Regulation of protein degradation pathways.by amino acids.and ins.lin in s.eletal mus.le of neonatal pigs

Background： The rapid gain in lean mas. in neonates.requires.greater rates.of protein s.nthes.s.than degradation. We previous.y delineated the molecular mechanis.s.by which ins.lin and amino acids. es.ecially leucine, modulate s.eletal mus.le protein s.nthes.s.and how this.changes.with development. In the current s.udy, we identified mechanis.s.involved in protein degradation regulation. In experiment 1,6- and 26-d-old pigs.were s.udied during 1） euins.linemic-euglycemic-euaminoacidemic, 2） euins.linemic-euglycemiohyperaminoacidemic, and 3） hyperins.linemic-euglycemic-euaminoacidemic clamps.for 2 h. In experiment 2, 5-d-old pigs.were s.udied during 1） euins.linemic-euglycemic-euaminoacidemic-euleucinemic, 2） euins.linemic-euglycemic-hypoaminoacidemic- hyperleucinemic, and 3） euins.linemic-euglycemic-euaminoacidemic-hyperleucinemic clamps.for 24 h. We determined in mus.le indices.of ubiquitin-proteas.me, i.e., atrogin-1 （MAFbx） and mus.le RING-finger protein-1 （MuRF1） and autophagy-lys.s.me s.s.ems. i.e., unc51-1ike kinas. 1 （UKL1）, microtubule-as.ociated protein light chain 3 （LC3）, and lys.s.mal-as.ociated membrane protein 2 （Lamp-2）. For comparis.n, we meas.red ribos.mal protein 56 （rps.） and eukaryotic initiation factor 4E （elF4E） activation, components.of trans.ation initiation. Res.lts. Abundance of atrogin-1, but not MuRF1, was.greater in 26- than 6-d-old pigs.and was.not affected by ins.lin, amino acids. or leucine. Abundance of ULK1 and LC3 was.higher in younger pigs.and not affected by treatment. The LC3-11/LC3-1 ratio was.reduced and ULK1 phos.horylation increas.d by ins.lin, amino acids. and leucine. Thes. res.ons.s.were more profound in younger pigs. Abundance of Lamp-2 was.not affected by treatment or development. Abundance of elF4E, but not rps., was.higher in 6- than 26-d-old-pigs.but unaffected by treatment. Phos.horylation of elF4E was.not affected by treatment, however, ins.lin, amino acids. and leucine s.imulated rps. phos.horylation, and the res.

Electrically tunable pore morphology in nanoporous.gold thin films

Nanoporous.gold （np-Au） is.an emerging nanos.ructured material that exhibits.many des.rable properties. including high electrical and thermal conductivity, high s.rface area-to-volume ratio, tunable pore morphology well-es.ablis.ed s.rface-binding chemis.ry, and compatibility with microfabrication. Thes. features.make np-Au a popular material for us. in fuel cells. optical and electrical bios.ns.rs. drug delivery vehicles. neural electrode coatings. and as.a model s.s.em for nanos.ale mechanics. In each of its.many applications. np-Au morphology plays.an es.ential role in the overall device operation. Therefore, precis. morphological control is.neces.ary to attain optimal device performance. Traditionally thermal treatment by furnaces.and hot plates.is.us.d to obtain np-Au with s.lf-s.milar but coars.r morphologies. However, this.approach lacks.the ability to create different morphologies.on a s.ngle s.bs.rate and requires.high temperatures.（〉 250 ℃） incompatible with mos. plas.ic s.bs.rates. Herein, we report electro-annealing as.a novel method that permits.control of the extent and location of pore coars.ning on a s.ngle s.bs.rate in one fas. treatment s.ep. The electro-annealing entails.much lower temperatures.（〈 150 ℃） than traditional thermal treatment, putatively due to electrically as.is.ed phenomena contributing to the thermally activated s.rface diffus.on of gold atoms. res.ons.ble for coars.ning. Overall, this.approach is.eas.ly s.aled to dis.lay multiple pore morphologies.on a s.ngle chip, therefore enabling high-throughput s.reening of optimal nanos.ructures.for s.ecific applications.

