Synthetic host-guest pairs as novel bioorthogonal tools for pre-targeting

Due to its simplicity, high efficiency, and chemo-selectivity, bioorthogonal chemistry has shown a great application potential in pre-targeting.Currently, four bioorthogonal pairs as targeting tools, including (strept)avidin/biotin, antibody/antigen, oligonucleotide hybridization and IEDDA tools, have been developed and applied in targeted delivery.Nevertheless, all of these tools still suffer from some limitations, such as difficult modification, biochemical fragility and larger molecular weight for biological association tools, as well as chemical instability for IEDDA tools.Synthetic host-guest pairs with relatively small molecular sizes not only possess strong chemical stability, but also have the features of fast conjugation rate, tunable binding affinity , easy modification, and high chemo-selectivity.Consequently, they can be used as a novel non-covalent bioorthogonal tool for pre-targeting.In order to further promote the development of host-guest pairs as novel bioorthogonal tools for pre-targeted delivery, we firstly calculate their conversion rate to make researcher aware of their unique advantages;next, we summarize the recent research progress in this area.The future perspectives and limitations of these unique tools will be discussed.This review will provide a systemic overview of the development of synthetic host-guest pairs as novel bioorthogonal tools for pre-targeting, and may serve as a “go for” resort for researchers who are interested in searching for new synthetic tools to improve pre-targeting.

Supramolecular systems feature dynamic,reversible and stimuli-responsive characteristics,which are not easily achieved by molecular entities.The last decade has witnessed tremendous advances in the investigations of supramolecular systems for various bioapplications,which include drug delivery,anticancer therapy,antibacterial therapy,photodynamic therapy,photothermal therapy,combination therapy,antidotes for residual drugs or toxins,and bioimaging and biosensing.Host-guest chemistry has played a key role in the development of such bioactive supramolecular systems,and natural macrocycles(such as cyclodextrins),synthetic macrocycles(such as calixarenes,cucurbit[n]urils,and pillararenes),and porous framework polymers(such as supramolecular organic frameworks and flexible organic frameworks)have been most successfully used as hosts to build different kinds of host-guest systems for attaining designed biofunctions.The self-assembly of rationally designed amphiphilic molecules,macromolecules and polymers represent another important approach for the construction of supramolecular architectures with advanced biofunctions.In this review,we summarize the important contributions made by Chinese researchers in this field,with emphasis on those reported in the past five years.

A polyphenol-assisted IL-10 mRNA delivery system for ulcerative colitis

With the development of synthesis technology, modified messenger RNA(mRNA) has emerged as a novel category of therapeutic agents for a broad of diseases. However, effective intracellular delivery of mRNA remains challenging, especially for its sensitivity to enzymatic degradation. Here, we propose a polyphenol-assisted handy delivery strategy for efficient in vivo delivery of IL-10 mRNA.IL-10 mRNA binds to polyphenol ellagic acid through supramolecular binding to yield a negatively charged core, followed by complexing with linear polyetherimide and coating with bilirubin-modified hyaluronic acid to obtain a layer-by-layer nanostructure. The nanostructure specifically up-regulated the level of IL-10, effectively inhibited the expression of inflammatory factors, promoted mucosal repair, protected colonic epithelial cells against apoptosis, and exerted potent therapeutic efficacy in dextran sulfate sodium salt-induced acute and chronic murine models of colitis. The designed delivery system without systemic toxicity has the potential to facilitate the development of a promising platform for mRNA delivery in ulcerative colitis treatment.

In spite of being a promising therapeutic modality,gene therapy has limited clinical applications,mostly due to the lack of spatiotemporal resolution and inadequate efficacy.Herein,we present a facile strategy to remotely control intracellular gene expression by using gold nanorod-(Au NR)derived,host-guest interaction-mediated supramolecular vesicles as a gene carrier and photothermal transducer.Upon pulsed laser irradiation,mild photothermal conditions dissociate supramolecular vesicles to release gene and simultaneously activate heat shock protein-70 promoter(Hsp70)for spatiotemporally initiating gene expression inside cancer cells.Subsequently,upon introducing a polymeric guest species specifically into cancer cells,the dissociated Au NRs functionalized with macrocyclic host molecules could re-aggregate rapidly in cells to retard exocytosis of these NRs,thereby allowing deferred photothermal therapy to enhance the overall therapeutic outcome.

