somatostatin is a neuropeptide and a key regulator of the growth *** sst encoding genes(sst1 to sst6)have been identified in teleost fish genomes but little is known about their *** present study aimed at replicating the context of the inflammatory bowel disease(IBD)and clarifying the involvement of sst3 in the intestine innate defence barrier in zebrafish *** first established a CRIsP/Cas9 sst3 deficient line(Mt)and analysed the morphological and transcriptomic response to 0.4%dextran sulfate sodium(Dss).Alcian blue staining of larval sections 6 days post fertilization showed an increased in acidic mucins in the intestinal bulb and mid-intestine,with a much stronger response in the Mt compared to wild type(Wt).the transcriptomic analysis revealed that Wt and Mt shared enriched gene ontology(GO)terms and pathways linked to catabolism,chondrocyte development,innate immune system,xenobiotic metabolism and oxidative *** contrast,the Wt specific response to Dss was enriched in GO terms and pathways linked to transcription and translation,various developmental processes,regulation of biosynthetic processes,apelin signalling and apoptosis while the Mt specific response included terms and pathways linked to protein metabolism and catabolic processes,extracellular matrix-receptor interaction and proteasome and chondrocyte ***,this study demonstrated that sst3 deficiency impairs insulin growth factor and adipocytokine signalling exacerbating the inflammatory response to Dss.

somatostatin(sst)plays important roles in growth and *** teleost fishes six sst encoding genes(sst1 to sst6)have been identified although few studies have addressed their *** we aim to determine the function of the teleost specific sst4 in the zebrafish.A CRIsPR/Cas9 sst4 zebrafish mutant with loss of function(sst4−/−)was produced which grew significantly faster and was heavier at the onset of gonadal maturation than the wild type(Wt).Consistent with their faster growth,liver igf1,igf2a and igf2b expression was significantly upregulated in the sst4−/−fish compared to the *** examination of the ovaries and testis indicated that sst4−/−fish had slightly delayed testicular gametogenesis compared to the *** lower expression of igf3,amh,insl3,hsd17b3,hsd11b2,hsd20b,cyp11b and cyp17 was consistently observed in the sst4−/−*** contrast,the ovaries had lower expression of igf1,igf2a and cyp19a1a but increased expression of igf2b and *** gonadotrophin beta subunits(fshb and lhb)in the brain were downregulated indicating the brain-pituitary-gonadal axis was downregulated in the sst4−/−fish and suggesting that the steroid production is compromised in the maturing *** addition,analysis of sst1 and sst3 mRNA levels in sst4−/−fish suggests a dosage compensation effect of sst1 in the brain and ***,the results from the zebrafish sst4−/−line support the idea that sst4 is involved in the regulation of igf signalling,somatic growth and reproduction since steroidogenesis and gametogenesis at pubertal onset were compromised.

One of the goals of the aquaculture industry is to understand and control growth associated traits through selective *** the present study the molecular basis of growth heterogeneity in the European sea bass(Dicentrarchus labrax)was *** establish growth heterogeneity in a group of hatchery bred sea bass individuals were tagged and their specific growth rates(sGR)determined at monthly *** expression in the brain,liver and white muscle from fish with the most divergent sustained sGR(6 individuals of the first and last quartile)was assessed using supersAGE(serial Analysis Gene Expression)combined with next generation sOLiD4 sequencing.A total of approx.11 million edited tags(26 bp),on average 2 million tags per sAGE library,that represented 47.071 unique transcripts were *** of transcripts in fish with high and low sGR yielded 344,698 and 601 differently expressed tags(0.01%false discovery rate and 4-fold change)in brain,liver and muscle,*** tags were mapped onto the sea bass genome and approximately one third of the tags could be assigned to annotated *** enrichment analysis revealed in liver,muscle and brain intricate gene expression changes in endocrine regulatory pathways involved in growth,metabolic and the stress axis,underlying divergent sGR in sea bass.

Free vibration and transient response of double curved beams connected by intermediate straight beams

this paper investigates the free vibration and transient response of interconnected structures including double curved beams and intermediate straight beams,which are all constructed from functionally graded porous(FGP) materials. the strain potential and kinetic energies of each beam along with the work done by the external force are calculated. Additionally, a higher-order beam element is introduced to derive stiffness and mass matrices, along with the force vector. the curved and straight beams are discretized, and their assembled stiffness, mass matrices, and force vectors, are *** conditions at the joints are used to derive the total matrices of the entire structure. subsequently, the natural frequencies and transient response of the system are determined. the accuracy of the mathematical model and the self-developed computer program is validated through the comparison of the obtained results with those of the existing literature and commercial software ANsYs, demonstrating excellent ***, a comprehensive study is conducted to investigate the effects of various parameters on the free vibration and transient response of the considered structure.

