Challenges facing the UsA in matching Germany on advanced manufacturing for green growth

This study explores the relative strength of public policies promoting advanced manufacturing by comparing the government programs and capacities of two world leaders in science and technology:the UsA and Germany.Based on insights from’varieties-of-capitalism’literature and using process tracing methodology,this study identifies significant differences in today’s pursuit of advanced manufacturing.The German model of Rhine capitalism has strength due to its long tradition of diversified quality work,built on a long-term commitment to government-industry collaboration,with significant indirect public support provided by vocational training and research institutes.The American model of Manchester capitalism has been challenged by the loss of millions of low-skilled manufacturing jobs due to outsourcing and competition from Asia.The study finds that the“Manufacturing UsA”policy initiative of the Obama era,although directly inspired by Germany’s Fraunhofer institutes,is hampered by the contextual and institutional circumstances identified.With biobased carbon fibers offering the potential of a novel,low-cost material,pursued with research and development(R&D)efforts in both countries,it provides a measure for evaluating policy outcomes,and the study confirms Germany’s technological lead.In addition,the study’s process-tracing methodology reveals that significant legacies from German knowledge creation in the past have spilled over to American science and technology,and may continue to do so,lured by government funds.Further studies on advanced manufacturing should test the robustness of these findings.

Impaired contractility and remodeling of the upper gastrointestinal tract in diabetes mellitus type-1

AIM: To investigate that both the neuronal function of the contractile system and structural apparatus of the gastrointestinal tract are affected in patients with longstanding diabetes and auto mic neuropathy. METHODs: The evoked esophageal and duodenal contractile activity to standardized bag distension was assessed using a specialized ultrasound-based probe. Twelve type-1 diabetic patients with autonomic neuropathy and severe gastrointestinal symptoms and 12 healthy controls were studied. The geometry and biomechanical parameters (strain, tension/stress, and stiffness) were assessed. REsULTs: The diabetic patients had increased frequency of distension-induced contractions (6.0 ± 0.6 vs 3.3 ± 0.5, P s increased reactivity was correlated with the duration of the disease (P = 0.009). Impaired coordination of the contractile activity in diabetic patients was demonstrated as imbalance between the time required to evoke the first contraction at the distension site and proximal to it (1.5 ± 0.6 vs 0.5 ± 0.1, P = 0.03). The esophageal wall and especially the mucosa-submucosa layer had increased thickness in the patients (P ssive stretch was less in diabetics (P sophageal and duodenal wall stiffness and circumferential deformation induced by the distensions were not affected in the patients (all P > 0.14). CONCLUsION: The impaired contractile activity with an imbalance in the distension-induced contractions likely reflects neuronal abnormalities due to autonomic neuropathy. However, structural changes and remodeling of the gastrointestinal tract are also evident and may add to the neuronal changes. This may contribute to the pathophysiology of diabetic gut dysfunction and impact on future management of diabetic patients with gastrointestinal symptoms.

Rethinking neurodegenerative diseases:neurometabolic concept linking lipid oxidation to diseases in the central nervous system

Currently,there is a lack of effective medicines capable of halting or reve rsing the progression of neurodegenerative disorde rs,including amyotrophic lateral sclerosis,Parkinson s disease,multiple sclerosis,or Alzheimer s disease.Given the unmet medical need,it is necessary to reevaluate the existing para digms of how to to rget these diseases.When considering neurodegenerative diseases from a systemic neurometabolic perspective,it becomes possible to explain the shared pathological features.This innovative approach presented in this paper draws upon exte nsive research conducted by the authors and researchers worldwide.In this review,we highlight the importance of metabolic mitochondrial dysfunction in the context of neurodegenerative diseases.We provide an overview of the risk factors associated with developing neurodegenerative disorders,including genetic,epigenetic,and environmental fa ctors.Additionally,we examine pathological mechanisms implicated in these diseases such as oxidative stress,accumulation of misfolded proteins,inflammation,demyelination,death of neurons,insulin resistance,dysbiosis,and neurotransmitter disturbances.Finally,we outline a proposal for the restoration of mitochondrial metabolism,a crucial aspect that may hold the key to facilitating curative therapeutic interventions for neurodegenerative disorders in forthcoming advancements.