Bioconjugation methods offer very important tools in studying biological systems.Synthetic host-guest pairs provide an alternative and complementary conjugation method to bioorthogonal reactions and biological association pairs.Nevertheless,macrocyclic hosts that can be used for in situ capture are limited and often rely on extremely high binding affinities.Herein,we report an alternative bioorthogonal host-guest pair that relies on highly selective molecular recognition in water.The host,namely amide naphthotube,possesses a biomimetic cavity with inward-directing hydrogen bonding sites and shows selective and strong binding to the guest(2-phenyl pyrimidine)even in biological media.Through anchoring the tetraphenyl ethylene-modi fied hosts to cell surfaces,the bioorthogonal host-guest pair can be applied in cell surface recognition,cell-cell interactions,and tissue imaging in mice.The bioorthogonality is originated from the high binding selectivity of the biomimetic macrocyclic host,which is different from other known host-guest pairs that have been applied in biological systems.This research provides a new noncovalent bioconjugation tool and a new concept for designing bioorthogonal host-guest pairs for biological applications.

Cucurbit[7]uril as a Broad-Spectrum Antiviral Agent against Diverse RNA Viruses

The emergence and re-emergence of RNA virus outbreaks highlight the urgent need for the development of broadspectrum antivirals.Polyamines are positively-charged small molecules required for the infectivity of a wide range of RNA viruses,therefore may become good antiviral targets.Cucurbit[7]uril(CB[7]),a synthetic macrocyclic molecule,which can bind with amine-based organic compounds with high affinity,has been shown to regulate bioactive molecules through competitive binding.In this study,we tested the antiviral activity of CB[7]against diverse RNA viruses,including a panel of enteroviruses(i.***.human enterovirus A71,coxsackievirus A16,coxsackievirus B3,and echovirus 11),some flaviviruses(i.***.dengue virus and Zika virus),and an alphavirus representative Semliki forest virus.CB[7]can inhibit virus replications in a variety of cell lines,and its mechanism of action is through the competitive binding with polyamines.Our findings not only for the first time provide evidence that CB[7]can be a promising broad-spectrum antiviral agent,but more importantly,offer a novel therapeutic strategy to fight against RNA viruses by supramolecular sequestration of polyamines.

pH-Responsive supramolecular DOX-dimer based on cucurbit[8]uril for selective drug release

A supramolecular dimer of doxorubicin(DOX)was constructed via ternary host-guest interactions between cucurbit[8]uril(CB[8])and tryptophan modified DOX(DOX-Trp,connected with an acid-labile bond)and we demonstrate for the first time that a supramolecular dimer of DOX can be formed upon homo-dimerization by CB[8],which may act as a stimuli pH-responsive,supramolecular DOX dimer prodrug system.This supramolecular DOX dimer transported DOX efficiently and selectively to cancer cells,thereby exhibiting significantly minimized cytotoxicity against noncancerous cells while maintaining effective cytotoxicity against cancer cells.Under this strategy,many other anticancer drugs could be chemically modified and loaded as a dimeric"ammunition"into CB[8]as supramolecular dimer prodrug systems(or a"jet fighter")for improved cancer therapy.

The ongoing outbreak of Coronavirus Disease 2019(COVID-19)has become a global public health emergency.SARScoronavirus-2(SARS-CoV-2),the causative pathogen of COVID-19,is a positive-sense single-stranded RNA virus belonging to the family Coronaviridae.For RNA viruses,virus-encoded RNA helicases have long been recognized to play pivotal roles during viral life cycles by facilitating the correct folding and replication of viral RNAs.Here,our studies show that SARS-CoV-2-encoded nonstructural protein 13(nsp13)possesses the nucleoside triphosphate hydrolase(NTPase)and RNA helicase activities that can hydrolyze all types of NTPs and unwind RNA helices dependently of the presence of NTP,and further characterize the biochemical characteristics of these two enzymatic activities associated with SARS-CoV-2 nsp13.Moreover,we found that some bismuth salts could effectively inhibit both the NTPase and RNA helicase activities of SARS-CoV-2 nsp13 in a dose-dependent manner.Thus,our findings demonstrate the NTPase and helicase activities of SARS-CoV-2 nsp13,which may play an important role in SARS-CoV-2 replication and serve as a target for antivirals.

同一个道德两难情境,使用不同语言(母语、外语)呈现时,会影响个体的道德选择倾向,外语条件中个体会做出更多的功利主义选择,即道德外语效应。使用不同方言呈现道德两难困境是否会影响个体的道德选择倾向?实验1和实验2都采用单因素被试内设计,在无音频(实验1)、普通话、本地方言和非本地方言条件下呈现道德两难困境。两个实验的被试分别为中国的大学生和非大学生成年群体,并分别在实验室和线上情境下完成对道德两难困境的决策,发现本地方言相比于非本地方言(实验1)和普通话(实验2)引发更少的功利主义道德选择。本研究首次发现类似道德外语效应的道德方言效应。
