The TNFα/TNFR2 axis mediates natural killer cell proliferation by promoting aerobic glycolysis

Natural killer(NK)cells are predominant innate lymphocytes that initiate the early immune response during *** cells undergo a metabolic switch to fuel augmented proliferation and activation following *** necrosis factor-alpha(TNFα)is a well-known inflammatory cytokine that enhances NK cell function;however,the mechanism underlying NK cell proliferation in response to TNFαis not well ***,we demonstrated that upon infection/inflammation,NK cells upregulate the expression of TNF receptor 2(TNFR2),which is associated with increased proliferation,metabolic activity,and effector ***,IL-18 can induce TNFR2 expression in NK cells,augmenting their sensitivity toward TNFα.Mechanistically,TNFα-TNFR2 signaling upregulates the expression of CD25(IL-2Rα)and nutrient transporters in NK cells,leading to a metabolic switch toward aerobic *** analysis revealed significantly reduced expression levels of genes involved in cellular metabolism and proliferation in NK cells from TNFR2 KO ***,our data affirmed that genetic ablation of TNFR2 curtails CD25 upregulation and TNFα-induced glycolysis,leading to impaired NK cell proliferation and antiviral function during MCMV infection in ***,our results delineate the crucial role of the TNFα-TNFR2 axis in NK cell proliferation,glycolysis,and effector function.

N6-methyladenosine methylation(m6A)is a common type of epigenetic alteration that prominently affects the prognosis of tumor ***,it is unknown how the m6A regulator affects the tumor microenvironment(TME)cell infiltration in adrenocortical carcinoma(ACC)and how it affects the prognosis of ACC patients *** m6A alteration patterns of 112 ACC patients were evaluated,furthermore,the association with immune infiltration cell features was *** unsupervised clustering method was applied to typify the m6A alteration patterns of ACC *** principal component analysis(PCA)technique was taken to create the m6A score to assess the alteration pattern in specific *** found two independent patterns of m6A alteration in ACC *** TME cell infiltration features were significantly in accordance with phenotypes of tumor immune-inflamed and immune desert in both *** m6Ascore also served as an independent predictive factor in ACC *** somatic copy number variation(CNV)and patients prognosis can be predicted by m6A alteration ***,the ACC patients with high m6A scores had better overall survival(OS)and higher efficiency in immune checkpoint blockade *** work demonstrated the significance of m6A alteration to the ACC patients *** individual m6A alteration patterns analysis might contribute to ACC patients prognosis prediction and immunotherapy choice.

Comparative Analysis of Dietary Fat Intake among Female University Students in Riyadh: A Cross-Sectional Assessment of Health Sciences versus Non-Health Sciences Disciplines

Background: In the global burden of non-communicable diseases, the top slot among them all is occupied by various cardiovascular diseases (CVD), which also claim primacy in mortality rates. Generally speaking, the pathophysiological pathway frequently involves either (or a combination of) elevated serum cholesterol levels, predominantly attributed to dietary patterns characterized by extremely high levels of saturated and trans-fat consumption. Day-to-day nutritional epidemiology among university students suggests that they represent a crucial demographic for the study of preventive interventions. Objective: This in-depth study and investigation were specifically aimed to quantitatively assess and compare prevailing patterns of dietary fat consumption between female students enrolled in health sciences and non-health sciences programs at major universities in Riyadh, Saudi Arabia. Methodology: This cross-sectional study was comprised of 434 Saudi female university students (age range: 18 - 25 years), utilizing a validated Block Dietary Fat Screener accompanied by anthropometric measurements and comprehensive demographic data collection. The sampling methodology consisted of a snowball technique that was spread across three major academic institutions in Riyadh. Results: Statistical analysis revealed a significant association between academic specialization and patterns of dietary fat consumption (p = 0.012). Paradoxically, students of health science exhibited a higher prevalence of very high fat intake (41.9%) compared with their non-health sciences counterparts (27.6%). Surprisingly, anthropometric stratification revealed that it was the underweight students who demonstrated very high levels of fat consumption. A notable socioeconomic gradient was observed, with students from households earning 15,000 - 20,000 SR monthly showing the highest prevalence of very high-fat consumption (47.1%), although this association did not achieve statistical significance (p = 0.104

Smart Home IoT Privacy and Security Preservation via Machine Learning Techniques

The development and use of Internet of Things(IoT)devices have grown significantly in recent *** IoT device characteristics are mainly to blame for the wide range of applications that may now be achieved with IoT *** have begun to embrace the IoT *** true and suitable devices,security faults that might be used for bad reasons,and administration of such devices are only a few of the issues that IoT,a new concept in technological progress,*** some ways,IoT device traffic differs from regular device *** with particular features can be classified into categories,irrespective of their function or ***-changing and complex environments,like a smart home,demand this classification scheme.A total of 41 IoT devices were employed in this *** build a multiclass classification model,IoT devices contributed 13 network traffic *** further preprocess the raw data received,preprocessing techniques like Normalization and Dataset Scaling were *** engineering techniques can extract features from the text data.A total of 117,423 feature vectors are contained in the dataset after stratification,which are used to further improve the classification *** this study,a variety of performance indicators were employed to show the performance of the logiboosted ***-XGB scored 80.2%accuracy following application of the logit-boosted algorithms to the dataset for classification,whereas Logi-GBC achieved 77.8%***,Logi-ABC attained 80.7%***-CBC,on the other hand,received the highest Accuracy score of 85.6%.The accuracy of Logi-LGBM and Logi-HGBC was the same at 81.37%*** suggested Logi-CBC showed the highest accuracy on the dataset when compared to existing Logit-Boosted Algorithms used in earlier studies.

