Low doses of ethanolic extract of Boldo(Peumus boldus) can ameliorate toxicity generated by cisplatin in normal liver cells of mice in vivo and in WRL-68 cells in vitro, but not in cancer cells in vivo or in vitro

OBJECTIVE： Use of cisplatin, a conventional anticancer drug, is restricted because it generates strong hepatotoxicity by accumulating in liver. Therefore its anticancer potential can only be fully exploited if its own toxicity is considerably reduced. Towards this goal, ethanolic extract of the plant, Boldo （Peumus boldus）, known for its antihepatotoxic effects, was used simultaneously with cisplatin, to test its ability to reduce cisplatin＇s cytotoxicity without affecting its anticancer potential. METHODs： The cytotoxicity of Boldo extract （BE） and cisplatin, administered alone and in combination, was determined in three cancer cell lines （A549, HeLa, and HepG2） and in normal liver cells （WRL-68）. Drug-DNA interaction, DNA damage, cell cycle, apoptosis, reactive oxygen species （ROs） and mitochondrial membrane potential （MMP, Aψ） were also studied. Hepatotoxicity and antioxidant activity levels were determined by alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase and glutathione assays in mice. The cytotoxicity of related proteins was tested by Western blotting. REsULTs： Co-administration of BE and cisplatin increased viability of normal cells, but had no effect on the viability of cancer cells. Boldo protected liver from damage and normalized different antioxidant enzyme levels in vivo and also reduced ROs and re-polarized MMP in vitro. Bax and cytochrome c translocation was reduced with caspase 3 down-regulation. Further, a drug- DNA interaction study revealed that BE reduced cisplatin＇s DNA-binding capacity, resulting in a reduction in DNA damage. CONCLUsION： Results indicated that a low dose of BE could be used beneficially in combination with cisplatin to reduce its toxicity without hampering cisplatin＇s anticancer effect. These findings siqnify a potential future use of BE in cancer therapy.

通过ButPCl2与两倍量原位形成的邻位胺基取代苯基锂的反应合成了二胺基-膦配体H2(MesN2PBu-t)和H2(dcpN2PBu-t).利用H2(MesN2PBu-t)和H2(dcpN2PBu-t)与[Fe(N(siMe3)2)2]2的胺消除反应合成得到二胺基-膦配位的高自旋亚铁配合物[(κ3-N,N,P-MesN2PBu-t)Fe(OEt2)](1)和[(κ3-N,N,P-dcpN2PBu-t)Fe(OEt2)](2).配合物1与重氮化合物(p-tolyl)2CN2和PhC(N2)CO2Et反应分别生成重氮配位的铁配合物[(κ3-N,N,P-MesN2PBu-t)Fe(η2-N,N-(p-tolyl)2CN2)](3)和[(κ3-N,N,P-MesN2PBu-t)Fe(κ2-N,O-PhC(N2)CO2Et)](4).配合物2可以催化重氮化合物(p-tolyl)2CN2转化为酮连氮化合物(p-tolyl)2-C=NN=C(tolyl-p)2.配合物2与重氮化合物DmpC(N2)H反应生成二胺基-膦叶立德配位的高自旋亚铁配合物[(κ3-N,N,C-dcpN2PBu-tCH,Dmp)Fe](5).二胺基-膦配体H2(MesN2PBu-t)和H2(dcpN2PBu-t)以及配合物1~5通过了各种谱学数据的表征.其中配合物2~5的结构还通过单晶X射线衍射得到确认.理论计算显示配合物3可看作为低自旋型四价铁末端亚胺基配合物.配合物4的电子结构可描述为中间自旋的三价铁中心与重氮自由基阴离子反铁磁性耦合形成基态自旋为s=1的配合物.

I describe a method for the observation of Positronium (Ps) involvement in the solar radiation spectrum. In this method, Rydberg-Ritz’s principle and Planck’s radiation formula are used to acquire information of the atomic transitions of Ps alike Hydrogen and Helium. In order to perform this experiment, an advanced solar spectrum monitor is constructed by utilizing light emitting diodes (LED) of various colors. A detailed study on this method provides qualitative agreement with experimental data, giving insight to the physical process involved in the solar radiation spectrum and confirming the existence of solar Ps.