Investigation of Hexavalent Chromium Reduction in strong saline and Acidic Nitro-Phosphate solutions

Phosphorus fertilizers from less pure sedimentary sources become increasingly important, due to depletion of phosphorus from igneous rock of high quality. Consequently, robust methods with potential to remove various types of hazardous elements are required. Among such impurities, hexavalent chromium (Cr(VI)) is very likely to become a future challenge. Different industrial ways to treat phosphate rock are currently being practised, and we have here studied how chromium behaves when using the nitro-phosphate process. The reduction mechanism of Cr (VI) in nitric acid and phosphoric acid solutions was investigated by measuring redox potential and UV-VIs spectra. The results show that Cr (VI) is not stable in strong nitric acid solutions. Reduction of Cr (VI) species decreased with decreasing temperature, NOsub>2sub> concentration, ionic strength and absence of light. Extracellular Vesicles from Mesenchymal stromal Cells (imEVs) Improve Cold Preservation of Isolated Mitochondria

Mitochondrial organelle transplantation (MOT) is an innovative strategy for the treatment of mitochondrial dysfunction such as cardiac ischemic reperfusion injuries, Parkinson’s diseases, brain and spinal cord injuries, and amyotrophic lateral sclerosis (ALs). However, one of the major challenges for widespread usage is a methodology for preservation of isolated mitochondria. Extracellular vesicles (EVs) are phospholipid bilayer-enclosed vesicles released from cells. EVs carry a cargo of proteins, nucleic acids, lipids, metabolites, and even organelles such as mitochondria. Purpose: To test if EVs enhance the stability of isolated mitochondria. Methods: We mixed isolated mitochondria of fibroblasts with EVs of mesenchymal stromal cells (imEVs) (9:1 in volume) and stored the mixture at 2°C - 6°C for different time periods. We measured morphology, mitochondrial membrane potential (MMP) and mitochondrial ATP content at 0, 2, 5 days. Key findings: After 2 days of storage, the mito-chondria without imEVs lost approximate 70% MMP (RFU: 1822 ± 68), compared to the fresh mitochondria (RFU: 5458 ± 52) (p 0.05). In agreement with MMP, mitochondria without imEVs lost significant mitochondrial ATP content (p 0.05), after 2 days of cold storage, compared to fresh mitochondria. Microscopy showed that imEVs promoted aggregation of isolated mitochondria. summary: The preliminary data showed that imEVs enhanced the stability of isolated mitochondria in cold storage.

Novel immune modulatory vaccines targeting TGFβ

Immune modulatory vaccines (IMVs) are novel therapies designed to activate anti-regulatory T cells (anti-Tregs) [1]. Anti-Tregs recognize HLA-restricted antigens on the surface of regulatory immune cells derived from tumor microenvironment antigens (TMAs) [2], such as IDO [3], PD-L1 [4], and TGFβ [5, 6]. Having established that specific T cells recognize target cells in a TGFβ-dependent manner, we reported that such TGFβ-specific T cells are associated with clinical benefit and improved survival after combined treatment with radiotherapy (RT) and immune checkpoint inhibitors (ICIs) in patients with pancreatic cancer (PC) [7]. Additionally, we found that a TGFβ-based IMV impacted tumor growth in murine models of PC via immune modulation of the tumor microenvironment (TME) [8]. These data suggest that combining a TGFβ-based IMV with RT/ICI therapy will be beneficial for patients with PC.