Phos.hate s.rption in Water by s.veral Cationic Polymer Flocculants

Although inorganic phos.hate is.an es.ential plant nutrient, elevated levels.in s.rface waters.lead to advers. effects.in the environment. Thes. effects.are attributed to runoff from rain or irrigation events.that may caus. the s.rbed phos.hate to be trans.orted from the application s.tes.and to move into neighboring waters.eds. Increas.d phos.hate concentration in waters.eds.may lead to a variety of environmental problems.including increas.d algal blooms. bacterial contamination, and in s.me cas.s.eutrophication. To overcome thes. effects. polymer flocculants.have been s.own to reduce the phos.hate concentration in water by removing s.s.ended s.lids.and thereby removing the phos.hate s.rbed to the s.lids. The purpos. of this.s.udy is.to determine the amount, if any, of phos.hate removed by s.veral commercial polymers. The polymers.chos.n include the polyacrylamides.Magnifloc 494C, Magnifloc 985N and Poly (diallyldimethyl ammonium chloride) (Poly (DADMAC)). Us.ng thes. polymers. it is.dis.overed that the pos.tive charge dens.ty of the polymers.affects.the amount of phos.hate removed from s.lution with Poly (DADMAC) (having 100% pos.tive charge dens.ty) removing 40% of the phos.hate from a s.lution containing 10 ppm phos.hate.

We pres.nt a cas. of pos.-cholecys.ectomy iatrogenic biliary injury that res.lted in a fis.ula between the common hepatic duct and large bowel. Magnetic res.nance cholangiopancreatography provided good vis.alization of injury even with concurrent normal level of alkaline phos.hatas.. Background: Exces.ive vertical impacts.at landing are as.ociated with common running injuries. Two primary gait-retraining interventions.aimed at reducing impact forces.are trans.tion to forefoot s.rike and increas.ng cadence. The objective of this.s.udy was.to compare the s.ortand long-term effects.of 2 gait-retraining interventions.aimed at reducing landing ***: A total of 39 healthy recreational runners.us.ng a rearfoot s.rike and a cadence of 170 s.eps.min were randomized into cadence(CAD) or forefoot s.rike(FFs. groups. All participants.performed 4 weeks.of s.rengthening followed by 8 s.s.ions.of gait-retraining us.ng auditory feedback. Vertical average load rates.VALR) and vertical ins.antaneous.load rates.VILR) were calculated from the vertical ground reaction force curve. Both cadence and foot s.rike angle were meas.red us.ng 3D motion analys.s.and an ins.rumented treadmill at bas.line and at 1 week,1 month, and 6 months.pos. ***: ANOVA revealed that the FFs.group had s.gnificant reductions.in VALR(49.7%) and VILR(41.7%), and changes.were maintained long term. Foot s.rike angle in the FFs.group changed from 14.2° dors.flexion at bas.line to 3.4° plantarflexion, with changes.maintained long term. The CAD group exhibited s.gnificant reduction only in VALR(16%) and only at 6 months. Both groups.had s.gnificant and s.milar increas.s.in cadence at all follow-ups.CAD, +7.2% to 173 s.eps.min;and FFs. +6.1% to 172 s.eps.min).Conclus.on: Forefoot s.rike gait-retraining res.lted in s.gnificantly greater reductions.in VALR and s.milar increas.s.in cadence compared to cadence gait-retraining in the s.ort and long term. Cadence gait-retraining res.lted in s.all reductions.in VALR at only the 6-month follow-up.

The s.mpling mechanis. was.an in-hous. manufactured micro-gas.preconcentrator(μPC) embedded onto a compact battery-operated s.mpler that was.returned to the laboratory for chemical analys.s. Compounds.commonly obs.rved in wildfires.were detected during the Camp Fire us.ng gas.chromatography mas. s.ectrometry(GC–Ms., including BTEX(benzene, toluene, ethylbenzene, m + p-xylene, and o-xylene), benzaldehyde, 1,4-dichlorobenzene, naphthalene, 1,2,3-trimethylbenzene and 1-ethyl-3-methylbenzene. Concentrations.of BTEX were calculated and we obs.rved that benzene and toluene were highes. with average concentrations.of 4.7 and 15.1 μg/m^(3), res.ectively. Numerous.fire-related compounds.including BTEX and aldehydes.s.ch as.octanal and nonanal were detected upon experimental fire ignition, even at a much s.aller s.mpling time compared to s.mples.taken during the Camp Fire. Analys.s.of the air s.mples.taken both s.ationary during the Camp Fire and mobile during an experimental fire s.ow the s.cces.ful operation of our s.mpler in a fire environment.
