Comparative study of galectin-3 and B-type natriuretic peptide as biomarkers for the diagnosis of heart failure

Background Heart failure （HF） is a common disease with complex pathophysiological causes. The diagnosis of HF commonly relies on comprehensive analyses of medical history and symptoms, and results from echocardiography and biochemical tests. Galectin-3, a rela-tively new biomarker in HF, was approved by the US Food and Drug Administration in 2010 as a marker in the stratification of risk for HF. We assessed galectin-3 as a biomarker for HF diagnosis in patients with preserved ejection fraction （pEF） and compared its performance with that of B-type natriuretic peptide （BNP）. Methods Thirty-five pEF patients with HF （HFpEF group） and 43 pEF patients without HF （control group） were enrolled. Plasma levels of galectin-3 and BNP in HFpEF and control subjects were determined. Sensitivity, specificity, pre dictive values, and accuracy of galectin-3 and BNP as markers for HF diagnosis were calculated and compared. Results Levels of galec- tin-3 and BNP were 23.09 ±6.97 ng/mL and 270.46 ± 330.41 pg/mL in the HFpEF group, and 16.74 ± 2.75 ng/mL and 59.94 ± 29.93 pg/mL in the control group, respectively. Differences in levels of galectin-3 and BNP between the two groups were significant （P 〈 0.01）. As a bio- marker for HF diagnosis in study subjects, galectin-3 showed sensitivity and specificity of 94.3% and 65.1%, respectively, at a cutoff value of 17.8 ug/mL. BNP showed sensitivity and specificity of 77.1% and 90.7%, respectively, at a cutoff value of 100 pg/mL. Galectin-3 was a significantly more sensitive （P 〈 0.05） but less specific （P 〈 0.01） biomarker compared with BNP. Differences in positive predictive value, negative predictive value, and accuracy between galectin-3 and BNP markers were not significant （P 〉 0.05）. Areas under the receiver operating characteristic curve （95% confidence interval） were 0.891 （0.808-0.974） and 0.896 （0.809-0.984） for galectin-3 and BNP, respec- tively, with no significant difference between the two values （P 〉 0.05）. In vivo Anti-tumor Effect of siRNA Against STAT3 on Transplanted Lewis Lung Cancer in Mice

This study aims at determining the therapeutic effect of knockdown of signal transducers and activators of transcription3（STAT3） gene expression by short interference RNA（siRNA） on transplanted Lewis lung cancer in mice in vivo. pSilencer 2.1-U6 STAT3 siRNA against STAT3（STAT3 siRNA） was synthesized. Lewis lung cancer cells were inoculated subcutaneously into C57BL/6 mice. Seven days after inoculation, the tumor-bearing mice were randomly divided into 3 groups and received intratumoral injection of （1） vehicle（PBS solution）, （2） vector（negative control）, and （3） STAT3 siRNA. Tumor volume and weight were calculated. Tumors and the lungs were excised for 21 days after inoculation. Expressions of STAT3, vascular endothelial growth factor（VEGF）, and matrix metalloproteinase-2（MMP2） were analyzed by RT-PCR, Western blot, and immunohistochemical staining. HE staining and TUNEL assay were used to confirm the apoptosis of tumors. The synthetic STAT3 siRNA effectively suppressed tumor growth, prevented tumor from pulmonary metastasis, and induced tumor apoptosis in vivo compared with vehicle and vectore in controls. It significantly inhibited STAT3 expression to contribute to downregulation of VEGF and MMP2 expression within tumors in vivo. This study demonstrates that STAT3 siRNA can effectively inhibit the expression of STAT3 gene within tumors, leading to suppression of tumor growth, prevention of cancer from pulmonary metastasis, and enhancing apoptosis of the transplanted Lewis lung cancer in mice in vivo.

Expression of Catalytic Domain of Protein Tyrosine Phosphatase 1B and Preparation of Its Polyclonal Antibody

This study focuses on the expression of human protein tyrosine phosphatase 1B （PTP1 B） catalytic domain （△PTP1B） and preparation of polyclonal antibody against △PTP1B. △PTP1B gene was PCR amplified with the cDNA of human PTP1B as the template, and cloned into the pT7 expression vector. The recombinant pT7-△PTP1B was expressed in E. eoli Rosetta（ DE3 ） host cells and purified. The antiserum was prepared by immunizing rabhit with purified recombinant △PTP1B. The polyclonal antibody against △PTP1B was purified by PVDF immobilized antigen affinity chromatography. △PTP1B was correctly cloned, expressed, and purified as confirmed by PCR, DNA sequence analysis, SDS-PAGE western blotting, and ILPLC. The titer and sensitivity of the antibody were 1：2500（ volume ratio） and 0. 1 ng, respectively. This study provides an important basis for further studying the biological function of PTP1B and its relationship with human diseases.

