Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(P<0.05). Furthermore, our research revealed that monotropein induced cell apoptosis by inhibiting Bcl-2 and increasing Bax, induced G_1–S cycle arrest in colorectal cancer by decreasing the expressions of CyclinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(P<0.05), and might play an anticancer role through Akt signaling pathway. The low-wavenumber components in the gradient of full waveform inversion(FWI)play a vital role in the stability of the ***,when FWI is implemented in some high frequencies and current models are not far away from the real velocity model,an excessive number of low-wavenumber components in the gradient will also reduce the convergence rate and inversion *** solve this problem,this paper firstly derives a formula of scattering angle weighted gradient in FWI,then proposes a hybrid *** hybrid gradient combines the conventional gradient of FWI with the scattering angle weighted gradient in each inversion frequency band based on an empirical formula derived *** weighted hybrid mode,we can retain some low-wavenumber components in the initial lowfrequency inversion to ensure the stability of the inversion,and use more high-wavenumber components in the high-frequency inversion to improve the convergence *** results of synthetic data experiment demonstrate that compared to the conventional FWI,the FWI based on the proposed hybrid gradient can effectively reduce the low-wavenumber components in the gradient under the premise of ensuring inversion *** also greatly enhances the convergence rate and inversion accuracy,especially in the deep part of the *** the field marine seismic data experiment also illustrates that the FWI based on hybrid gradient(HGFWI)has good stability and adaptability.

Enhancing photoresponsiveness of metal-organic polyhedra by modifying microenvironment

Photoresponsiveness of materials is critical to their tunability and efficiency in terminal *** metal-organic polyhedra(PMOPs)feature intrinsic pores and remote controllability,but aggregation of PMOPs in solid state hampers their photoresponsiveness ***,we report the construction of a new PMOP(Cu_(24)(C_(16)H_(12)N_(2)O_(4))_(12)(C_(18)H_(22)O_(5))12,denoted as MOP-PR-LA),where long alkyl(LA)chains act as the intermolecular poles,propping against adjacent PMOP molecules to create individual microenvironment benefiting the isomerization of photoresponsive(PR)*** ultraviolet(UV)-and visible-light irradiation,MOP-PR-LA is much easier to isomerize than the counterpart MOP-PR without *** propylene adsorption,MOP-PR has a low change of adsorption capacity(9.9%),while that of MOP-PR-LA reaches 58.6%.Density functional theory calculations revealed that PR in the cis state has a negative effect on adsorption,while the trans state of PR favors *** work might open an avenue for the construction of photoresponsive materials with high responsiveness and controllability.

Combined use of fly ash and silica to prevent the long-term strength retrogression of oil well cement set and cured at HPHT conditions

The long-term strength retrogression of silica-enriched oil well cement poses a significant threat to wellbore integrity in deep and ultra-deep wells, which is a major obstacle for deep petroleum and geothermal energy development. Previous attempts to address this problem has been unsatisfactory because they can only reduce the strength decline rate. This study presents a new solution to this problem by incorporating fly ash to the traditional silica-cement systems. The influences of fly ash and silica on the strength retrogression behavior of oil well cement systems directly set and cured under the condition of 200°C and 50 MPa are investigated. Test results indicate that the slurries containing only silica or fly ash experience severe strength retrogression from 2 to 30 d curing, while the slurries containing both fly ash and silica experience strength enhancement from 2 to 90 d. The strength test results are corroborated by further evidences from permeability tests as well as microstructure analysis of set cement. Composition of set cement evaluated by quantitative X-ray diffraction analyses with partial or no known crystal structure(PONKCS) method and thermogravimetry analyses revealed that the conversion of amorphous C-(A)-S-H to crystalline phases is the primary cause of long-term strength *** addition of fly ash can reduce the initial amount of C-(A)-S-H in the set cement, and its combined use with silica can prevent the crystallization of C-(A)-S-H, which is believed to be the working mechanism of this new admixture in improving long-term strength stability of oil well cement systems.

Microphase Separation of Semiflexible Ring Diblock Copolymers

Aiming at the difficult problem of solving the conformation statistics of complex polymers, this study presents a novel and concise conformation statistics theoretical approach based on Monte Carlo and Neural Network method. This method offers a new research idea for investigating the conformation statistics of complex polymers, characterized by its simplicity and practicality. It can be applied to more complex topological structure, more higher degree of freedom polymer systems with higher dimensions, theory research on dynamic self-consistent field theory and polymer field theory, as well as the analysis of scattering experimental data. The conformation statistics of complex polymers determine the structure and response properties of the system. Using the new method proposed in this study, taking the semiflexible ring diblock copolymer as an example, Monte Carlo simulation is used to sample this ring conformation to construct the dataset of polymer. The structure factor describing conformation statistics are expressed as continuous functions of structure parameters by neural network supervised learning. This is the innovation of this work. As an application, the structure factors represented by neural networks were introduced into the random phase approximation theory to study the microphase separation of semiflexible ring diblock copolymers. The influence of the ring's topological properties on the phase transition behavior was pointed out.