利用CRIsPR/Cas9基因编辑技术在斑马鱼中构建了可稳定遗传的生长抑素(somatostatin,sst)基因(sst1)突变体鱼系。通过比较6 dpf(days post fertilization,dpf)的sst1^(-/-)突变体和野生型仔鱼转录组,发现sst1^(-/-)突变体相关生化过程和信号通路发生了显著变化。相比野生型,sst1^(-/-)突变体仔鱼中有354个基因显著上调,504个基因显著下调。GO(gene ontology)富集分析表明,大部分差异基因与氨基酸和蛋白质生物合成相关。KEGG(kyoto encyclopedia of genes and genomes)通路富集显示代谢通路、氨基酸合成、氨基酰基-tRNA合成、核糖体合成和rRNA加工相关通路表达增强。结果表明,sst1^(-/-)突变体蛋白质生物合成和代谢过程更加活跃。Rt-qPCR验证结果显示,随机选取的10个基因mRNA表达变化趋势与转录组测序结果一致,说明转录组测序结果真实可靠。6月龄突变体体质量、体长和野生型相比并无显著差异。本研究结果为进一步挖掘生长抑素基因家族的潜在功能奠定基础。

Photon shifting and trapping in perovskite solar cells for improved efficiency and stability

Advanced light management techniques can enhance the sunlight absorption of perovskite solar cells(PsCs).When located at the front,they may act as a UV barrier,which is paramount for protecting the perovskite layer against UVenabled *** it was recently shown that photonic structures such as Escher-like patterns could approach the theoretical Lambertian-limit of light trapping,it remains challenging to also implement UV protection properties for these diffractive structures while maintaining broadband absorption ***,we propose a checkerboard(CB)tile pattern with designated UV photon conversion *** a combined optical and electrical modeling approach,this photonic structure can increase photocurrent and power conversion efficiency in ultrathin PsCs by 25.9%and 28.2%,*** further introduce a luminescent down-shifting encapsulant that converts the UV irradiation into Visible photons matching the solar cell absorption *** this end,experimentally obtained absorption and emission profiles of state-of-the-art down-shifting materials(i.e.,lanthanidebased organic-inorganic hybrids)are used to predict potential gains from harnessing the UV *** demonstrate that at least 94%of the impinging UV radiation can be effectively converted into the Visible spectral *** protection from high-energy photons contributes to the market deployment of perovskite solar cell technology,and may become crucial for space applications under AM0 *** combining light trapping with luminescent downshifting layers,this work unravels a potential photonic solution to overcome UV degradation in PsCs while circumventing optical losses in ultrathin cells,thus improving both performance and stability.

Lower back pain is one of the most common medical problems in the world and it is experienced by a huge percentage of people *** to its ability to produce a detailed view of the soft tissues,including the spinal cord,nerves,intervertebral discs,and vertebrae,Magnetic Resonance Imaging is thought to be the most effective method for imaging the *** semantic segmentation of vertebrae plays a major role in the diagnostic process of lumbar *** is difficult to semantically partition the vertebrae in Magnetic Resonance Images from the surrounding variety of tissues,including muscles,ligaments,and intervertebral discs.U-Net is a powerful deep-learning architecture to handle the challenges of medical image analysis tasks and achieves high segmentation *** work proposes a modified U-Net architecture namely MU-Net,consisting of the Meijering convolutional layer that incorporates the Meijering filter to perform the semantic segmentation of lumbar vertebrae L1 to L5 and sacral vertebra ***-colour mask images were generated and used as ground truth for training the *** work has been carried out on 1312 images expanded from t1-weighted mid-sagittal MRI images of 515 patients in the Lumbar spine MRI Dataset publicly available from Mendeley *** proposed MU-Net model for the semantic segmentation of the lumbar vertebrae gives better performance with 98.79%of pixel accuracy(PA),98.66%of dice similarity coefficient(DsC),97.36%of Jaccard coefficient,and 92.55%mean Intersection over Union(mean IoU)metrics using the mentioned dataset.

structural Optimization of Metal and Polymer Ore Conveyor Belt Rollers

Ore conveyor belt rollers operate in harsh environments,making them prone to premature *** service lives are highly dependent on the stress field and bearing misalignment angle,for which limit values are defined in a *** this work,an optimization methodology using metamodels based on radial basis functions is implemented to reduce themass of twomodels of *** a structural point of view,one of the rollers ismade completely of metal,while the other also has some components made of polymeric *** objective of this study is to develop and apply a parametric structural optimization methodology to minimize the mass of the two models of *** represent the mechanical behavior of the rollers,simulations were performed using the finite element *** the numerical optimization process,the variable parameters were the dimensions of the shaft and external *** geometric configuration that corresponded at the same time to the lowest mass and acceptable ranges for the stress and bearingmisalignment angle was *** the proposed methodology,a 32.3% reduction in mass was obtained for a metal roller design and an 18.9% reduction for a polymer *** both cases,the constraints were not *** the all-metal roller,the safety factors for the maximum stress and bearingmisalignment angle were 1.44 and 1.75,respectively,while for the polymer roller the corresponding figures were 1.50 and *** work describes a low-computational-cost optimization methodology for roller designs that have been little studied in the ***,the methodology could be adapted for use with other types of rollers and rollers made of different materials.