Does the Fifth sate of Matter Originate the Early Universe？

In this article we have demonstrated BEC （Bose-Einstein Condensate） which is the fifth state of matter. Recent development of gamma-ray laser and scenario of black holes are highlighted. From the stephen W. Hawking＇s lecture at kTH, stockholm and the experimental evidence of LIGO scientific collaboration, UsA, it is found that binary type black holes exist and can emit information which is detectable. Hence the source of information of black holes is not relay black; constituents are the densest state of material considered to be the fifth state and the primordial state of the Universe.

study Chromaticity of solar spectrum

The chromaticity of solar spectrum is studied with the help of a solar spectrum Monitor system that can detect individual color in the spectrum. Recent observations done by the detector on the solar Radiation and the Erath Milieu Experiment suggest that the sun’s visible spectral irradiance changes from May 2009 to september 2012. The data of Earth’s coordinates and environment have been taken since April 2005 after the devastating Tsunami (December, 2004) of India. The bizarre data of?zenith angle,?azimuth angle, and?temperature?of the Earth’s atmosphere show their changes of maxima and minima epoch to epoch. The data of solar spectrum monitor have been taking since 2009 and significant transformations of colored ratios?ΔRB/W?and?ΔRR/W?per hour are observed among the regions of each solstice between 2009 and 2012. The author advocates that the abrupt vagaries of the Earth’s movements may cause devastating tsunamis, earthquake, volcanic eruption, cyclones and tornadoes in addition of anomalous changes of solar spectral irradiation, humidity, temperature and pressure;those effects may spoil ecological balance and extinct some living species from the soil of the earth.

Effects of AG1 and AG2 QTLs on Nonstructural Carbohydrate and seed Management Options for Rice seedling Growth and Establishment under Flooding stress

Rice(Oryza sativa)plants acquired excess photosynthates in the form of nonstructural carbohydrates(NsCs)in their stems and *** keen interest in rice NsC,the dynamics of NsC accumulation,translocation and re-accumulation have not yet been well ***1 and AG2 QTLs associated with flooding tolerance through catalyzing starch into soluble sugar in germinating *** we conducted three experiments,greenhouse and field to lay the groundwork for large-scale diversity studies on grain NsC and some agronomic traits under direct-seeded rice(DsR)system,using elite lines incorporating AG1,AG2 and AG1-AG2 QTLs into the popular varieties PsB Rc82 and Ciherang-sub1 along with the donors kho Hlan On(AG1)and Ma-Zhan Red(AG2).In germinating seedlings,soluble sugars increased,while starch concentration decreased gradually especially in the tolerant checks and AG1-AG2 introgression lines under flooded *** sugar accumulation in stem started to increase from the vegetative stage and peaked at the panicle initiation stage then gradually decreased towards the maturity *** sub1-AG lines had higher sugar and starch concentrations at different growth stages than other genotypes in wet season 2016 and dry season *** survival rate was positively correlated with the stem NsC at the early vegetative stage(21 days after sowing),and stem NsC was positively associated with plant height at different growth *** the tested seeding rate,the most suitable seeding rate,4 g/m2 with shallow burial depth(0.5 cm),resulted in better seedling establishment,relatively higher seedling vigor index and higher leaf area index under flooding in DsR *** of AG1-AG2 QTLs had no any negative impact on nonstructural carbohydrate,germination rate,and growth and biomass production.

High yield expression of proteins in <i>E. coli</i>for NMR studies

In recent years, high yield expression of proteins in E. coli has witnessed rapid progress with developments of new methodologies and technologies. An important advancement has been the development of novel recombinant cloning approaches and protocols to express heterologous proteins for Nuclear Magnetic Resonance (NMR) studies and for isotopic enrichment. Isotope labeling in NMR is necessary for rapid acquisition of high dimensional spectra for structural studies. In addition, higher yield of proteins using various solubility and affinity tags has made protein over-expression cost-effective. Taken together, these methods have opened new avenues for structural studies of proteins and their interactions. This article deals with the different techniques that are employed for over-expression of proteins in E. coli and different methods used for isotope labeling of proteins vis-à-vis NMR spectroscopy.

