A comprehensive evaluation of flowering plant diversity and conservation priority for national park planning in China

Establishment of a national park protection system in China,including the latest target proposed to protect at least 30%of the land area,calls for a comprehensive exploration of conservation priorities incorporating multiple diversity *** herein evaluate the spatial distribution of Chinese flowering plants from the perspectives of richness,uniqueness,vulnerability,and evolutionary history,by integrating three mega-phylogenies and comprehensive distribution *** detect significantly high consistency among hotspots of different diversity measures for Chinese flowering plants,suggesting that multiple facets of evolutionary diversity are concentrically distributed in *** legal protection to these areas is expected to maximize positive conservation *** propose two integrative diversity indices by incorporating three richness-based and three phylogeny-based measures,*** methods identify areas with high species richness,but the integrative phylogeny-based index also locates key areas with ancient and unique evolutionary histories(e.g.,Ailao-Wuliang Mts,Dabie Mts,Hainan rainforest,Karst area of Yunnan-Guizhou-Guangxi,Nanling Mts,and southeast coastal regions).Of all the diversity indices explored,phylogenetic endemism maximizes the incidental protection of other indices in most cases,emphasizing its significance for conservation ***,42 priority areas are identified by combining the 5%-criterion hotspots of two integrative indices and the minimum area to protect all threatened species *** priorities cover only 13.3%of China's land area,but host 97.1%of species richness(23,394/24,095),96.5%of endemic species(11,841/12,274),100%of threatened species(2,613/2,613),and 99.3%of phylogenetic diversity for flowering plants involved in this *** frameworks provide a solid scientific basis for national park planning in China.

RNA secondary structure has become the most exploitable feature for ab initio detection of non-coding RNA（nc RNA） genes from genome sequences. Previous work has used Minimum Free Energy（MFE） based methods developed to identify nc RNAs by measuring sequence fold stability and certainty. However, these methods yielded variable performances across different nc RNA species. Designing novel reliable structural measures will help to develop effective nc RNA gene finding tools. This paper introduces a new RNA structural measure based on a novel RNA secondary structure ensemble constrained by characteristics of native RNA tertiary structures. The new method makes it possible to achieve a performance leap from the previous structure-based methods. Test results on standard nc RNA datasets（benchmarks） demonstrate that this method can effectively separate most nc RNAs families from genome backgrounds.

Measuring NO Production by Plant Tissues and Suspension Cultured Cells

We describe an inexpensive and reliable detector for measuring NO emitted in the gas phase from plants. The method relies on the use of a strong oxidizer to convert NO to NO2 and subsequent capture of NO2 by a Griess reagent trap. The set-up approaches the sensitivity for NO comparable to that of instruments based on chemiluminescence and photoacoustic detectors. We demonstrate the utility of our set-up by measuring NO produced by a variety of well established plant sources. NO produced by nitrate reductase （NR） in tobacco leaves and barley aleurone was readily detected, as was the production of NO from nitrite by the incubation medium of barley aleurone. Arabidopsis mutants that overproduce NO or lack NO-synthase （AtNOS1） also displayed the expected NO synthesis phenotype when assayed by our set-up. We could also measure NO production from elicitor-treated suspension cultured cells using this set-up. Further, we have focused on the detection of NO by a widely used fluorescent probe 4-amino-5-methylamino-2＇,7＇-difluorofluorescein （DAF-FM）. Our work points to the pitfalls that must be avoided when using DAF-FM to detect the production of NO by plant tissues. In addition to the dramatic effects that pH can have on fluorescence from DAF-FM, the widely used NO scavengers 2-phenyl- 4,4,5, 5-tetramethylimidazoline- 1-oxyl-3-oxide （PTIO） oxide （cPTIO） can produce anomalous and unexpected to bind to dead cells and remain NO-sensitive. and 2-（4-carboxyphenyl）-4,4,5,5-tetramethylimidazoline- 1-oxyl-3- results. Perhaps the most serious drawback of DAF-FM is its ability to bind to dead cells and remain NO-sensitive.

The fundamental unit of rapid, physiological color change in vertebrates is the dermal chromato- phore unit. This unit, comprised of cellular associations between different chromatophore types, is relatively conserved across the fish, amphibian, and reptilian species capable of physiological color change and numerous attempts have been made to understand the nature of the four major chro- matophore types （melanophores, erythrophores, xanthophores, and iridophores） and their bio- chemical regulation. In this review, we attempt to describe the current state of knowledge regard- ing what classifies a pigment cell as a dynamic chromatophore, the unique characteristics of each chromatophore type, and how different hormones, neurotransmitters, or other signals direct pig- ment reorganization in a variety of vertebrate taxa.
