Dietary fatty acids and adipose tissue inflammation at the crossroad between obesity and colorectal cancer

Excess adiposity, a worldwide-growing pathological condition, is now recognized as a main risk factor for most chronic diseases including colorectal cancer (CRC). Obese subjects show an increased cancer incidence with obesity representing an important indicator of survival, prognosis, recurrence and response to therapy. A low-grade chronic inflammation of metabolically active tissues including the adipose tissue (AT), defined as meta-inflammation, is a main feature of obesity. Fatty acids (FA), the main AT components, are important modulators of inflammation, and the type of FA stored in AT critically affects tissue functions. Their profile within AT mirrors FA dietary intake but also depends on a metabolic control. Obesity, changes in the habitual diet, weight loss or pathological conditions like CRC influence FA profile of AT pointing to these molecules as important actors in AT dysfunction and meta-inflammation, that characterize metabolic diseases and may favor cancer development. Worth of note, diet is receiving growing attention as a main determinant in cancer prevention due to its capacity to modulate immune response and inflammation. Alterations in the balance between different families of FA may contribute to generate a pro-inflammatory profile with deleterious effects on metabolic and immune homeostasis at both local and systemic levels. This review focuses on FA as regulators of human AT inflammation discussing the role of obesity-, diet-, and weight loss-associated changes in FA profile in this process. The relevance of FA composition of AT in linking diet, obesity and CRC will be also reviewed.

Chemotherapy-induced peripheral neuropathy （CIPN） is one of the most common and severe adverse effects related to cancer treatment. Unfortunately, although several agents and protocols have been proposed, no prophylactic strategies have yet to be proven useful. Therefore, new alternative therapies have been considered for CIPN prevention. Herbal medicine in Japan, called Kampo medicine, is derived from traditional Chinese medicine. Goshajinkigan （GJG） is a Kampo medicine, that is comprised of ten herbs. The aim of this work is to analyse the results of pre-clinical and clinical studies on the potential applications of GJG in CIPN prevention.

Heterogeneous catalysis under flow for the 21st century fine chemical industry

Due to metal leaching and poor catalyst stability, the chemical industry's fine chemical and pharmaceutical sectors have been historically reluctant to use supported transition metal catalysts to manufacture fine chemicals and active pharmaceutical ingredients. With the advent of new generation supported metal catalysts and flow chemistry, we argue in this study, this situation is poised to quickly change. Alongside heterogenized metal nanoparticles, both single-site molecular and single-atom catalyst will become ubiquitous. This study offers a critical outlook taking into account both technical and economic aspects.

Size contrast of Pt nanoparticles formed on neighboring domains within suspended and supported graphene

The relatively small size of thin （one or few layers） graphene flakes makes it extremely difficult to study the behavior of suspended graphene by characterization techniques other than the electron microscopies. Herein, we exploited the capability of spatially resolved photoemission in combination with high resolution transmission electron microscopy to investigate the interaction of thermally evaporated Pt atoms on suspended and supported graphene. Spectroscopic and microscopic analyses reveal that the nucleation of nanometer- sized Pt particles in these two regions exhibit different trends. While only small nanometer-sized islands are present on the supported graphene, relatively larger clusters of islands were also found on the suspended flakes. The X-ray photoemission C ls core levels acquired after the Pt deposition show an increase in the number of vacancies in the graphene sheets.

Neurological diseases associated with autoantibodies targeting the voltage-gated potassium channel complex:immunobiology and clinical characteristics

Voltage-gated potassium channels(VGKCs)represent a group of tetrameric signaling proteins with several functions,including modulation of neuronal excitability and neurotransmitter release.Moreover,VGKCs give a key contribution to the generation of the action potential.VGKCs are complexed with other neuronal proteins,and it is now widely known that serum autoantibodies directed against VGKCs are actually directed against the potassium channel subunits only in a minority of patients.By contrast,these autoantibodies more commonly target three proteins that are complexed with alpha-dendrotoxin-labeled potassium channels in brain extracts.These three proteins are contactin-associated protein-2(Caspr-2),leucine-rich,glioma inactivated 1(LGI-1)protein and the protein Tag-1/contactin-2.Neoplasms are detected only in a minority of seropositive patients for VGKC complex-IgG and do not significantly associate with Caspr-2 or LGI-1.Among all the cancers described in association with VGKC complex-IgG,lung carcinoma,thymoma,and hematologic malignancies are the most commonly detected.We will review all the major neurological conditions associated with VGKC complex-IgG.These include Isaacs’syndrome,Morvan syndrome,limbic encephalitis,facio-brachial dystonic seizures,chorea and other movement disorders,epilepsy,psychosis,gastrointestinal neuromuscular diseases,a subacute encephalopathy that mimics Creutzfeldt-Jakob prion disease both clinically and radiologically and autoimmune chronic pain.The vast majority of these conditions are reversible by immunotherapy,and it is becoming increasingly recognized that early diagnosis and detection of VGKC complex-IgG is critical in order to rapidly start the treatment.As a result,VGKC complex-IgG are now part of the investigation of patients with unexplained subacute onset of epilepsy,memory or cognitive problems,or peripheral nerve hyperexcitability syndromes.