Recently,the number of Internet of Things(IoT)devices connected to the Internet has increased dramatically as well as the data produced by these *** would require offloading IoT tasks to release heavy computation and storage to the resource-rich nodes such as Edge Computing and Cloud ***,different service architecture and offloading strategies have a different impact on the service time performance of IoT ***,this paper presents an Edge-Cloud system architecture that supports scheduling offloading tasks of IoT applications in order to minimize the enormous amount of transmitting data in the ***,it introduces the offloading latency models to investigate the delay of different offloading scenarios/schemes and explores the effect of computational and communication demand on each one.A series of experiments conducted on an EdgeCloudSim show that different offloading decisions within the Edge-Cloud system can lead to various service times due to the computational resources and communications ***,this paper presents a comprehensive review of the current state-of-the-art research on task offloading issues in the Edge-Cloud environment.

Text visualization is concerned with the representation of text in a graphicalform to facilitate comprehension of large textual data. Its aim is to improve the ability tounderstand and utilize the wealth of text-based information available. An essential task inany scientific research is the study and review of previous works in the specified domain,a process that is referred to as the literature survey process. This process involves theidentification of prior work and evaluating its relevance to the research question. With theenormous number of published studies available online in digital form, this becomes acumbersome task for the researcher. This paper presents the design and implementationof a tool that aims to facilitate this process by identifying relevant work and suggestingclusters of articles by conceptual modeling, thus providing different options that enablethe researcher to visualize a large number of articles in a graphical easy-to-analyze *** tool helps the researcher in analyzing and synthesizing the literature and building aconceptual understanding of the designated research area. The evaluation of the toolshows that researchers have found it useful and that it supported the process of relevantwork analysis given a specific research question, and 70% of the evaluators of the toolfound it very useful.

Exploring and Modelling IoT Offloading Policies in Edge Cloud Environments

The Internet of Things(IoT)has recently become a popular technology that can play increasingly important roles in every aspect of our daily *** collaboration between IoT devices and edge cloud servers,edge server nodes provide the computation and storage capabilities for IoT devices through the task offloading process for accelerating tasks with large resource ***,the quantitative impact of different offloading architectures and policies on IoT applications’performance remains far from clear,especially with a dynamic and unpredictable range of connected physical and virtual *** this end,this work models the performance impact by exploiting a potential latency that exhibits within the environment of edge ***,it investigates and compares the effects of loosely-coupled(LC)and orchestrator-enabled(OE)*** LC scheme can smoothly address task redistribution with less time consumption for the offloading sceneries with small scale and small task ***,the OE scheme not only outperforms the LC scheme in the large-scale tasks requests and offloading occurs but also reduces the overall time by 28.19%.Finally,to achieve optimized solutions for optimal offloading placement with different constraints,orchestration is important.

A Novel Energy and Communication Aware Scheduling on Green Cloud Computing

The rapid growth of service-oriented and cloud computing has created large-scale data centres *** data centres’operating costs mostly come from back-end cloud infrastructure and energy *** cloud computing,extensive communication resources are ***,cloud applications require more bandwidth to transfer large amounts of data to satisfy end-user *** is also essential that no communication source can cause congestion or bag loss owing to unnecessary switching *** paper proposes a novel Energy and Communication(EC)aware scheduling(EC-scheduler)algorithm for green cloud computing,which optimizes data centre energy consumption and traffic *** primary goal of the proposed EC-scheduler is to assign user applications to cloud data centre resources with minimal utilization of data *** first introduce a Multi-Objective Leader Salp Swarm(MLSS)algorithm for task sorting,which ensures traffic load balancing,and then an Emotional Artificial Neural Network(EANN)for efficient resource ***-scheduler schedules cloud user requirements to the cloud server by optimizing both energy and communication delay,which supports the lower emission of carbon dioxide by the cloud server system,enabling a green,unalloyed *** tested the proposed plan and existing cloud scheduling methods using the GreenCloud simulator to analyze the efficiency of optimizing data centre energy and other scheduler *** EC-scheduler parameters Power Usage Effectiveness(PUE),Data Centre Energy Productivity(DCEP),Throughput,Average Execution Time(AET),Energy Consumption,and Makespan showed up to 26.738%,37.59%,50%,4.34%,34.2%,and 33.54%higher efficiency,respectively,than existing state of the art schedulers concerning number of user applications and number of user requests.
