Transcriptional coactivators regulate the rate of gene expression in the *** receptor coactivator 6(NCOA6),a coactivator,has been implicated in embryonic development,metabolism,and cancer pathogenesis,but its role in innate immunity and inflammatory diseases remains ***,we demonstrated that NCOA6 was expressed in monocytes and macrophages and that its level was increased under proinflammatory ***,nuclear NCOA6 was found to translocate to the cytoplasm in activated monocytes and then become incorporated into the inflammasome with NLRP3 and AsC,forming cytoplasmic ***,NCOA6 associated with the ATP hydrolysis motifs in the NACHT domain of NLRP3,promoting the oligomerization of NLRP3 and AsC and thereby instigating the production of IL-1βand active *** note,Ncoa6 deficiency markedly inhibited NLRP3 hyperactivation caused by the Nlrp3^(R258W) gain-of-function mutation in *** ablation of Ncoa6 substantially attenuated the severity of two NLRP3-dependent diseases,folic-induced acute tubular necrosis and crystal-induced arthritis,in *** with these findings,NCOA6 was highly expressed in macrophages derived from gout patients,and NCOA6-positive macrophages were significantly enriched in gout macrophages according to the transcriptome profiling ***,NCOA6 is a critical regulator of NLRP3 inflammasome activation and is therefore a promising target for NLRP3-dependent diseases,including gout.

With the advancement of wireless network technology,vast amounts of traffic have been generated,and malicious traffic attacks that threaten the network environment are becoming increasingly *** signature-based detection methods,static analysis,and dynamic analysis techniques have been previously explored for malicious traffic detection,they have limitations in identifying diversified malware traffic *** research has been focused on the application of machine learning to detect these ***,applying machine learning to lightweight devices like IoT devices is challenging because of the high computational demands and complexity involved in the learning *** this study,we examined methods for effectively utilizing machine learning-based malicious traffic detection approaches for lightweight *** introduced the suboptimal feature selection model(sFsM),a feature selection technique designed to reduce complexity while maintaining the effectiveness of malicious traffic *** performance was evaluated on various malicious traffic,benign,exploits,and generic,using the UNsW-NB15 dataset and sFsM sub-optimized hyperparameters for feature selection and narrowed the search scope to encompass all *** improved learning performance while minimizing complexity by considering feature selection and exhaustive search as two steps,a problem not considered in conventional *** experimental results showed that the detection accuracy was improved by approximately 20%compared to the random model,and the reduction in accuracy compared to the greedy model,which performs an exhaustive search on all features,was kept within 6%.Additionally,latency and complexity were reduced by approximately 96%and 99.78%,respectively,compared to the greedy *** study demonstrates that malicious traffic can be effectively detected even in lightweight device *** verified the possibility of detecting various attack

Plants are aerobic organisms that rely on molecular oxygen for respiratory energy *** conditions,with oxygen levels ranging between 1%and 5%,usually limit aerobic respiration and affect plant growth and ***,we demonstrate that the hypoxic microenvironment induced by active cell proliferation during the two-step plant regeneration process intrinsically represses the regener-ation competence of the callus in Arabidopsis *** showed that hypoxia-repressed plant regener-ation is mediated by the RELATED TO APETALA2.12(RAP2.12)protein,a memberof the Ethylene Response Factor VIl(ERF-Vll)*** found that the hypoxia-activated RAP2.12 protein promotes salicylic acid(sA)biosynthesis and defense responses,thereby inhibiting pluripotency acquisition and de novo shoot regeneration in *** and genetic analyses revealed that RAP2.12 could bind directly to the sALICyLIC ACID INDUCTION DEFICIENT 2(sID2)gene promoter and activate sA biosynthesis,repressing plant regeneration possibly via a PLETHORA(PLT)-dependent ***,the rap2.12 mutant calli exhibits enhanced shoot regeneration,which is impaired by sA *** together,these find-ings uncover that the cell proliferation-dependent hypoxic microenvironment reduces cellular pluripotency and plant regeneration through the RAP2.12-sID2 module.

