen in Chinese),Lycii Fructus(Gouqizi in Chinese),Asini Corii Colla(Ejiao in Chinese),Aurantii Fructus Immaturus(Zhishi in Chinese),and Atractylodis Macrocephalae Rhizoma(Baizhu in Chinese),which could help to release excessive turbid,and nourishing yin and supplementing qi in the treatment.This study has been carried out to review the latest advances of SHTB in the treatment of constipation.The results showed that significant effect of SHTB was found in the treatment of constipation,such as functional constipation,and constipation associated with tumor chemotherapy,colitis,type 2 diabetes and chronic cardiac failure.Besides,obvious adverse reactions were not observed.SHTB could effectively treat five types of constipation,provide direction for the future exploration of SHTB in the treatment of other types of constipation。

N-glycosylation of Wnt3 regulates the progression of hepatocellular carcinoma by affecting Wnt/β-catenin signal pathway

BACKGROUND Wnt/FZD-mediated signaling pathways are activated in more than 90%of hepatocellular carcinoma(HCC)cell lines.As a well-known secretory glycoprotein,Wnt3 can interact with FZD receptors on the cell surface,thereby activating the Wnt/β-catenin signaling pathway.However,the N-glycosylation modification site of Wnt3 and the effect of this modification on the biological function of the protein are still unclear.AIM To investigate the effect of Wnt3 N-glycosylation on the biological function of HCC cells.METHODS Site-directed mutagenesis was used to verify the Wnt3 N-glycosylation sites,actinomycin D treatment was used to detect the stability of Wnt3 after site-directed mutation,the binding of the N-glycosylation site-directed mutant Wnt3 to FZD7 was observed by laser confocal microscopy,and the effects of the N-glycosylation site-directed mutation of Wnt3 on the Wnt/β-catenin signaling pathway and the progression of HCC cells were detected by western blot and cell function experiments.RESULTS Wnt3 has two N-glycosylation-modified sites(Asn90 and Asn301);when a single site at amino acid 301 is mutated,the stability of Wnt3 is weakened;the binding ability of Wnt3 to FZD7 decreases when both sites are mutated simultaneously;and the level of proteins related to the Wnt/β-catenin signaling pathway is downregulated.Cell proliferation,migration and invasion are also weakened in the case of single 301 site and double-site mutations.CONCLUSION These results indicate that by inhibiting the N-glycosylation of Wnt3,the proliferation,migration,invasion and colony formation abilities of liver cancer cells can be weakened,which might provide new therapeutic strategies for clinical liver cancer in the future.

Computerized cognitive remediation therapy on cognitive impairment and social function in patients with chronic schizophrenia

BACKGROUND Patients with schizophrenia may have various disease manifestations,most of which gradually tend toward incurable chronic decline,leading to mental disability.The basic symptoms of the disease can impair social function,whereas long-term hospitalization produces hospitalization syndrome,causing serious damage to social function.AIM To investigate the effects of Computerized Cognitive Remediation Therapy(CCRT)on cognitive and social functioning in patients with chronic schizophrenia.METHODS A retrospective analysis of 120 patients with chronic schizophrenia in Shanghai Pudong New Area Mental Health Center was performed.They were divided into an intervention group(60 cases treated with CCRT combined with conventional medication)and a control group(60 cases treated with conventional medication).After treatment,effects on cognitive function and social roles were observed in both groups.The Positive and Negative Syndrome Scale(PANSS)was used to assess the patients'psychiatric symptoms.The Wisconsin Card Sorting Test(WCST)was used to assess the patients'cognitive functioning,and the Social Functioning Scale for Psychiatric Inpatients(SSPI)was used to assess the social functioning of the inpatient psychiatric patients.RESULTS No significant differences were observed in the PANSS,WCST,and SSPI intergroup scores before treatment(P>0.05).After 2,4,and 6 wk of therapy,general psychopathological factors,positive symptoms,negative symptoms,and total PANSS scores of PANSS in the intervention group were lower than in the control group(P<0.05).After 2,4,and 6 wk of treatment,the number of false responses,number of persistent bugs,and total responses in the WCST were significantly lower in the intervention group than in the control group(P<0.05),and the amount of completed classification was significantly higher than in the control group(P<0.05).After 2,4,and 6 wk of therapy,the SSPI scores were significantly greater than those of the controls(P<0.05).After 6 wk of treatment,the efficacy rates of the control and interv

农业遗产地种质资源丰富,加强立法保护对维护生物多样性、保障粮食安全具有重要意义。本文采取法学与资源科学交叉的研究方法,通过实地调查与文献分析,探讨当前我国农业遗产地种质资源保护中的问题,在革新保护模式的基础上,探究保护制度的完善。研究表明,我国农业遗产地种质资源保护的规范主要依据相关资源环境类法律法规,种质资源保护以采取“保护单一对象的损害应对模式”为主。然而,在管理中存在保护对象要素化、保护责任单向化、保护权限碎片化、保护政策同质化等问题。基于整体主义生态观,借鉴当代生态环境治理经验,可以引入系统性保护的方法,将种质资源保护纳入农业系统生态保护全过程,建立保护整体系统的风险治理模式。在此基础上,制定《农业文化遗产保护法》。进而,细化农民主体参与激励、联席保护、名录保护、第三方协议保护等制度的运用。最终,通过法律制度的完善,为农业遗产地种质资源保护提供规范性依据。

Successful conversion of advanced gastric cancer treated by immunotherapy combined with chemotherapy: A case report

Gastric cancer is one of the most common malignant tumors,for patients with advanced gastric cancer,the traditional treatment is mainly chemotherapy or combined targeted therapy;however,these have not achieved ideal efficacy.However,with the continuous deepening of research on molecular targeted drugs and immunosuppressants,the treatment of advanced gastric cancer patients has made new progress,and some new technologies have also been continuously emerged and applied,which brings hope for the treatment of advanced gastric cancer.Recently,several multicenter randomized controlled phase III studies on immunotherapy for advanced gastric cancer yielded encouraging results,demonstrating its superior efficacy in the treatment of advanced gastric cancer.However,prospective data to support the acceptance of surgery and the R0 removal rate following conversion therapy with immune checkpoint inhibitors are lacking.In this study,a 58-year-old woman with advanced gastric cancer and positive PD-L1 expression,negative HER-2 expression,and microsatellite stable status received immunochemotherapy combined with traditional Chinese medicine to achieve R0 resection and satisfactory efficacy.
