Endothelial glycocalyx as a potential theriapeutic target in organ injuries

Objective:The endothelial glycocalyx(eGC)is a dynamic and multicomponent layer of macromolecules found at the surface of vascular endothelium,which is largely underappreciated.It has recently been recognized that eGC is a major regulator of endothelial function and may have therapeutic value in organ injuries.This study aimed to explore the role of the eGC in various pathologic and physiologic conditions,by reviewing the basic research findings pertaining to the detection of the eGC and its clinical significance.We also explored different pharmacologic agents used to protect and rebuild the eGC.Data sources:An in-depth search was performed in the PubMed database,focusing on research published after 2003 with keywords including eGC,permeability,glycocalyx and injuries,and glycocalyx protection.Study selection:Several authoritative reviews and original studies were identified and reviewed to summarize the characteristics of the eGC under physiologic and pathologic conditions as well as the detection and protection of the eGC.Results:The eGC degradation is closely associated with pathophysiologic changes such as vascular permeability,edema formation,mechanotransduction,and clotting cascade,together with neutrophil and platelet adhesion in diverse injury and disease states including inflammation(sepsis and trauma),ischemia-reperfusion injury,shock,hypervolemia,hypertension,hyperglycemia,and high na+as well as diabetes and atherosclerosis.Therapeutic strategies for protecting and rebuilding the eGC should be explored through experimental test and clinical verifications.Conclusions:Disturbance of the eGC usually occurs at early stages of various clinical pathophysiologies which can be partly prevented and reversed by protecting and restoring the eGC.The eGC seems to be a promising diagnostic biomarker and therapeutic target in clinical settings.

PHOSPHO1 Serves as a Key Metabolism-Related Biomarker in the Tumorigenesis of Diffuse Large B-cell Lymphoma

Objective:Diffuse large B-cell lymphoma(DLBCL)is an aggressive type of non-Hodgkin lymphoma.Due to its genetic heterogeneity and abnormal metabolism,many DLBCL patients have a poor prognosis.This study investigated the key metabolism-related genes and potential mechanisms.Methods:Differentially expressed genes,differentially expressed transcription factors(TFs),and differentially expressed metabolism-related genes(DEMRGs)of glucose and lipid metabolic processes were identified using the edgeR package.Key DEMRGs were screened by Lasso regression,and a prediction model was constructed.The cell type identification by estimating relative subsets of Rna transcripts algorithm was utilized to assess the fraction of immune cells,and Gene Set Enrichment Analysis was used to determine immune-related pathways.A regulatory network was constructed with significant co-expression interactions among TFs,DEMRGs,immune cells/pathways,and hallmark pathways.Results:A total of 1551 DEMRGs were identified.A prognostic model with a high applicability(area under the curve=0.921)was constructed with 13 DEMRGs.Tumorigenesis of DLBCL was highly related to the neutrophil count.Four DEMRGs(PRXL2AB,CCN1,DECR2 and PHOSPHO1)with 32 TF-DEMRG,36 DEMRG-pathway,14 DEMRG-immune-cell,9 DEMRG-immune-gene-set,and 67 DEMRG-protein-chip interactions were used to construct the regulatory network.Conclusion:We provided a prognostic prediction model based on 13 DEMRGs for DLBCL.We found that phosphatase,orphan 1(PHOSPHO1)is positively regulated by regulatory factor X5(RFX5)and mediates MYC proto-oncogene(MYC)targeting the V2 pathway and neutrophils.

Microstructure and properties of an Al–Ti–Cu–Si brazing alloy for SiC–metal joining

An Al–Ti–Cu–Si solid–liquid dual-phase alloy that exhibits good wettability and appropriate interfacial reaction with SiC at 500–600°C was designed for SiC–metal joining. The microstructure, phases, differential thermal curves, and high-temperature wetting behavior of the alloy were analyzed using scanning electron microscopy, X-ray diffraction analysis, differential scanning calorimetry, and the sessile drop method. The experimental results show that the 76.5Al–8.5Ti–5Cu–10Si alloy is mainly composed of Al–Al2Cu and Al–Si hypoeutectic low-melting-point microstructures (493–586°C) and the high-melting-point intermetallic compound AlTiSi (840°C). The contact angle, determined by high-temperature wetting experiments, is approximately 54°. Furthermore, the wetting interface is smooth and contains no obvious defects. Metallurgical bonding at the interface is attributable to the reaction between Al and Si in the alloy and ceramic, respectively. The formation of the brittle Al4C3phase at the interface is suppressed by the addition of 10wt% Si to the alloy. ? 2017, University of Science and Technology Beijing and Springer-Verlag Berlin Heidelberg.