Levels of L-carnitine in human seminal plasma are associated with sperm fatty acid composition

The fatty acid composition of spermatozoa has been shown to be important for their function,and L-carnitine is crucial for fatty acid metabolism.Its levels in the seminal plasma positively correlate with semen quality,whereas high body mass index(BMI)is associated with both reduced semen quality and altered sperm fatty acid composition.Here,we examined the associations between free seminal L-carnitine levels and sperm fatty acid composition as well as BMI.semen samples were collected and analyzed from 128 men with unknown fertility status and with BMI ranging from 19 kg m^(−2) to 63 kg m^(−2).sperm fatty acid composition was assessed by gas chromatography,while free seminal L-carnitine analysis was performed using high-performance liquid chromatography.Multiple linear regression analysis showed a positive correlation of free seminal L-carnitine levels with the amount of sperm palmitic acid(β=0.21;P=0.014),docosahexaenoic acid(DHA;β=0.23;P=0.007),and total n-3 polyunsaturated fatty acids(β=0.23;P=0.008)and a negative correlation of free seminal L-carnitine levels with lignoceric acid(β=−0.29;P=0.001)and total n-6 polyunsaturated fatty acids(β=−0.24;P=0.012)when adjusted for covariates.There was no relationship between free seminal L-carnitine levels and BMI.since free seminal L-carnitine levels are associated with semen quality,the absence of a correlation with BMI suggests that reduced semen quality in obese men is independent of seminal L-carnitine.

Linking monitoring and modelling for river basin management:Danish experience with combating nutrient loadings to the aquatic environment from point and non-point sources

Nationwide monitoring of the aquatic environment was initiated in 1988 in Denmark as a means to follow the outcome of the Action Plans for nutrient pollution of the aquatic environment. Five Action Plans have been adopted by the Danish Parliament since 1985 and the nationwide monitoring programme can be used to quantify the outcome as shown by reductions in nutrient discharges from both point and non-point sources. Moreover, the empirical experience gathered from nearly 20 years of monitoring is assisting the development and calibration of models for simulation of nitrogen leaching, nitrogen removal in groundwater and surface waters and the establishment of a P-index all covering the entire land area of Denmark.

Total Variation Based Parameter-Free Model for Impulse Noise Removal

We propose a new two-phase method for reconstruction of blurred im-ages corrupted by impulse noise.In the first phase,we use a noise detector to iden-tify the pixels that are contaminated by noise,and then,in the second phase,we reconstruct the noisy pixels by solving an equality constrained total variation mini-mization problem that preserves the exact values of the noise-free pixels.For images that are only corrupted by impulse noise(i.e.,not blurred)we apply the semismooth Newton’s method to a reduced problem,and if the images are also blurred,we solve the equality constrained reconstruction problem using a first-order primal-dual algo-rithm.The proposed model improves the computational efficiency(in the denoising case)and has the advantage of being regularization parameter-free.Our numerical results suggest that the method is competitive in terms of its restoration capabilities with respect to the other two-phase methods.

Understanding the effects of structured self-assessment in directed,self-regulated simulation-based training of mastoidectomy:A mixed methods study

Objective:self-directed training represents a challenge in simulation-based training as low cognitive effort can occur when learners overrate their own level of performance.This study aims to explore the mechanisms underlying the positive effects of a structured self-assessment intervention during simulation-based training of mastoidectomy.Methods:A prospective,educational cohort study of a novice training program consisting of directed,self-regulated learning with distributed practice(5x3 procedures)in a virtual reality temporal bone simulator.The intervention consisted of structured self-assessment after each procedure using a rating form supported by small videos.semi-structured telephone interviews upon completion of training were conducted with 13 out of 15 participants.Interviews were analysed using directed content analysis and triangulated with quantitative data on secondary task reaction time for cognitive load estimation and participants’self-assessment scores.Results:six major themes were identified in the interviews:goal-directed behaviour,use of learning supports for scaffolding of the training,cognitive engagement,motivation from self-assessment,selfassessment bias,and feedback on self-assessment(validation).Participants seemed to self-regulate their learning by forming individual sub-goals and strategies within the overall goal of the procedure.They scaffolded their learning through the available learning supports.Finally,structured self-assessment was reported to increase the participants’cognitive engagement,which was further supported by a quantitative increase in cognitive load.Conclusions:structured self-assessment in simulation-based surgical training of mastoidectomy seems to promote cognitive engagement and motivation in the learning task and to facilitate self-regulated learning.