Expression and Characterization of Catalytic Domain of T Cell Protein Tyrosine Phosphatase(ΔTC-PTP)——Immunohistochemical Study of ΔTC-PTP Expression in Non-small Cell Lung Carcinomas

This study objective was to express and characterize the catalytic domain of the human T cell protein tyrosine phosphatase（△TC-PTP） and to study immunohistochemically the expression of △TC-PTP in human non-small cell lung cancers. △TC-PTP gene was PCR amplified with the cDNA of human TC-PTP as template, and cloned into the pT7 expression vector. The recombinant pT7-△TC-PTP was expressed in E. coli Rosetta （ DE3 ） host cells and puri- fied. The enzymatic characteristics of △TC-PTP including enzyme activity and kinetics assay were measured. The antiserum was prepared by immunizing rabbit with the purified recombinant △TC-PTP. Rabbit polyclonal antibody against △TC-PTP was purified by PVDF immobilized antigen affinity chromatography. Immunohistochemical staining of lung cancer tissues was performed with antibody against △TC-PTP protein. △TC-PTP gene was correctly cloned, expressed, and purified. The recombinant △TC-PTP had a highly catalytic activity of PTPase. Squamous cell lung carcinoma showed a significantly higher expression rate of △TC-PTP （76. 92%, 10/13 ） than adenocarcinoma （57.14%, 4/7） and normal lung tissue（20%, 1/5 ）. This study represents the first demonstration that △TC-PTP is highly expressed in human squamous cell lung carcinomas. In addition, this study provides an important basis for further studying the biological function of TC-PTP and its relationship with lung carcinomas and other diseases.

In the past, living donor nephrectomy required an open flank incision that results in postoperative morbidity and a prolonged hospital stay. Since its introduction in 1995, laparoscopic living donor nephrectomy has been shown to decrease postoperative pain and hospital stay, reduce blood loss, and improve cosmesis while hastening recovery of normal activities of donors. With decreased morbidity and favorable graft function, this procedure as a novel approach has been used to address the increasing disparity between organ need and availability. Better graft function and survival are noted in living-donor kidney transplantation than in cadavefic kidney transplantation.

Cells are the fundamental units of biological systems and exhibit unique development trajectories and molecular features. Our exploration of how the genomes orchestrate the formation and maintenance of each cell, and control the cellular phenotypes of various organismsis, is both captivating and intricate. Since the inception of the first single-cell RNA technology, technologies related to single-cell sequencing have experienced rapid advancements in recent years. These technologies have expanded horizontally to include single-cell genome, epigenome,proteome, and metabolome, while vertically, they have progressed to integrate multiple omics data and incorporate additional information such as spatial sc RNA-seq and CRISPR screening. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Low-temperature chlorination roasting technology for the simultaneous recovery of valuable metals from spent LiCoO_(2)cathode material

With the continuous increase in the disposal volume of spent lithium-ion batteries(LIBs),properly recycling spent LIBs has become essential for the advancement of the circular *** study presents a systematic analysis of the chlorination roasting kinetics and proposes a new two-step chlorination roasting process that integrates thermodynamics for the recycling of LIB cathode *** activation energy for the chloride reaction was 88.41 kJ/mol according to thermogravimetric analysis–derivative thermogravimetry data obtained by using model-free,model-fitting,and Z(α)function(αis conversion rate).Results indicated that the reaction was dominated by the first-order(F1)model when the conversion rate was less than or equal to 0.5 and shifted to the second-order(F2)model when the conversion rate exceeded *** conditions were determined by thoroughly investigating the effects of roasting temperature,roasting time,and the mass ratio of NH_(4)Cl to LiCoO_(2).Under the optimal conditions,namely 400℃,20 min,and NH_(4)Cl/LiCoO_(2)mass ratio of 3:1,the leaching efficiency of Li and Co reached 99.43% and 99.05%,*** of the roasted products revealed that valuable metals in LiCoO_(2)transformed into CoCl_(2) and ***,the reaction mechanism was elucidated,providing insights for the establishment of a novel low-temperature chlorination roasting technology based on a crystal structure *** technology can guide the development of LIB recycling processes with low energy consumption,low secondary pollution,high recovery efficiency,and high added value.

目的:对比两孔式胸腔镜肺叶切除术(TPTL)与传统三孔式胸腔镜肺叶切除术(TTTL)的临床应用效果,阐述TPTL在临床治疗中的意义及价值。方法:回顾性分析行胸腔镜下肺叶切除的124例患者的临床资料。124例患者按照所行术式的不同分为TPTL组60例(采用TPTL、淋巴结廓清术)和TTTL组64例(采用TTTL、淋巴结廓清术)。比较2组患者手术时间、术中出血量、术后盐酸哌替啶用量、术后拔除胸引管时间、术后住院时间和术后6个月患侧上肢活动后不适者的例数。结果:TPTL组患者切口长度[(4.0±0.2)cm]小于TTTL组患者切口长度[(6.2±0.3)cm](P0.05)。结论:与TTTL比较,TPTL可以减小切口长度、减轻患者术后疼痛、提高患者术后生活质量。

Comparison of the extraperitoneal and transperitoneal laparoscopic radical prostatectomy

The laparoscopic radical prostatectomy （LRP） is a developing technique for treatment of localizedprostate cancer, while the extraperitoneal approach has been highlighted recently.1 From May 2003 to April 2006, we performed laparoscopic radical prostatectomy by transperitoneal or extraperitoneal approaches in 31 patients with localized prostate cancer. Some parameters of these patients are compared in this article.

Boosting hydrocarbon conversion via Cu-doping induced oxygen vacancies on CeO2 in CO2 electroreduction