Plasma spray of biofunctional(Mg, Sr)-substituted hydroxyapatite coatings for titanium alloy implants

Plasma-sprayed hydroxyapatite (HA) coatings have been widely utilized in load-bearing titanium alloy implants. In this study, Mg, Sr co-substituted HA ((Mg,Sr)-HA) nano-scale powders have been synthesized, which are further used to prepare (Mg,Sr)-HA coatings on Ti-6A1-4V alloys in order to improve the biological functions. The average size of (Mg,Sr)-HA nano particles is ~75nm. The average bonding strength for (Mg,Sr)-HA coating and samples after heat treatment at 500℃ or 600℃ for 3h are 26.17±2.11 MPa, 36.07±4.48 MPa and 37.07 ±2.95 MPa, respectively. There is a significantly increase of bonding strength likely due to low residual stress after heated treatment.MC3T3-E1 cells show a high proliferation rate when cultured with (Mg,Sr)-HA coating extract compared to the normal culture medium, which also exhibit large extension and depositi on of extracellular matrices when adhered on the coating surfaces. Thus, these (Mg,Sr)-HA coatings show high bonding strength and improved biological functions, which offer promising future applications in the fields of orthopedics and dentistry.

Sky-blue perovskite light-emitting diodes based on quasi-two-dimensional layered perovskites

A mixed organic（4-phenylbutylamine, 4-PBA） and inorganic（cesium, Cs） cations are used to deposit quasi-two-dimensional layered perovskites. This layered perovskites exhibit good film coverage as twodimensional perovskites and high emission performance close to three-dimensional organic–inorganic hybrid perovskites. Light-emitting diodes（LEDs） are fabricated by using solution process based on the quasi-two-dimensional layered perovskites. The perovskite LEDs exhibit a sky-blue emission with electroluminescence peak at 491 nm and a low turn on voltage at 2.9 V. The maximum external quantum efficiency reaches 0.015% at brightness of 186 cd/m^2.

Transfer characteristics of nitrogen fixed by leguminous green manure crops when intercropped with maize in northwestern China

To ascertain the possibility of cultivating maize using biological nitrogen fixation(BNF)by leguminous green manure crops in maize/leguminous green manure intercropping systems,BNF and nitrogen(N)transfer were studied in Xining and Wuwei,two typical northwestern Chinese cities.The experimental treatments included monocultured maize,monocultured green manures(hairy vetch and common vetch),and their intercropping systems.The proportions of N derived from the atmosphere(%N_(dfa))in intercropping systems were not significantly different from that in monocultured green manure systems at either experimental site,except for that in hairy vetch(HV)in Xining.The amount of N derived from the atmosphere(N_(dfa))of common vetch(CV)significantly decreased from 1.16 and 1.10 g/pot in monoculture to 0.77 and 0.55 g/pot when intercropped with maize,in Xining and Wuwei,respectively,and the N_(dfa) of HV when intercropped significantly decreased from 1.02 to 0.48 g/pot in Xining.In the intercropping systems in Xining and Wuwei,the amounts of N transferred(N_(transfer))from CV to maize were 21.54 and 26.81 mg/pot,accounting for 32.9 and 5.9%respectively of the N accumulation in maize,and the values of N_(transfer) from HV to maize were 39.61 and 46.22 mg/pot,accounting for 37.0 and 23.3%,respectively,of the N accumulation in maize.Path analysis showed that soil nutrient and green manure biomass were mainly related to N_(dfa),and thatδ^(15) N had a primary relationship with N_(transfer).We found that 5.9-37.0%of N accumulation in maize was transferred from green manures,and that the N transfer ability to maize of HV was higher than that of CV.In conclusion,intercropping with leguminous green manures provided a feasible way for maize to effectively utilize biologicallyfixed N.

