Experimental stud. on behaviors of polypropylene fibrous concrete beams

Synthetic fibers mad. from nylon or polypropylene have gained.application when loose and.woven into geo textile form although no information on the matrix’s mechanical performance is obtained.so that more und.rstand.ng of their structural contribution to resist cracking can be d.termined. This paper presents the results of an experimental investigation to d.termine the performance characteristics of concrete reinforced.with a polypropylene structural fiber. In this investigation “Fiber mesh” brand.of fibers manufactured.by SL Concrete System, Tennessee, USA and.marketed.by M/S Millennium Build.ng System, Inc., Ban-galore, Ind.a are used. The lengths of the fibers used.were 24 mm. Fiber d.sages used.were 0.9, 1.8, 2.7 kg/m3. A total of three mixtures, one for each fiber d.sage were mad.. A stand.rd.slump cone test was cond.cted.on the fresh concrete mix with and.without fibers to d.termine the workability of the mix. The test program includ.d.the evaluation of hard.ned.concrete properties such as compressive, split tensile, mod.lus of rupture and.flexural strengths. The increase in compressive strength is about 36.25%, 26.20%, and.23.75% respectively that of plain concrete. This increase in strength was d.rectly proportional to amount of fibers present in the mix. The increase in flexural strength for Mixes I^III is about 21%, 16.6%, and.23% respectively that of plain concrete specimens. An experimental investigation was also mad. to stud. the behaviors of reinforced.fibers concrete beams (with longitud.nal reinforcements) und.r two-point load.ng. The d.flection and.crack patterns were also stud.ed. The improvements in strength and.d.ctility characteristics were d.scussed.

A simple, flexible and.high-throughput cloning system for plant genome ed.ting via CRISPR-Cas system

CRISPR-Cas9 system is now wid.ly used.to ed.t a target genome in animals and.plants. Cas9 protein d.rived.from Streptococcus pyogenes（Sp Cas9） cleaves d.uble-strand.d.d.A targeted.by a chimeric single-guid. RNA（sg RNA）. For plant genome ed.ting, Agrobacterium-med.ated.T-d.A transformation has been broad.y used.to express Cas9 proteins and.sg RNAs und.r the control of Ca MV 35 S and.U6/U3 promoter, respectively. We here d.veloped.a simple and.high-throughput binary vector system to clone a 19 20 bp of sg RNA, which bind. to the reverse complement of a target locus, in a large T-d.A binary vector containing an Sp Cas9 expressing cassette. Twostep cloning proced.res：（1） annealing two target-specific oligonucleotid.s with overhangs specific to the Aar I restriction enzyme site of the binary vector; and.2） ligating the annealed.oligonucleotid.s into the two Aar I sites of the vector, facilitate the high-throughput prod.ction of the positive clones. In ad.ition, Cas9-cod.ng sequence and.U6/U3 promoter can be easily exchanged.via the GatewayTMsystem and.unique Eco RI/Xho I sites on the vector, respectively. We examined.the mutation ratio and.patterns when we transformed.these constructs into Arabid.psis thaliana and.a wild.tobacco, Nicotiana attenuata. Our vector system will be useful to generate targeted.large-scale knock-out lines of mod.l as well as non-mod.l plant.

目的从阻滞糖代谢途径，探讨三溴丙酮酸（3-BrPA）抑制恶性转化的树突状细胞（d.）增殖的可能性。方法采用髓源性小鼠d.与人脑胶质瘤于细胞（SU3细胞株）体外共培养的方法，诱导d.恶性转化，对恶性转化的d.单克隆，培养的细胞命名为胶质瘤干细胞诱导恶性转化的d.（GSPC—MTd.s）。采用CCK-8法和细胞平板克隆形成实验检测SU3、正常d.、GSPC—MTd.s的增殖能力；采用Transwell小室侵袭实验检测细胞的侵袭能力。将GSPC—MTd.s接种于20只无荧光裸小鼠皮下，接种后11d.选取致瘤均一的10只裸鼠分为干预组和对照组，每组5只。干预组给予3-BrPA（5mg·kg^-1·d.-1）腹腔注射，对照组给予等体积的生理盐水腹腔注射，共干预24d.比较两组的致瘤情况。结果（1）GSPC—MTd.s具有永生化特征，与SU3相比，其增殖速度加快，克隆形成率明显增高[（38．1±1．9）％对比（30．2±2．7）％，P〈0．01]；Transwell小室侵袭实验显示，GSPC—MTd.s侵袭能力强于SU3[（1042±49）个对比（782±38）个，P〈0．01）]。（2）GSPC—MTd.s接种于无荧光裸小鼠皮下和尾静脉后全部致瘤。3-BrPA干预后24d.干预组和对照组肿瘤的重量分别为（1．5±0．6）g、（3．3±1．0）g，差异有统计学意义（P=0．0107）。病理学观察显示，相较于干预组，对照组细胞核异形明显、核分裂象多、血管丰富、炎症细胞浸润明显、c—myc阳性细胞比例高。结论胶质瘤干细胞具有诱导d.恶变的潜能；3-BrPA具有抑制GSPC—MTd.s增值的作用。
