A novel prognostic target-gene signature and nomogram based on an integrated bioinformatics analysis in hepatocellular carcinoma

There is currently no effective solution to the problem of poor prognosis and recurrence of HCC.The technology of immunotherapy and prognosis of genetic material has made continuous progress in recent years.In the study,a 5-gene signature was established for the prognosis of HCC through biological information,and the immune infiltration of HCC patients was studied.After studied HCC patients’immune infiltration,the paper screened the differential target genes of miR-126-3p in HCC downloaded from TCGA database,and uses WGCNA method to select the modular genes highly relevant to M2 macrophage.Then we use LASSO and COX regression analysis technology to establish the 5-gene signature.The nomogram is established by combining the prognostic score and clinical phenotype.Cibersort was empolyed to observe the immune infiltration in HCC patients.We revealed the biological pathways of HCC-related genes through GSEA and Metascape.The bioinformatics analysis of 2495 differential target genes finally constructed a 5-gene signature with a reliable prognostic ability(CDCA8,SLC41A3,PPM1G,TCOF1,GRPEL2).The combination of prognostic score and AJCC_Stage resulted in a more reliable prognosis ability.At the same time,10 immune cells that are differentially expressed in HCC patients were also found.8 GSEA pathways related to the prognosis were found.In the study,a reliable 5-gene signature was established based on the differential target gene of miR-126-3p to study the immune infiltration in HCC patients.It provides help for HCC-related prognosis research and immunotherapy.

Investigation of the mechanical and ballistic properties of hybrid carbon/aramid woven laminates

High-performance ballistic fibers,such as aramid fiber and ultra-high-molecular-weight polyethylene(UHMWPE),are commonly used in anti-ballistic structures due to their low density,high tensile strength and high specific modulus.However,their low modulus in the thickness direction and insufficient shear strength limits their application in certain ballistic structure.In contrast,carbon fiber reinforced epoxy resin matrix composites(CFRP)have the characteristics of high modulus in the thickness direction and high shear resistance.However,carbon fibers are rarely used and applied for protection purposes.A hybridization with aramid fiber reinforced epoxy resin matrix composites(AFRP)and CFRP has the potential to improve the stiffness and the ballistic property of the typical ballistic fiber composites.The hybrid effects on the flexural property and ballistic performance of the hybrid CFRP/AFRP laminates were investigated.Through conducting mechanical property tests and ballistic tests,two sets of reliable simulation parameters for AFRP and CFRP were established using LS-DYNA software,respectively.The experimental results suggested that by increasing the content of CFRP that the flexural properties of hybrid CFRP/AFRP laminates were enhanced.The ballistic tests'results and the simulation illustrated that the specific energy absorption by the perforation method of CFRP achieved 77.7%of AFRP.When CFRP was on the striking face,the shear resistance of the laminates and the resistance force to the projectiles was promoted at the initial penetration stage.The proportion of fiber tensile failures in the AFRP layers was also enhanced with the addition of CFRP during the penetration process.These improvements resulted in the ballistic performance of hybrid CFRP/AFRP laminates was better than AFRP when the CFRP content was 20 wt%and 30 wt%.

Bone tissue engineering (BTE) is a rapidly developing strategy for repairing critical-sized bone defects to address the unmet need for bone augmentation and skeletal repair. Effective therapies for bone regeneration primarily require the coordinated combination of innovative scaffolds, seed cells, and biological factors. However, current techniques in bone tissue engineering have not yet reached valid translation into clinical applications because of several limitations, such as weaker osteogenic differentiation, inadequate vascularization of scaffolds, and inefficient growth factor delivery. Therefore, further standardized protocols and innovative measures are required to overcome these shortcomings and facilitate the clinical application of these techniques to enhance bone regeneration. Given the deficiency of comprehensive studies in the development in BTE, our review systematically introduces the new types of biomimetic and bifunctional scaffolds. We describe the cell sources, biology of seed cells, growth factors, vascular development, and the interactions of relevant molecules. Furthermore, we discuss the challenges and perspectives that may propel the direction of future clinical delivery in bone regeneration.

