目的探索脑型疟发病过程中,星形胶质细胞被脑血管周炎性微环境诱导活化及其机制,及其对神经元损伤的影响。方法4~5周C57BL/6雄性小鼠经腹腔接种5×10^(6)个感染伯氏疟原虫ANKA株的红细胞,7~10 d后死亡,作为实验型脑型疟模型。新生3~5 d的乳鼠,安乐死后分离脑皮层原代星形胶质细胞;2. h内的乳鼠,安乐死后分离脑皮层原代神经元。以2. ng/mlγ干扰素(IFN-γ)、1 ng/ml肿瘤坏死因子α(TNF-α)和感染疟原虫的小鼠红细胞(pRBC)诱导星形胶质细胞活化,作为活化组,同时设置未处理静息态细胞作为对照组。2. h后用细胞裂解提取液提取总RNA测序,聚类分析生成热图,选取代表性的数个基因绘制小提琴图。EliSA测定细胞培养上清中CXCL10表达水平。分离脑型疟小鼠脾脏CD8^(+)T细胞,与活化的星形胶质细胞共孵育,荧光显微镜下观察免疫突触及与CXCL10抗体共孵育后的CXCL10水平,流式细胞仪检测CD80等分子表达水平。原代神经元加入两组星形胶质细胞培养上清,在MAP2.体中孵育,设置空白培养基作为空白组,乳酸脱氢酶(LDH)检测试剂盒检测神经元损伤程度,CCK-8试剂盒检测神经元活力。Western blotting检测STAT1、STAT3及其磷酸化分子水平,活化组加入STAT1通路抑制剂氟达拉滨,免疫荧光染色观察神经元形态。采用PRISM Graph Pad 8.0软件进行统计学分析,两两比较采用t检验。结果活化组星形胶质细胞形态由扁平星状变为长梭形,胶质纤维酸性蛋白(GFAP)相对含量为(1.36±0.03),高于对照组的(1.00±0.00)(t=13.33,P1);活化组细胞培养上清中的CXCL10含量为(7.07±0.81)ng/ml,高于对照组的(2.57±0.2.)ng/ml(t=9.05,P1)。转录组分析显示,活化组抗原加工、提呈与T细胞趋化因子转录水平上调(均P1),CD80、CD86、MHCⅠ表达水平上调。活化组和CD8^(+)T细胞共孵育形成“免疫突触”,CD8^(+)T细胞CD69表达量提高。LDH结果显示,活化组上清刺激后神经元相对死亡率为(50.2.2.4)%,高于空白组(0%)和对照组(0%)(t=2..62.2..62.均P1);CCK-8检测结果显示,活化组上清刺激后神经元相对活力为0.52.0.03,低于空白组(1.00±0.00)和对照组(1.42.0.06)(t=18.92.16.65,均P1);Western blotting结果显示,神经元经活化星形胶质细胞上清刺激后β-淀粉样前体蛋白(β-APP)相对表达量为0.44±0.02.低于空白组(1.00±0.00)和对照组(0.55±0.02.(t=37.2.、4.93,均P1);促凋亡蛋白Bax与抑凋亡蛋白Bcl-2.比例为1.01±0.07,与空白组(1.00±0.00)、对照组(1.00±0.06)比较差异无统计学意义(t=0.31、0.13,均P>0.05)。Western blotting结果显示,活化组胞浆中STAT1、p-STAT1蛋白相对表达量分别为3.40±1.08、4.00±0.82.均高于对照组(1)(t=3.13、5.13,均P1.00±0.03、1.01±0.05,与对照组(1)差异无统计学意义(t=0.2.、0.52.均P>0.05)。在活化组胞核中p-STAT1蛋白相对表达量为1.78±0.2.,高于对照组(1)(t=5.081,P1)、p-STAT3蛋白相对表达量为1.02.0.02.与对照组(1)差异无统计学意义(t=1.38,均P>0.05)。对MAP2.疫荧光染色结果显示,活化组上清可造成明显的神经元损伤,经STAT1抑制剂处理后损伤得到缓解。结论脑型疟发病过程中血管周炎性微环境可通过STAT1分子诱导神经毒性星形胶质细胞产生,可导致神经元死亡。活化星形胶质细胞与活化的CD8^(+)T细胞相互作用,进一步加重中枢神经系统损伤。

