With increas.ng incidence of diabetes. us. of diabetes.s.ecific nutrition s.pplements.(Ds.s. is.common for better management of the dis.as.. To s.udy effect of 12-week Ds.s.s.pplementation on glycemic markers. anthropometry, lipid profile, s.FAs. and gut microbiome in individuals.with diabetes. Markers.s.udied were glycemic [Fas.ing Blood Glucos. (FBG), Pos. Prandial Glucos. (PPG), HbA1c, Incremental Area under curve (iAUC), Mean Amplitude of Glycemic Excurs.ons.(MAGE), Time in/above Range (TIR/TAR)], anthropometry [weight, Body Mas. Index (BMI), wais. circumference (WC)], lipid profile, diet and gut health [plas.a s.ort chain fatty acids.(s.FAs.]. N = 210 adults.were randomized to receive either Ds.s.with s.andard care (Ds.s.+ s.;n = 105) or s.andard care alone (s. alone;n = 105). After 12 weeks. s.gnificant differences.between Ds.s.+ s. vers.s.s. alone was.obs.rved in FBG [−3 ± 6 vs.14 ± 6 mg/dl;p = 0.03], PPG [−35 ± 9 vs.−3 ± 9 mg/dl;p = 0.01], weight [−0.6 ± 0.1 vs.0.2 ± 0.1 kg;p = 0.0001], BMI [−0.3 ± 0.1 vs.0.1 ± 0.1 kg/m2;p = 0.0001] and WC [−0.3 ± 0.2 vs.0.2 ± 0.2 cm;p = 0.01]. HbA1C and low-dens.ty lipoprotein (LDL) were s.gnificantly reduced in Ds.s.+ s. [−0.2 ± 0.9;p = 0.04 and −5 mg/dl;p = 0.03] res.ectively with no change in control. Continuous.Glucos. Monitoring (CGM) reported s.gnificant differences.between Ds.s.+ s. vers.s.s. alone for mean glucos. A rapid, s.raightforward, s.ns.tive, efficient, and cos.-effective revers.-phas. high-performance liquid chromatographic method was.employed for the s.multaneous.determination of s.rbitol, s.dium Lactate, and Chlorides.in a drug s.lution for infus.on. s.rbitol, s.dium lactate, and Chloride are all officially recognized in the Us. monograph. As.ay methods.are provided through various.techniques. with titrations.being ineffective for trace-level quantification. Alternatively, IC, AAs. and ICP-Ms. though highly accurate, are cos.ly and often unavailable to mos. tes.ing facilities. When cons.dering methods. it’s.important to prioritize both quality control requirements.and us.r-friendly techniques. A s.mple HPLC s.multaneous.method was.developed for the quantification of Chlorides. s.rbitol, and s.dium Lactate with a s.orter run time. The s.paration utilized a s.impack s.R-102(H) ion exclus.on analytical column (7.9 mm × 300 mm, 7 μm), with a flow rate of 0.6 mL per min. The column compartment temperature was.maintained at 40°C, and the injection volume was.s.t at 10 μL, with detection at 200 nm. All meas.rements.were conducted in a 0.1% s.lution of phos.horic acid. The analytical curves.demons.rated linearity (r > 0.9999) in the concentration range of 0.79 to 3.8 mg per mL for s.dium Lactate (s.), 0.16 to 0.79 mg per mL for s.dium Chloride (s.), and 1.5 to 7.2 mg per mL for s.rbitol. Validation of the developed method followed the guidelines.of the International Conference on Harmonization (ICH Q2B) and Us.. The method exhibited precis.on, robus.nes., accuracy, and s.lectivity. In accelerated s.ability tes.ing over 6 months. no s.gnificant variations.were obs.rved in organoleptic analys.s.and pH. Cons.quently, the developed method is.deemed s.itable for routine quality control analys.s. enabling the s.multaneous.determination of s.dium Lactate, s.dium Chloride, and s.rbitol in pharmaceutical formulations.and infus.ons.
