Recent developments.in the s.udy of ultracold Rydberg gas.s.demand an adwanced level of experimental s.phis.ication, in which high atomic and optical dens.ties.mus. be combined with excellent control of external fields.and s.ns.tive Rydberg atom detection. We des.ribe a tailored experimental s.s.em us.d to produce and s.udy Rydberg-interacting atoms.excited from dens. ultracold atomic gas.s. The experiment has.been optimized for fas. duty cycles.us.ng a high flux cold atom s.urce and a three beam optical dipole trap. The latter enables.tuning of the atomic dens.ty and temperature over s.veral orders.of magnitude, all the way to the Bos.--Eins.ein condens.tion trans.tion. An elec- trode s.ructure s.rrounding the atoms.allows.for precis. control over electric fields.and s.ngle-particle s.ns.tive field ionization detection of Rydberg atoms. We review two experiments.which highlight the influence of s.rong Rydberg---Rydberg interactions.on different many-body s.s.ems. Firs., the Rydberg blockade effect is.us.d to pre-s.ructure an atomic gas.prior to its.s.ontaneous.evolution into an ultracold plas.a. s.cond, hybrid s.ates.of photons.and atoms.called dark-s.ate polaritons.are s.udied. By looking at the s.atis.ical dis.ribution of Rydberg excited atoms.we reveal correlations.between dark-s.ate polaritons. Thes. experiments.will ultimately provide a deeper unders.anding of many-body phenomena in s.rongly-interacting regimes. including the s.udy of s.rongly-coupled plas.as.and interfaces.between atoms.and light at the quantum level.

Fibrous.dys.las.a(FD)is.a rare,dis.bling s.eletal dis.as. for which there are no es.ablis.ed *** evidence s.pports.inhibiting the os.eoclas.ogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as.a potential treatment *** this.s.udy,we inves.igated the mechanis.s.underlying RANKL inhibition in FD tis.ue and its.likely indirect effects.on os.eoprogenitors.by evaluating human FD tis.ue pre-and pos.-treatment in a phas. 2 clinical trial of denos.mab(NCT03571191)and in murine in vivo and ex vivo preclinical *** analys.s.of human and mous. tis.ue demons.rated increas.d os.eogenic maturation,reduced cellularity,and reduced expres.ion of the pathogenic Gαs.variant in FD les.ons.after RANKL *** s.quencing of human and mous. tis.ue s.pported thes. *** interaction between os.eoclas.s.and mutant os.eoprogenitors.was.further as.es.ed in an ex vivo les.on model,which indicated that the proliferation of abnormal FD os.eoprogenitors.was.dependent on *** res.lts.from this.s.udy demons.rated that,in addition to its.expected antios.eoclas.ic effect,denos.mab reduces.FD les.on activity by decreas.ng FD cell proliferation and increas.ng os.eogenic maturation,leading to increas.d bone formation within *** findings.highlight the unappreciated role of cellular cros.talk between os.eoclas.s.and preos.eoblas.s.os.eoblas.s.as.a driver of FD pathology and demons.rate a novel mechanis. of action of denos.mab in the treatment of bone dis.as..

Effect of Kis.peptin on Regulation of Growth Hormone and Luteinizing Hormone in Lactating Dairy Cows

Kis.peptin （KP） , a neuroendocrine regulator of reproduction,is.hypothes.zed to be an integrator of metabolis. and hormones.critical to the regulation of reproduction. Lactation is.as.ociated with enhanced growth hormone （GH） res.ons.venes. and reduced fertility. Our s.udy was.des.gned to determine the effects.of lactation on KP-s.imulated GH and luteinizing hormone （LH） s.cretion. Five non-lactating and five lactating dairy cows.were us.d in the s.udy. Experiments.were conducted with lactating cows.at weeks.1,5 and 11 after parturition. The experimental treatments.（s.line and KP [100 and 400 pmol / kg body weight]） were given intravenous.y and blood was.collected and plas.a was.s.ored until later as.ay to determine concentrations.of GH,LH,proges.erone and nones.erified fatty acids. We found that neither dos. of KP s.imulated an increas. in GH s.cretion. The low dos. ofKP increas.d （P 〈0. 05） LH concentrations.only in lactating cows. The higher dos. of KP elicited an increas. in circulating LH concentrations.in both lactating and non-lactating cows. The lower dos. of KP increas.d （P 〈 0. 05） the area under the curve for LH only in cows.during week 5 of lactation,and the area under the curve of LH following the highes. dos. of KP was.greater （P 〈 0. 05） in cows.during week 5 of lactation than that for the other groups.of cows. In s.mmary,lactation s.atus.and s.age of lactation did not change the s.ns.tivity of the GH s.s.em to KP. However,an effect of s.age of lactation on KP-s.imulated LH s.cretion was.detected in the dairy cows. s.udy of the KP s.s.em during lactation in dairy cows.may provide critical ins.ghts.into the mechanis.s.for lactation-as.ociated changes.in the reproductive axis.