Aims:Surveys and research on the applications of the hepatic venous pressure gradient(HVPG)are important for understanding the current status and future development of this technology in *** article aimed to investigate the status of hepatic venous pressure gradient measurement in china in ***:We investigated the overall status of HVPG technology in china-including hospital distribution,hospital level,annual number of cases,catheters used,average cost,indications,and current challenges by using online *** counting the number and percentages of cases of these results,we hope to clarify the current status of HVPG measurements in ***:According to the survey,85 hospitals in china used HVPG technology in 2.2. distributed across 2. provinces.A total of 4989 HVPG measurements were performed in all of the surveyed hospitals in 2.2.,of which 2.13 cases(56.4%)were measured *** average cost of HVPG measurement was 5646.8±2.2..9 *** the clinical teams who performed the measurements(sometimes multiple per hospital),94.3%(82.87)used the balloon method,and the majority of the teams(72.4%,63/87)used embolectomy ***:This survey clarified the clinical application status of HVPG in china and confirmed that some medical institutions in china have established a foundation for this *** is still necessary to continue promoting and popularizing this technology in the future.

Osteosarcoma is the most common primary bone sarcoma that mostly occurs in young adults. The causes of osteosarcoma are heterogeneous and still not fully understood. Identification of novel, important oncogenic factors in osteosarcoma and development of better, effective therapeutic approaches are in urgent need for better treatment of osteosarcoma patients. In this study, we uncovered that the oncogene MYC is significantly upregulated in metastastic osteosarcoma samples. In addition, high MYC expression is associated with poor survival of osteosarcoma patients. Analysis of MYC targets in osteosarcoma revealed that most of the osteosarcoma super enhancer genes are bound by MYC. Treatment of osteosarcoma cells with super enhancer inhibitors THZ1 and JQ1 effectively suppresses the proliferation, migration, and invasion of osteosarcoma cells. Mechanistically,THZ1 treatment suppresses a large group of super enhancer containing MYC target genes including CDK6 and TGFB2. These findings revealed that the MYC-driven super enhancer signaling is crucial for the osteosarcoma tumorigenesis and targeting the MYC/super enhancer axis represents as a promising therapeutic strategy for treatment of osteosarcoma patients.

Grain formation is fundamental for crop yield but is vulnerable to abiotic and biotic *** grain production is threatened by the false smut fungus Ustilaginoidea virens,which specifically infects rice floral organs,disrupting fertilization and seed ***,little is known about the molecular mechanisms of the ***-rice interaction and the genetic basis of floral ***,we report that *** secretes a cytoplasmic effector,UvCBP1,to facilitate infection of rice ***,UvCBP1 interacts with the rice scaffold protein OsRACK1A and competes its interaction with the reduced nicotinamide adenine dinucleotide phosphate oxidase OsRBOHB,leading to inhibition of reactive oxygen species(ROS)*** the analysis of natural variation revealed no OsRACK1A variants that could avoid being targeted by UvCBP1,expression levels of OsRACK1A are correlated with field resistance against *** in rice *** of OsRACK1A restores the OsRACK1A-OsRBOHB association and promotes OsRBOHB phosphorylation to enhance ROS production,conferring rice floral resistance to *** without yield *** together,our findings reveal a new pathogenic mechanism mediated by an essential effector from a flower-specific pathogen and provide a valuable genetic resource for balancing disease resistance and crop yield.

Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2. of emergency department *** syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in china, 2.18 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). The present guideline includes the underlying diseases of syncope in children and adolescents, the diagnostic procedures, methodology and clinical significance of standing test and headup tilt test, the clinical diagnosis vasovagal syncope, postural orthostatic tachycardia syndrome, orthostatic hypotension and orthostatic hypertension, and the treatment of syncope as well as follow-up.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2.11 and ended in June 2.12. and the data have been released to the public as the LAMOST Pilot Data Release in August 2.12. The regular survey started in September 2.12. and completed its first year of operation in June 2.13. The LAMOST DR1 includes a total of 12.2.plates containing 2.955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2.are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2.2.4 696 spectra, of which 1 944 32. are stellar spectra, 12.082.are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（12. 52. entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.