Unraveling the Effects of Austenitizing Temperature and Austenite Grain Size on the Crystallographic Characteristics and Mechanical Properties of Martensitic Transformation Products in a Low‑Alloy Steel

The influence of austenitizing temperature and austenite grain size on the crystallographic characteristics and mechanical properties of transformation products was investigated in a low-alloy steel.Annealing at a lower temperature after austenitization at 1050℃ can reduce the vacancy concentration and enhance the stability of austenite,thereby determining the martensite-start(Ms)temperature and density of twin-related V1/V2 variant pairs.V1/V2 variant pairs are predominately generated by autocatalytic nucleation,which can promote transformation by self-accommodation.Annealing at 800℃ after austenitization at 1050℃ generated the highest content of V1/V2 variant pairs,which contributed to high values of strength,Charpy impact energy,and elongation.In addition to refining the austenite grain size,niobium(Nb)microalloying increases the packet boundaries of the transformation products,mainly because the refined austenite grains provide more nucleation sites for the martensitic transformation.However,the density of block boundaries decreases after austenite refinement by Nb microalloying,owing to insufficient autocatalytic nucleation.

Objective:Cytogenetic abnormalities have been proven to be the most valuable parameter for risk stratification of childhood acute lymphoblastic leukemia(ALL).However,studies on the prevalence of cytogenetic abnormalities and their correlation to clinical features in chinese pediatric patients are limited,especially large-scale studies.Methods:We collected the cytogenetics and clinical data of 1541 children newly diagnosed with ALL between 2001 and 2014 in four chinese hospitals,and retrospectively analyzed their clinical features,prognosis and risk factors associated with pediatric ALL.

Primary orbital monophasic synovial sarcoma with calcification:A case report

BACKGROUND Synovial sarcoma is a malignant mesenchymal neoplasm with variable epithelial differentiation.Most synovial sarcoma cases are reported in young adults and can arise in any body site.Notably,primary orbital synovial sarcoma is rare.CASE SUMMARY An 8-year-old east Asian girl with 1-month history of gradual painless proptosis and lacrimation of the right eye was admitted.The patient presented with painless proptosis,downward eyeball displacement,and upward movement disorders.According to clinical manifestations,imaging examinations and postoperative immunohistochemical examinations,the diagnosis was monophasic synovial sarcoma with calcification.The patient underwent anterior orbitotomy procedure for removal of the right orbital mass under general anesthesia.The diagnosis of monophasic synovial sarcoma with calcification was confirmed finally through histological and immunohistochemical exam.The follow-up period was 6 mo,and no recurrence was observed during this period.CONCLUSION Primary orbital monophasic synovial sarcoma with calcification is a rare sarcoma,and clinical manifestations and imaging results are not specific.The tumor may present similar features as a benign tumor.Comprehensive analysis of clinical,radiological,and pathological findings is critically important for making the right diagnosis.Conventional treatment approach for synovial sarcoma is surgical resection with adjuvant or neoadjuvant radiotherapy,which is highly effective for localized tumors.

Association of emergency room visits for respiratory diseases with sources of ambient PM2.5

Previous studies have reported associations of short-term exposure to different sources of ambient fine particulate matter(PM2.5)and increased mortality or hospitalizations for respiratory diseases.Few studies,however,have focused on the short-term effects of source-specific PM2.5 on emergency room visits(ERVs)of respiratory diseases.Source apportionment for PM2.5 was performed with Positive Matrix Factorization(PMF)and generalized additive model was applied to estimate associations between source-specific PM2.5 and respiratory disease ERVs.The association of PM2.5 and total respiratory ERVs was found on lag4(RR=1.011,95%CI:1.002,1.020)per interquartile range(76μg/m3)increase.We found PM2.5 to be significantly associated with asthma,bronchitis and chronic obstructive pulmonary disease(COPD)ERVs,with the strongest effects on lag5(RR=1.072,95%CI:1.024,1.119),lag4(RR=1.104,95%CI:1.032,1.176)and lag3(RR=1.091,95%CI:1.047,1.135),respectively.The estimated effects of PM2.5 changed little after adjusting for different air pollutants.Six primary PM2.5 sources were identified using PMF analysis,including dust/soil(6.7%),industry emission(4.5%),secondary aerosols(30.3%),metal processing(3.2%),coal combustion(37.5%)and traffic-related source(17.8%).Some of the sources were identified to have effects on ERVs of total respiratory diseases(dust/soil,secondary aerosols,metal processing,coal combustion and traffic-related source),bronchitis ERVs(dust/soil)and COPD ERVs(traffic-related source,industry emission and secondary aerosols).Different sources of PM2.5 contribute to increased risk of respiratory ERVs to different extents,which may provide potential implications for the decision making of air quality related policies,rational emission control and public health welfare.