Complementary to mainstream cardiac imaging modalities for preclinical research,photoacoustic computed tomography(PACT)can provide functional optical contrast with high imaging speed and resolution.However,PACT has not been demonstrated to reveal the dynamics of whole cardiac anatomy or vascular system without surgical procedure(thoracotomy)for tissue penetration.Here,we achieved non-invasive imaging of rat hearts using the recently developed three-dimensional PACT(3D-PACT)platform,demonstrating the regulated illumination and detection schemes to reduce the effects of optical attenuation and acoustic distortion through the chest wall;thereby,enabling unimpeded visualization of the cardiac anatomy and intracardiac hemodynamics following rapidly scanning the heart within 10 s.We further applied 3D-PACT to reveal distinct cardiac structural and functional changes among the healthy,hypertensive,and obese rats,with optical contrast to uncover differences in cardiac chamber size,wall thickness,and hemodynamics.Accordingly,3D-PACT provides high imaging speed and nonionizing penetration to capture the whole heart for diagnosing the animal models,holding promises for clinical translation to cardiac imaging of human neonates.

Ticks are recognized as one of the most important vectors that cause diseases in animals and humans,and have significant public health impacts worldwide[1].They have a wide range of hosts among terrestrial vertebrates,including mammals,birds,amphibians.The numerous zoonotic pathogens transmitted by ticks are the causative agents of Lyme disease,rickettsiosis,ehrlichioses,relapsing fever,Q fever,and tularemia.Cases of tick-borne diseases are increasing around the world and some emerging or re-emerging pathogens are being discovered,which pose great challenges for prevention and control.

Flumazenil-insensitive benzodiazepine effects in vitro and in vivo

OBJECTIVE To identify benzodi⁃azepine(BZD)effects that are insensitive to the classical BZD binding site antagonist,flumazenil.Whether the flumazenil-insensitive BZD effects have selectivity on different GABAA receptor sub⁃types was also investigated.METHODS The high-concentration effects of BZDs and their sensitivity to flumazenil were determined on recombi⁃nant synaptic(α1β2γ2,α2β2γ2,α5β2γ2)and extra-synaptic(α4β2δ)GABAA receptors using the voltage-clamp electrophysiology technique.The in vivo evaluation of flumazenil-insensitive BZD effects was conducted in mice loss of right reflex(LORR)test.RESULTS Diazepam induced a biphasic potentiation for theα1β2γ2,α2β2γ2 andα5β2γ2 receptor channels,but did not affect theα4β2δreceptor.In contrast to the nanomolar com⁃ponent of potentiation,the second potentiation elicited by micromolar diazepam was insensitive to flumazenil.Midazolam,clonazepam,and loraz⁃epam at 200μmol·L-1 exhibited similar flumaze⁃nil-insensitive effects onα1β2γ2,α2β2γ2 andα5β2γ2 receptors,whereas the potentiation induced by 200μmol·L-1 zolpidem or triazolam was abol⁃ished by flumazenil.Consistent with the in vitro results,flumazenil antagonized the zolpidem(50 mg·kg-1)-induced LORR,but not those induced by 50 mg·kg-1 diazepam or 100 mg·kg-1 midazolam.CONCLUSION The existence of non-classical BZD binding sites on certain GABAA receptor subtypes and the flumazenil-insensitive effects depend on the chemical structures of the allosteric modulators.

目的探究国内外近十年体外膜肺氧合(extracorporeal membrane oxygenation,ECMO)研究的热点和趋势。方法采用CiteSpace分析软件对2013~2023年期间WOS数据库、中国知网(CNKI)和万方数据库中检索到的以ECMO为主题的核心数据进行分析,对发文量,机构,国家,关键词频次、聚类和时间线等进行可视化分析。结果2013~2023年期间共检索出2378篇中文文献和3544篇英文文献。对作者、国家、地区和研究机构的分析显示,WOS数据库中发文量最多的是美国,发文量达1146篇,远超过其他国家,且中介中心性值高(0.18),影响力较大。中文数据库中较活跃的关键词聚类是"器官移植"和"新生儿婴幼儿";WOS数据库中较活跃的关键词聚类是"acute respiratory distress syndrome"和"cardiogenic shock"。时间线分析显示,国内近两年的研究方向是"预后""输血"和"抗凝",国外近两年的研究方向是"COVID-19""guidelines""cohort"和"heparin"。结论国内学者多为独立性研究,文章发表量大,但影响力不足,缺乏与国内外学者的合作,建议建立ECMO领域共享数据库。与国外研究进展相比,国内研究缺乏指南共识的建立和探讨,建议关注此方向的深入研究,加强与国内外研究者的合作与交流,促进ECMO应用科学化、规范化发展。