Semiannual oscillation,annual oscillation,quasibiennial oscillation,and solar cycle variation of the OH airglow emission in the mesopause region

The vertically integrated emission rate,centroid altitude,peak emission rate,and peak height of the hydroxyl(OH)airglow were calculated from Thermosphere Ionosphere Mesosphere Energetics and Dynamics(TIMED)/Sounding of the Atmosphere using Broadband Emission Radiometry(SABER)observations to study the seasonal and interannual variations in the intensity and location of the OH *** emission rate is inversely proportional to the height of the emission,with the semiannual oscillation dominating at low latitudes and the annual oscillation dominating at higher *** OH emission is modulated by the quasibiennial oscillation at the equator,and the quasibiennial oscillation signal is weak at other *** represented the vertical transport of atomic oxygen by using atomic oxygen concentrations obtained from a global atmospheric model,the Specified Dynamics Whole Atmosphere Community Climate Model with thermosphere and ionosphere eXtension *** with the amplitudes of the migrating diurnal tide(DW1)calculated from temperature data observed by TIMED/SABER,we found that both the vertical transport of atomic oxygen and DW1 amplitudes in the equatorial region exhibit semiannual oscillation and quasibiennial oscillation,which have a strong correlation with the variations in the amplitude and phase of semiannual oscillation and quasibiennial oscillation in OH *** is likely that the DW1 affects the vertical transport of atomic oxygen that is involved in the reaction to produce O3,thus affecting the OH *** analyzed the relationship between OH emission and solar activity by using the solar radio flux at 10.7 cm as a proxy for solar *** results showed that the OH emission is well correlated with solar activity,and the modulation of OH emission by solar activity has a significant latitudinal *** small correlation between emission height and solar activity indicates that solar activity modulates OH emission mainly thr

Distribution and geochemical significance of alkylbenzenes for crude oil with different depositional environments and thermal maturities

A total of 45 alkylbenzenes were detected and identified in crude oils with different depositional environments and thermal maturities from the Tarim Basin,Beibuwan Basin,and songliao Basin using comprehensive two-dimensional gas chromatography time-of-flight mass spectrometry(GC×GCTOFMS).By analyzing the distribution characteristics of C0-C5alkylbenzenes,it is found that the content of some alkylbenzenes varies greatly in crude *** on the distribution characteristics of 1,2,4,5-tetramethylbenzene(Te MB)and 1,2,3,4-Te MB,the ratio of 1,2,4,5-Te MB to 1,2,3,4-Te MB is proposed to indicate the organic matter origin and depositional environment of ancient *** samples originated mainly from lower hydrobiont,algae,bacteria and source rocks deposited under reducing/anoxic conditions have low 1,2,4,5-/1,2,3,4-Te MB values(less than 0.6),while oil samples originated mainly from terrestrial higher plants and source rocks deposited under oxic/sub-oxic conditions have higher 1,2,4,5-/1,2,3,4-Te MB values(greater than 1.0).The significant difference of 1,2,4,5-/1,2,3,4-Te MB values is controlled by 1,2,4,5-Te MB content.1,2,4,5-Te MB content in oils derived from source rocks deposited in oxidized sedimentary environment(greater than 1.0 mg/g whole oil)is higher than that in oils from source rocks deposited in reduced sedimentary environment(less than 1.0 mg/g whole oil).1,2,4,5-/1,2,3,4-Te MB ratio might not or slightly be affected by evaporative fractionation,biodegradation and thermal maturity.1,2,4,5-/1,2,3,4-Te MB ratio and 1,2,4,5-Te MB content can be used as supplementary parameter for the identification of sedimentary environment and organic matter *** should be noted that compared to the identification of organic matter sources,the 1,2,4,5-/1,2,3,4-Te MB parameter is more effective in identifying sedimentary environments.

We previously developed a powered hip prosthetic mechanism with kinematic functions of hip flexion-extension and abduction-adduction,and its theoretical and simulation-based kinematics were *** internal-external hip rotation has a positive effect on the movements of human lower limbs according to medical research,we developed a novel hip prosthetic mechanism based on a previous hip prosthesis that possesses motion characteristics similar to those of a human bionic hip,and the motion characteristics of multiple Degrees-of-Freedom(DoFs)were analyzed after kinematic ***,a walking model of the human‒machine model was established,and the walking stability of an amputee,which reflects the rehabilitation effect,was explored while the hip prosthetic mechanism considered the internal-external rotation of the ***,a prototype and its verification platform were built,and kinematic validation of the hip prosthetic mechanism was carried *** results showed that the designed Parallel Mechanism(PM)possesses human-like motion characteristics similar to those of a human bionic hip and can be used as a hip ***,the existing motion characteristic of internal-external hip rotation can enhance the walking stability of an amputee via this hip prosthetic mechanism.
