Excessive fat deposition in obese subjects promotes the occurrence of metabolic diseases,such as type 2 diabetes mellitus(T2DM),cardiovascular diseases,and non-alcoholic fatty liver disease(NAFLD).Adipose tissue is not only the main form of energy storage but also an endocrine organ that not only secretes adipocytokines but also releases many extracellular vesicles(EVs)that play a role in the regulation of whole-body metabolism.Exosomes are a subtype of EVs,and accumulating evidence indicates that adipose tissue exosomes(AT Exos)mediate crosstalk between adipose tissue and multiple organs by being transferred to targeted cells or tissues through paracrine or endocrine mechanisms.However,the roles of AT Exos in crosstalk with metabolic organs remain to be fully elucidated.In this review,we summarize the latest research progress on the role of AT Exos in the regulation of metabolic disorders.Moreover,we discuss the potential role of AT Exos as biomarkers in metabolic diseases and their clinical application.

This research investigates the ecological importance,changes,and status of mangrove wetlands along China’s coastline.Visual interpretation,geological surveys,and ISO clustering unsupervised classification methods are employed to interpret mangrove distribution from remote sensing images from 2021,utilizing ArcGIS software platform.Furthermore,the carbon storage capacity of mangrove wetlands is quantified using the carbon storage module of InVEST model.Results show that the mangrove wetlands in China covered an area of 278.85 km2 in 2021,predominantly distributed in Hainan,Guangxi,Guangdong,Fujian,Zhejiang,Taiwan,Hong Kong,and Macao.The total carbon storage is assessed at 2.11×10^(6) t,with specific regional data provided.Trends since the 1950s reveal periods of increase,decrease,sharp decrease,and slight-steady increases in mangrove areas in China.An important finding is the predominant replacement of natural coastlines adjacent to mangrove wetlands by artificial ones,highlighting the need for creating suitable spaces for mangrove restoration.This study is poised to guide future mangroverelated investigations and conservation strategies.

Whispering gallery mode(WGM)microtoroid resonators are one of the most sensitive biochemical sensors in existence,capable of detecting single molecules.The main barrier for translating these devices out of the laboratory is that light is evanescently coupled into these devices though a tapered optical fiber.This hinders translation of these devices as the taper is fragile,suffers from mechanical vibration,and requires precise positioning.Here,we eliminate the need for an optical fiber by coupling light into and out from a toroid via free-space coupling and monitoring the scattered resonant light.A single long working distance objective lens combined with a digital micromirror device(DMD)was used for light injection,scattered light collection,and imaging.We obtain Q-factors as high as 1:6´108 with this approach.Electromagnetically induced transparency(EIT)-like and Fano resonances were observed in a single cavity due to indirect coupling in free space.This enables improved sensing sensitivity.The large effective coupling area(~10μm in diameter for numerical aperture=0.14)removes the need for precise positioning.Sensing performance was verified by combining the system with the frequency locked whispering evanescent resonator(FLOWER)approach to perform temperature sensing experiments.A thermal nonlinear optical effect was examined by tracking the resonance through FLOWER while adjusting the input power.We believe that this work will be a foundation for expanding the implementation of WGM microtoroid resonators to real-world applications.

Malignant hyperthermia(MH)is a genetic disorder of skeletal muscle cells that affects muscle cytoplasmic calcium homeostasis,with high mortality and low morbidity.Generally,it presents with non-specific signs of a hypermetabolic response,including high fever,tachycardia,and elevated end-tidal carbon dioxide(ETCO_(2)).The successful treatment lies in the timely recognition and early use of dantrolene.[1]As an inhibitor of Ca2+release through ryanodine receptor(RYR)channels,the skeletal muscle relaxant dantrolene has proven to be both a valuable experimental probe of intracellular Ca2+signaling and a lifesaving treatment for MH.[2]Dominant mutations in the skeletal muscle RYR1 gene are well-recognized causes of both malignant hyperthermia susceptibility(MHS)and central core disease(CCD).

Enabling flexible fibers with light-emitting capabilities has the potential to revolutionize the design of smart wearable interactive devices.A recent publication in Light Science&Application,an interdisciplinary team of scientists led by Prof.Yan-Qing Lu and Prof.Guangming Tao has realized a highly flexible,uniformly luminescent photochromic fiber based on a mass-produced thermal drawing method.It overcomes the shortcomings of existing commercial lightdiffusing fibers,exhibiting outstanding one-dimensional linear illumination performance.The research team integrated controllable photochromic fibers into various wearable interaction interfaces,providing a novel approach and insights to enable human-computer interaction.

The author studies the 2D isentropic Euler equations with the ideal gas law.He exhibits a set of smooth initial data that give rise to shock formation at a single point near the planar symmetry.These solutions to the 2D isentropic Euler equations are associated with non-zero vorticity at the shock and have uniform-in-time■-H■lder bound.Moreover,these point shocks are of self-similar type and share the same profile,which is a solution to the 2D self-similar Burgers equation.The proof of the solutions,following the3D construction of Buckmaster,Shkoller and Vicol (in 2023),is based on the stable 2D self-similar Burgers profile and the modulation method.