Agricultural areas of land are deteriorating every day owing to population increase, rapid urbanization, and industrialization. To feed today’s huge populations, increased crop production is required from smaller areas, which warrants the continuous application of high doses of inorganic fertilizers to agricultural land. These cause damage to soil health and, therefore, nutrient imbalance conditions in arable soils. Under these conditions, the benefits of microbial inoculants (such as Actinobacteria) as replacements for harmful chemicals and promoting ecofriendly sustainable farming practices have been made clear through recent technological advances. There are multifunctional traits involved in the production of different types of bioactive compounds responsible for plant growth promotion, and the biocontrol of phytopathogens has reduced the use of chemical fertilizers and pesticides. There are some well-known groups of nitrogen-fixing Actinobacteria, such as Frankia, which undergo mutualism with plants and offer enhanced symbiotic *** addition to nitrogen fixation, increasing availability of major plant nutrients in soil due to the solubilization of immobilized forms of phosphorus and potassium compounds, production of phytohormones, such as indole-3-acetic acid, indole-3-pyruvic acid, gibberellins, and cytokinins, improving organic matter decomposition by releasing cellulases, xylanase, glucanases, lipases, and proteases, and suppression of soil-borne pathogens by the production of siderophores, ammonia, hydrogen cyanide, and chitinase are important features of Actinobacteria useful for combating biotic and abiotic stresses in *** positive influence of Actinobacteria on soil fertility and plant health has motivated us to compile this review of important findings associated with sustaining plant productivity in the long run.

Preliminary pharmacological evaluation of Alocasia indica schott tuber

To elucidate potential antioxidant, antidiarrheal, cytotoxic, and antibacterial activities of the ethanol extract of A/ocasia indica schott tuber in different experimental models established in vitro and in vivo. METHODs： In vitro antioxidant activity was evaluated by 2, 2-diphenyl-l-picrylhydrazyl （DPPH） radical-scavenging assay. Phenolic content was estimated by using Folin-Ciocalteu＇s reagent while reducing ability was measured by ferric reducing power assay./n vivo antidiarrheal studies were carried out in mice, and the activity was evaluated in castor oil and magnesium sulfate- induced diarrhea. Disk diffusion assay was utilized to determine antibacterial activity against a number of pathogenic bacterial strains. Acute toxicity test was carried out to measure the safe doses for the extract. REsULTs： In DPPH radical-scavenging assay, the extract exhibited strong radical-scavenging activity with the 50% inhibitory concentration value of 42.66 IJg/mL. Total phenolic content was found to be 542.26 mg gallic acid equivalent per 100 g of dried tuber extract, whereas flavonoid content was found to be 4.30 mg quercetin equivalent/g of dried tuber extract. In reducing power assay, the extract showed strong reducing power in a concentration-dependent manner. The extract significantly （P 〈 0.01） enhanced the latent period and decreased defecation in both castor oil- and magnesium sulfate-induced diarrhea. The extract also lessened gastrointestinal motility in mice. Potential antibacterial activity was exhibited by the extract against all the tested bacterial strains in disk diffusion assay. The 50% lethal concentration against brine shrimp nauplii was 81.09 μg/mL. CONCLUsION： The results demonstrated that the ethanol extract of A. inc/ica has potential antioxidant, antidiarrheal, cytotoxic, and antibacterial activity.

Technology advances in genomics,proteomics,and metabolomics largely expanded the pool of potential therapeutic *** with the in vitro setting,cell-based screening assays have been playing a key role in the processes of drug discovery and *** the commonly used strategies based on colorimetric and cell viability,we reason that methods that capture the dynamic cellular events will facilitate optimal hit identification with high sensitivity and ***,we propose a live-cell screening strategy using structured illumination microscopy (sIM) combined with an automated cell colocalization analysis software,CellprofilerTM,to screen and discover drugs for mitochondria and lysosomes interaction at a nanoscale resolution in living *** strategy quantitatively benchmarks the mitochondria-lysosome interactions such as mitochondria and lysosomes contact (MLC) and *** automatic quantitative analysis also resolves fine changes of the mitochondria-lysosome interaction in response to genetic and pharmacological ***-resolution live-cell imaging on the basis of quantitative analysis opens up new avenues for drug screening and development by targeting dynamic organelle interactions at the nanoscale resolution,which could facilitate optimal hit identification and potentially shorten the cycle of drug discovery.
