Aim： To investigate the transformation of characteristics of epidermal cells from foreskin which were used to reconstruct male rabbit anterior urethra in combination with acellular collagen matrices. Methods： In three rabbits, autologous foreskin epidermal cells were isolated, expanded in vitro, and seeded （inoculated） onto a tubular acellular collagen matrix, acquired from allogeneic rabbit bladder submucosa. A urethral mucosal defect was created, and urethral reconstruction was performed with the tubular acellular collagen matrix seeded with epidermal cells. Results： On gross examination at 12 months following the procedure, the mucosa of the urethral grafts appeared lubricous and smooth. Urethrography showed that a wide urethral caliber had been maintained without any sign of strictures. Histological examination showed a transitional cell layer in the graft without evidence of a margin between the graft and the host tissue at 12 months postoperatively. Conclusion： Epidermal cells seeded onto acellular collagen matrices can be successfully used to reconstruct urethras that have defects and are transformed to transitional epithelial cells.

AIM To investigate alterations in the fecal microbiome using 16 S r RNA amplicon sequencing in couples in the same cohabitation *** Fecal samples were collected from eight ulcerative colitis(UC) patients and their healthy partners at Lishui People's Hospital, Zhejiang Province, China. DNA was extracted and the variable regions V3 and V4 of the 16 S r RNA genes were PCR amplified using a two-step protocol. Clear reads were clustered into operational taxonomic units(OTUs) at the 97% sequence similarity level using UCLUST v1.2.22. The Wilcoxon rank-sumtest(R v3.1.2) was used to compare inter-individual differences. Differences with a P value mins were more abundant in the healthy *** Our results suggested that the microbial composition might be affected in healthy partners cohabiting with UC patients, especially in terms of microbiota dysbiosis.

目的探讨大鼠脂肪干细胞(ADSC)的培养及诱导分化为成肌细胞的方法。方法取SD大鼠脂肪组织,酶消化法分离、培养ADSC,免疫荧光和流式细胞仪检测相关表面抗原CD90、CD105和CD34的表达。取培养至第2代处于对数生长期细胞,分别用含5-氮杂胞苷(5-aza)的诱导培养液(诱导组)和基础培养液(对照组)进行培养。诱导时间为7、14、21、28和35 d,倒置相差显微镜观察细胞的生长情况和形态变化,免疫荧光和流式细胞仪检测成肌细胞特异性抗原desmin和myosin的表达。结果成功从大鼠脂肪组织分离、培养出ADSC,相关表面抗原表达检测证实其干细胞特性。诱导组细胞诱导28 d后呈现成肌细胞特有的"漩涡"样生长形态,单个细胞表现出多核化;免疫荧光和流式细胞仪检测显示,desmin和myosin的表达率在诱导28 d时达最高,分别为52.57%和50.04%,而诱导前和对照组细胞均呈阴性表达。结论成功从大鼠脂肪组织分离、培养ADSC,含5-aza的诱导培养液可将其诱导分化为成肌细胞,诱导28 d成肌细胞特异性抗原表达率最高。

背景:脂肪干细胞是目前组织工程研究中的热点种子细胞,但由于其缺乏特异性的表面标记,故目前尚缺乏有效的纯化方法。通过纯化取材的脂肪来提高干细胞纯度是一种简单可行的方法。目的:分析在原代培养脂肪干细胞前,如何对取材的脂肪组织进行纯化。设计、时间及地点:观察对比动物实验,于2007-12/2008-03在上海市第六人民医院上海市实验医学动物中心完成。材料:选用4周龄SD大鼠,用于获得腹股沟处的脂肪组织。方法:解剖SD大鼠腹股沟处取材的脂肪组织,游离出其表面血管及内部的血管分支。剔除内部血管的同时将分布在血管周围的椭圆形结节组织一并切除。主要观察指标:对椭圆性结节行苏木精-伊红染色,显微镜下观察以确定其组织类型。分别对经过纯化处理和未纯化处理的脂肪组织行苏木精-伊红染色,显微镜下观察其组织结构上的差异。结果:①椭圆性小结节经苏木精-伊红染色后在显微镜下观察证实为淋巴组织。②纯化的脂肪组织结构一致,细胞成分单一;未纯化的脂肪组织结构紊乱,细胞种类多样、成分复杂。结论:脂肪干细胞原代培养所用的脂肪组织中含有多种组织成分,在剔除脂肪组织表面的血管、皮肤和肌肉组织的同时应一并剔除组织内的血管及其周围淋巴结。