Hyperplasia and migration of fibroblast-like synoviocytes(FLSs)are the key drivers in the pathogenesis of rheumatoid arthritis(RA)and joint destruction.Abundant Yes-associated protein(YAP),which is a powerful transcription co-activator for proliferative genes,was observed in the nucleus of inflammatory FLSs with unknown upstream mechanisms.Using Gene Expression Omnibus database analysis,it was found that Salvador homolog-1(SAV1),the pivotal negative regulator of the Hippo-YAP pathway,was slightly downregulated in RA synovium.However,SAV1 protein expression is extremely reduced.subsequently,it was revealed that SAV1 is phosphorylated,ubiquitinated,and degraded by interacting with an important serine-threonine kinase,G protein-coupled receptor(GPCR)kinase 2(GRK2),which was predominately upregulated by GPCR activation induced by ligands such as prostaglandin E2(PGE2)in RA.This process further contributes to the decreased phosphorylation,nuclear translocation,and transcriptional potency of YAP,and leads to aberrant FLSs proliferation.Genetic depletion of GRK2 or inhibition of GRK2 by paroxetine rescued SAV1 expression and restored YAP phosphorylation and finally inhibited RA FLSs proliferation and migration.Similarly,paroxetine treatment effectively reduced the abnormal proliferation of FLSs in a rat model of collagen-induced arthritis which was accompanied by a significant improvement in clinical manifestations.Collectively,these results elucidate the significance of GRK2 regulation of Hippo-YAP signaling in FLSs proliferation and migration and the potential application of GRK2 inhibition in the treatment of FLSs-driven joint destruction in RA.

Anthocyanins are secondary metabolites induced by environmental stimuli and developmental signals.The positive regulators of anthocyanin biosynthesis have been reported,whereas the anthocyanin repressors have been neglected.Although the signal transduction pathways of gibberellin(GA)and jasmonic acid(JA)and their regulation of anthocyanin biosynthesis have been investigated,the cross-talk between GA and JA and the antagonistic mechanism of regulating anthocyanin biosynthesis remain to be investigated.In this study,we identified the anthocyanin repressor MdbHLH162 in apple and revealed its molecular mechanism of regulating anthocyanin biosynthesis by integrating the GA and JA signals.MdbHLH162 exerted passive repression by interacting with MdbHLH3 and MdbHLH33,which are two recognized positive regulators of anthocyanin biosynthesis.MdbHLH162 negatively regulated anthocyanin biosynthesis by disrupting the formation of the anthocyanin-activated MdMYB1-MdbHLH3/33complexes and weakening transcriptional activation of the anthocyanin biosynthetic genes MdDFR and MdUF3GT by MdbHLH3 and MdbHLH33.The GA repressor MdRGL2a antagonized MdbHLH162-mediated inhibition of anthocyanins by sequestering MdbHLH162 from the MdbHLH162-MdbHLH3/33 complex.The JA repressors MdJAZ1 and MdJAZ2 interfered with the antagonistic regulation of MdbHLH162 by MdRGL2a by titrating the formation of the MdRGL2a-MdbHLH162 complex.Our findings reveal that MdbHLH162 integrates the GA and JA signals to negatively regulate anthocyanin biosynthesis.This study provides new information for discovering more anthocyanin biosynthesis repressors and explores the cross-talk between hormone signals.

On the half-quantized Hall conductance of massive surface electrons in magnetic topological insulator films

In topological insulators,massive surface states resulting from local symmetry breaking were thought to exhibit a half-quantized Hall conductance,obtained from the low-energy effective model in an infinite Brillouin zone.In a lattice model,the surface band is composed of a combination of surface states and bulk states.The massive surface states alone may not be enough to support an exact one-half quantized surface Hall conductance in a finite Brillouin zone and the whole surface band always gives an integer quantized Hall conductance as enforced by the TKNN theorem.To explore this,we investigate the band structures of a lattice model describing the magnetic topological insulator film that supports the axion insulator,Chern insulator,and semi-magnetic topological insulator phases.We reveal that the gapped and gapless surface bands in the three phases are characterized by an integer-quantized Hall conductance and a half-quantized Hall conductance,respectively.We propose an effective model to describe the three phases and show that the low-energy dispersion of the surface bands inherits from the surface Dirac fermions.The gapped surface band manifests a nearly half-quantized Hall conductance at low energy near the center of Brillouin zone,but is compensated by another nearly half-quantized Hall conductance at high energy near the boundary of Brillouin zone because a single band can only have an integer-quantized Hall conductance.The gapless band hosts a zero Hall conductance at low energy but is compensated by another half-quantized Hall conductance at high energy,and thus the half-quantized Hall conductance can only originate from the gapless band.Moreover,we calculate the layer-resolved Hall conductance of the system.The conclusion suggests that the individual gapped surface band alone does not support the half-quantized surface Hall effect in a lattice model.