Background:Visceral adipose tissue-derived serine protease inhibitor(vaspin),a secretory adipokine,protects against insulin resistance.Recent studies have demonstrated that serum vaspin levels are decreased in patients with coronary artery disease and that vaspin protects against myocardial ischemia-reperfusion injury and atherosclerosis.However,it remains unclear whether vaspin exerts specific effects on pathological cardiac hypertrophy.Methods:An in vivo study was conducted using a cardiac hypertrophy model established by subcutaneous injection of isoproterenol(ISO)in C57BL/6 and vaspin-ko mice.Rapamycin was administered intraperitoneally to mice,for further study.H9c2 cells and neonatal rat ventricular myocytes(NRVMs)were treated with ISO to induce hypertrophy.Human vaspin fusion protein,the proteasome inhibitor MG132,and chloroquine diphosphate were used for further mechanistic studies.Results:Here,we provide the first evidence that vaspin knockdown results in markedly exaggerated cardiac hypertrophy,fibrosis,and cardiomyocyte senescence in mice treated with ISO.Conversely,the administration of exogenous recombinant human vaspin protected NRVMs in vitro against ISO-induced hypertrophy and senescence.Furthermore,vaspin significantly potentiated the ISO-induced decrease in autophagy.Both rapamycin and chloroquine diphosphate regulated autophagy in vivo and in vitro,respectively,and participated in vaspin-mediated cardioprotection.Moreover,the PI3K-AKT-mTOR pathway plays a critical role in vaspin-mediated autophagy in cardiac tissues and NRVMs.Our data showed that vaspin downregulated the p85 and p110 subunits of PI3K by linking p85 and p110 to NEDD4L-mediated ubiquitination degradation.Conclusion:Our results show,for the first time,that vaspin functions as a critical regulator that alleviates pathological cardiac hypertrophy by regulating autophagy-dependent myocardial senescence,providing potential preventive and therapeutic targets for pathological cardiac hypertrophy.

Synthesis and Properties of High Performance Functional Polyimides Containing Rigid Nonplanar Conjugated Fluorene Moieties

A diamine(wuFDA) containing vertical rigid non-planar conjugated fluorene moiety and low polarizability group(C―F)was designed and synthesized through three steps of reactions(halogenated reaction, Suzuki coupling reaction, and reduction reaction).Four kinds of high performance functional polyimides(wuFPI-6 F, wuFPI-BP, wuFPI-BT, and wuFPI-PM) were thus prepared by the condensation polymerization of wuFDA with four commercial dianhydride 6 FDA, BPDA, BTDA, and PMDA, respectively. The polyimides exhibited low dielectric constant, excellent thermal stability, outstanding solubility, good film-forming property, and mechanical properties. The dielectric constants of the polyimides were in the range of 2.28-2.88(f = 10~4 Hz). The 5% weight-loss temperatures(Td 5%)in nitrogen were in the range of 555-584 °C, and the glass transition temperatures(T_g) were in the range of 408-448 °C. The weight loss of wuFPI-BP maintaining at 450 and 500 °C for half an hour was only 0.33% and 1.26%, respectively. All the wuFPIs could be dissolved in almost all organic solvents, even chloroform. The tensile strength and tensile modulus of these films were in the ranges of 78.6-85.7 MPa and 3.1-3.2 GPa, respectively. In addition, the polyimides displayed light color with special fluorescent and resistive switching(ON-OFF) characteristics; the maximum fluorescence emission was observed at 422-424 nm in NMP solution and at 470-548 nm in film state. The memory devices with the configuration of indium tin oxide/wuFPIs/aluminum(ITO/wuFPIs/Al) exhibited distinct volatile memory characteristics of static random access memory(SRAM), with an ON/OFF current ratio of 10~5-10~6. These functional polyimides showed attractive potential applications in the field of high performance flexible polymer photoelectronic devices or polymer memory devices.

Study of neural network disturbance learning and application in RoboCup

To solve the problem of convergence to a local optimum in the multi-layer feedforward neural network, a new disturbance gradient algorithm is proposed. Through introducing random disturbance into the training process, the algorithm can avoid being trapped into the local optimum. The random disturbance obeys the Boltzmann distribution. The convergence of the algorithm to the global optimum is statistically guaranteed. The application of the algorithm in RoboCup, which is a complex multi-agent system, is discussed. Experiment results illustrate the learning efficiency and generalization ability of the proposed algorithm.