Ferrofluid measurements of bottom velocities and shear stresses

A novel direct measurement strategy of bottom velocities and shear stresses based on the use of ferrofluids is presented. Such a strategy overcomes some of the limits of state-of-the-art instruments. A preliminary experimental campaign has been carried out in the presence of currents in steady flow conditions in order to test the effects of ferrofluid quantity and of the controlling permanent magnetic force. An alternating current （AC） circuit and a direct current （DC） conditioning circuit have been tested. For velocities larger than 0.05 m/s, the near-bottom velocity-output voltage calibration curve has a monotone parabolic shape. The sensitivity of the instrument is increased by a factor of 30 when the DC circuit is used.

Towards a better understanding of anesthesia emergence mechanisms: Research and clinical implications

Emergence from anesthesia(AE) is the ending stage of anesthesia featuring the transition from unconsciousness to complete wakefulness and recovery of consciousness(RoC). A wide range of undesirable complications, including coughing, respiratory/cardiovascular events, and mental status changes such as emergence delirium, and delayed RoC, may occur during this critical phase. In general anesthesia processes, induction and AE represent a neurobiological example of "hysteresis". Indeed, AE mechanisms should not be simply considered as reverse events of those occurring in the induction phase. Anesthesia-induced loss of consciousness(LoC) and AE until RoC are quite distinct phenomena with, in part, a distinct neurobiology. Althoughanaesthetics produce LoC mostly by affecting cortical connectivity, arousal processes at the end of anesthesia are triggered by structures deep in the brain, rather than being induced within the neocortex. This work aimed to provide an overview on AE processes research, in terms of mechanisms, and EEG findings. Because most of the research in this field concerns preclinical investigations, translational suggestions and research perspectives are proposed. However, little is known about the relationship between AE neurobiology, and potential complications occurring during the emergence, and after the RoC. Thus, another scope of this review is to underline why a better understanding of AE mechanisms could have significant clinical implications, such as improving the patients' quality of recovery, and avoiding early and late postoperative complications.

International transferability of macro-level safety performance functions: a case study of the United States and Italy

Safety performance functions(SPFs),or crash-prediction models,have played an important role in identifying the factors contributing to crashes,predicting crash counts and identifying hotspots.Since a great deal of time and effort is needed to estimate an SPF,previous studies have sought to determine the transferability of particular SPFs;that is,the extent to which they can be applied to data from other regions.Although many efforts have been made to examine micro-level SPF transferability,few studies have focused on macro-level SPF transferability.There has been little transferability analysis of macro-level SPFs in the international context,especially between western countries.This study therefore evaluates the transferability of SPFs for several states in the USA(Illinois,Florida and Colorado)and for Italy.The SPFs were developed using data from counties in the United States and provincias in Italy,and the results revealed multiple common significant variables between the two countries.Transferability indexes were then calculated between the SPFs.These showed that the Italy SPFs for total crashes and bicycle crashes were transferable to US data after calibration factors were applied,whereas the US SPFs for total and bicycle crashes,with the exception of the Colorado SPF,could not be transferred to the Italian data.On the other hand,none of the pedestrian SPFs developed was transferable to other countries.This paper provides insights into the applicability of macro-level SPFs between the USA and Italy,and shows a good potential for international SPF transferability.Nevertheless,further investigation is needed of SPF transferability between a wider range of countries.

Totally robotic vs 3D laparoscopic colectomy: A single centers preliminary experience

AIM: to compare robotic and three-dimensional(3D) laparoscopic colectomy based on the literature and our preliminary experience.METHODS: This retrospective observational study compared operative measures and postoperative outcomes between laparoscopic 3D and robotic colectomy for cancer. From September 2013 to September 2014,24 robotic colectomies and 23 3D laparoscopic colectomy were performed at our Department. Data were analyzed and reported both by approach and by colectomy side. Robotic left colectomy(RL) vs laparoscopic 3D left colectomy(LL 3D) and Robotic right colectomy(RR) vs laparoscopic 3D(LR 3D). Rectal cancer procedures were not included.RESULTS: There were 18 RR and 11 LR 3D,6 RL and 12 LL 3D. As regards LR 3D,extracorporeal anastomosis(EA) was performed in 7 patients and intracorporeal anastomosis(IA) in 4; the RR group included 14 IA and 4 EA. There was no mortality. Median operative time was higher for the robotic group while conversion rate(12.5% vs 13%) and lymph nodes removed(14 vs 13) were similar for both. First flatus time was 1 d for RR and 2 d the other patient groups. Oral intake was resumed in 1 d by LR and in 2 d by the other patients(P = 0.012). Overall cost was €4950 and €1950 for RL and LL 3D,and €4450 and €1450 for RR and LR 3D,respectively. CONCLUSION: There were no differences betweenRR and LR 3D,except that IA was easier with RR,and probably contributed with the learning curve to the longer operative time recorded. Both techniques offer similar advantages for the patient with significantly different costs. In left colectomies robotic colectomy provided better outcomes,especially in resections approaching the rectum.