Recommendation Information systems(RIs)are pivotal in helping users in swiftly locating desired content from the vast amount of information available on the *** Convolution Network(GCN)algorithms have been employed to implement the RIs ***,the GCN algorithm faces limitations in terms of performance enhancement owing to the due to the embedding value-vanishing problem that occurs during the learning *** address this issue,we propose a Weighted Forwarding method using the GCN(WF-GCN)*** proposed method involves multiplying the embedding results with different weights for each hop layer during graph *** applying the WF-GCN algorithm,which adjusts weights for each hop layer before forwarding to the next,nodes with many neighbors achieve higher embedding *** approach facilitates the learning of more hop layers within the GCN *** efficacy of the WF-GCN was demonstrated through its application to various *** the MovieLens dataset,the implementation of WF-GCN in LightGCN resulted in significant performance improvements,with recall and NDCG increasing by up to+163.64%and+132.04%,***,in the *** dataset,LightGCN using WF-GCN enhanced with WF-GCN showed substantial improvements,with the recall and NDCG metrics rising by up to+174.40%and+169.95%,***,the application of WF-GCN to self-supervised Graph Learning(sGL)and simple Graph Contrastive Learning(simGCL)also demonstrated notable enhancements in both recall and NDCG across these datasets.

Tip-enhanced nano-spectroscopy and-imaging have significantly advanced our understanding of low-dimensional quantum materials and their interactions with light,providing a rich insight into the underlying physics at their natural length ***,various functionalities of the plasmonic tip expand the capabilities of the nanoscopy,enabling dynamic manipulation of light-matter interactions at the *** this review,we focus on a new paradigm of the nanoscopy,shifting from the conventional role of imaging and spectroscopy to the dynamical control approach of the tip-induced light-matter *** present three different approaches of tip-induced control of light-matter interactions,such as cavity-gap control,pressure control,and near-field polarization ***,we discuss the nanoscale modifications of radiative emissions for various emitters from weak to strong coupling regime,achieved by the precise engineering of the ***,we introduce recent works on light-matter interactions controlled by tip-pressure and near-field polarization,especially tunability of the bandgap,crystal structure,photoluminescence quantum yield,exciton density,and energy transfer in a wide range of quantum *** envision that this comprehensive review not only contributes to a deeper understanding of the physics of nanoscale light-matter interactions but also offers a valuable resource to nanophotonics,plasmonics,and materials science for future technological advancements.

Most microfluidic separation techniques rely largely on object size as a separation *** ability to separate micro-objects based on their shape is crucial in various biomedical and chemical ***,we develop an ondemand,label-free acoustofluidic method to separate prolate ellipsoids from spherical microparticles based on traveling surface acoustic wave-induced acoustic radiation force and *** freely rotating non-spherical microobjects were aligned under the progressive acoustic field by the counterrotating radiation torque,and the major axis of the prolate ellipsoids was parallel to the progressive wave *** specific alignment of the ellipsoidal particles resulted in a reduction in the cross-sectional area perpendicular to the wave *** a consequence,the acoustic backscattering decreased,resulting in a decreased magnitude of the radiation *** the variation in radiation force,which depended on the micro-object morphology enabled the acoustofluidic shape-based *** conducted numerical simulations for the wave scattering of spherical and prolate objects to elucidate the working mechanism underlying the proposed method.A series of experiments with polystyrene microspheres,prolate ellipsoids,and peanut-shaped microparticles were performed for *** quantitative analysis of the separation efficiency,we confirmed the high purity and high recovery rate of the proposed acoustofluidic shapebased separation of *** a bioparticle,we utilize Thalassiosira eccentrica to perform shape-based separation,as the species has a variety of potential applications in drug delivery,biosensing,nanofabrication,bioencapsulation and immunoisolation.