The Circular Electron Positron Collider(CEPC)is a large scientific project initiated and hosted by China,fostered through extensive collaboration with international *** complex comprises four accelerators:a 30 GeV Linac,a 1.1 GeV Damping Ring,a Booster capable of achieving energies up to 180 GeV,and a Collider operating at varying energy modes(Z,W,H,and tt).The Linac and Damping Ring are situated on the surface,while the subterranean Booster and Collider are housed in a 100 km circumference underground tunnel,strategically accommodating future expansion with provisions for a potential Super Proton Proton Collider(SPPC).The CEPC primarily serves as a Higgs *** its baseline design with synchrotron radiation(SR)power of 30 MW per beam,it can achieve a luminosity of 5×10^(34)cm^(-2)s^(-1)per interaction point(IP),resulting in an integrated luminosity of 13 ab^(-1)for two IPs over a decade,producing 2.6 million Higgs *** the SR power to 50 MW per beam expands the CEPC's capability to generate 4.3 million Higgs bosons,facilitating precise measurements of Higgs coupling at sub-percent levels,exceeding the precision expected from the HL-LHC by an order of *** Technical Design Report(TDR)follows the Preliminary Conceptual Design Report(Pre-CDR,2015)and the Conceptual Design Report(CDR,2018),comprehensively detailing the machine's layout,performance metrics,physical design and analysis,technical systems design,R&D and prototyping efforts,and associated civil engineering ***,it includes a cost estimate and a preliminary construction timeline,establishing a framework for forthcoming engineering design phase and site selection *** is anticipated to begin around 2027-2028,pending government approval,with an estimated duration of 8 *** commencement of experiments and data collection could potentially be initiated in the mid-2030s.

Malnutrition is a common comorbidity among patients with ***,no nutrition-screening tool has been recognized in this population.A quick and easy screening tool for nutrition with high sensitivity and easy-to-use is *** on the previous 25 nutrition-screening tools,the Delphi method was made by the members of the Chinese Society of Nutritional Oncology to choose the most useful item from each *** to these results,we built a nutrition-screening tool named age,intake,weight,and walking(AIWW).Malnutrition was defined based on the scored patient-generated subjective global assessment(PG-SGA).Concurrent validity was evaluated using the Kendall tau coefficient and kappa consistency between the malnutrition risks of AIWW,nutritional risk screening 2002(NRS-2002),and malnutrition screening tool(MST).Clinical benefit was calculated by the decision curve analysis(DCA),integrated discrimination improvement(IDI),and continuous net reclassification improvement(c NRI).A total of 11,360 patients(male,n=6,024(53.0%)were included in the final study cohort,and 6,363 patients had malnutrition based on *** on AIWW,NRS-2002,and MST,7,545,3,469,and1,840 patients were at risk of malnutrition,*** sensitivities of AIWW,NRS-2002,and MST risks were 0.910,0.531,and 0.285,and the specificities were 0.768,0.946,and *** Kendall tau coefficients of AIWW,NRS-2002,and MST risks were 0.588,0.501,and 0.326,*** area under the curve of AIWW,NRS-2002,and MST risks were0.785,0.739,and 0.630,*** IDI,c NRI,and DCA showed that AIWW is non-inferior to NRS-2002(IDI:0.002(-0.009,0.013),c NRI:-0.015(-0.049,0.020)).AIWW scores can also predict the survival of patients with *** missed diagnosis rates of AIWW,NRS-2002,and MST were 0.09%,49.0%,and 73.2%,*** showed a better nutritionscreening effect than NRS-2002 and MST for patients with cancer and could be recommended as an alternative nutritionscreening tool

目的探讨脐动脉平滑肌细胞（HUASMC）与阴茎海绵体脱细胞基质（ACCM）复合构建海绵体平滑肌的可行性。方法以1％的Triton-X100与0．1％NH3H2O混合液对兔阴茎海绵体进行脱细胞处理，制备ACCM。采用贴块法分离、培养、扩增HUASMC。HUASMC以30×10^6／ml密度接种ACCM，细胞ACCM体外复合10d后将复合物植人9只5周龄BALB／C裸鼠背部皮下，术后10、20和40d分别对移植物进行HE染色、免疫组织化学染色和器官浴槽实验，评价其裸鼠体内构建情况。结果ACCM为白色圆柱状，镜下为富含胶原的疏松多孔结构，不含细胞成分。HUASMC与ACCM相容性良好，HUASMC在与ACCcM接触部位充分伸展，并沿ACCM窦隙活跃生长。9只裸鼠均存活，植人部位无感染，无植人物排斥发生。随培育时间延长，裸鼠体内ACCM逐渐降解，植入的HUASMC分化形成结构良好、交错排列的平滑肌组织。器官浴槽实验显示，构建组织对去氧肾上腺素和电刺激均表现出收缩功能，去氧肾上腺素和电刺激所诱导的最大收缩力分别为（3．64±0．18）和（2．50±0．21）g。结论HUASMC作为种子细胞与ACCM复合可构建出具有一定形态和功能的组织工程海绵体平滑肌。
