The role of element homeostasis in neoplastic disease pathogenesis is be-yond question. The imbalance of trace elements precisely underlies the ini-tiation and promotion of tumor pathology. The aim of the study was to in-vestigate blood and tissue macroelements, microelements and hemoproteins level in brain tumors and their intermolecular interactions. samples of blood and brain tumor tissues were investigated. Detection of myoglobin level was implemented by the reaction of passive hemagglutination and immunoturbidimetric test. Catalase activity was determined by the method of Beer and sizer. Free radical activity was determined by the method of induced biochemiluminescence. Microelements level was investigated by usage of atomic emission spectrometry. To build the networks of studied hemoprotein interactions with signaling pathways of proteins, expressed in brain tumors, molecular interaction databases (sTRiNG, BioGrid) were used. Modern databases of signaling pathways (KEGG) suggest that in normal cells hypoxia can lead to HiF-1A protein synthesis. ros synthesis inhibits the PHD enzyme and triggers the release of calcium ions, and increases proliferation. Calcium ions are triggering factor of apoptosis and cell proliferation. Myoglobin can possibly be the cell adaptation factor towards hypoxia, oxidative stress and element homeostasis violation, and myoglobin level decreasing can additionally stimulate proliferation, by apoptosis inhibition.

Background: Progressive supranuclear palsy (PsP) is a clinicopathological syndrome related to τdeposits and in linkage disequilibrium with τpolymorphisms. some rare familial PsP cases have been related to τgene mutations. Objective: To present the clinical, pathological, and molecular data of one family with early-onset autosomal dominant PsP. Design: We performed clinical examinations, quantitative neurological tests, positron emission tomographic scans with fluorodopa F 18 and raclopride C 11, analysis of τmutations, neuropathological examinations, and protein analyses on brain specimens. Results: Three family members had PsP confirmed by pathological features in the proband. A novel mutation of τ, G303V, was found in the proband and other family members. τisoforms with 4 microtubule-binding repeats were overexpressed in the proband brain. Conclusions: The G303V mutation of τis associated with autosomal dominant PsP. Expression of 4 microtubule-binding repeat τisoforms is increased in the proband.

Genetic and Phenotypic Parameters in the selection of Upland Rice Genotypes

To assist in upland rice breeding programs, estimated genetic and phenotypic parameters are extremely important in the selection of superior lines. Thus, the objective of this study was estimate the genetic and phenotypic parameters of upland rice lines of the breeding program of UFLA in partnership with Embrapa Arroz e Feijao and Epamig. The experiment was installed in an experimental area of the Department of Agriculture at UFLA. it was evaluated 36 upland rice lines of the preliminary trial of the 2014/15 season, in randomized blocks, with three replications and plots consisted of five rows and each row has four meters. For this, it was evaluated the characters: plant height (cm), yield (kg·ha-1), 100-grain weight, days until flowering, percentage of filled grains and rice blast severity. The variance analysis and genetic parameters were estimated using the software sAs. To evaluate the experimental precision was estimated the accuracy, which varied between 63% and 98%, showing experimental precision, moderate to high. The lines differed significantly for all characters, meaning there is genetic variability among genotypes evaluated. With intensity of selection of 10% of lines, it was obtained a genetic gain of 325.29 kg·ha-1 in the character productivity, a plant height (cm) reduction in 5.50 cm, and days until flowering, with a reduction of 9.23 days, with selection of earlier plants. The estimated values for heritability ranged from 38% - 96%. Estimates confirm the possible success with the selection of the upland rice lines for these characters.

1996～1997期间,在一个富营养湖(湖E)和一个贫营养湖(湖O)中进行了富营养化和有机污染物(PCB,DDTs及γ-HCH)的交互作用研究,两湖的地貌形态和水化学相似。集水区的土壤样品表明两湖暴露于相似的大气条件下。与湖O相比湖E没有观测到生物稀释。湖E的沉积物(1.5倍)、表面沉积物(1.4倍)和河鲈(Perca fluviatilis)(2.4倍)的有机碳和脂质物质中的PCB水平较高,并且微粒的沉降流较大,沉积物捕集器和沉淀物表面PCB的水平也较高。湖E中PCB的大部分转运主要是靠浮游植物。DDD/DDE比值较高似乎与p,p’-DDT的高负载相关而非厌氧的原因。所记录的湖O深层沉积岩心的高DDD/DDE比值和sDDT的高水平以及在此期间沉积最多,大概是由于从集水区流入的前体DDT最高的缘故。

目的评价患者临床特征及实验室检查结果对18F-脱氧葡萄糖(FDG) PET/CT显像诊断不明原因发热(FUO)的影响。方法回顾性纵向分析FUO患者的PET/CT显像结果。参考标准为血清学检查、各种细菌培养或活组织检查结果,必要时参考其他实验室检查或临床随访结果。收集患者PET/CT显像近几天内的临床特征、炎性标志物、蛋白质分析、血清学检查和细菌培养结果。根据不同病因最终诊断分为3组:第1组为感染或肿瘤,第2组为脉管炎、自身免疫性疾病或非感染性炎性疾病,第3组为未明确诊断的自限性发热或持续性发热。PET/CT显像结果分为阳性或阴性、有助于或无助于诊断发热来源。分析患者临床特征和实验室检查结果对PET/CT结果的影响。结果 67例患者最终诊断分组为:第1组25例,第2组20例,第3组22例。89.6%的患者炎性标志物阳性,28.4%的蛋白质检测阳性,20.9%的细菌培养结果阳性。67例患者中,PET/CT显像阳性52例,25例有助于诊断发热来源,其中对第1组和第2组帮助尤其大。PET/CT显像的灵敏度、特异性和准确性分别为84%、31%和61%。PET/CT显像结果与最终诊断(P=0.035)和细菌培养结果(P=0.037)相关,而与其他临床特征或实验室检查结果无明显相关性。结论18F-FDG PET/CT显像诊断FUO的灵敏度高,但特异性低,可能的原因是FUO的病原范围很广,也有一些是自限性发热。最有可能从PET/CT显像获益的患者是那些多种炎性标志物阳性的患者,这些患者有活动性病变的可能性更高。

Occurrence of Nanobacteria-Like Particles in Renal stones of a southern Brazilian Population

Purpose: identifying the source of stone formation in recurrent stone formers has always been a big problem. Material and Methods: in this study kidney stones from 52 patients were submitted to direct examination by scanning electronic microscopy (sEM) after manual fracture and 27 calculi were cultured in Eagle’s Minimum Essential Medium (E-MEM) and Brain-Heart infusion (BHi) for eight weeks at 37°C in 5% CO2 atmosphere. Twenty-seven powdered and demineralized stones were suspended in sterile PBs, filtered through 0.22 m-pore-size sterile filters Millex (Millipore, Massachusetts, UsA) and submitted to DNA extraction (Quiagen-Brazil). Platinum PCR superMiX (GiBCO-BRL), primers (invitrogen), and Ultra Pure Water (Advanced Biotechnologies, Columbia, MD) were used for PCR (Polymerase Chain Reaction), which was generally conducted for 30 or 35 cycles with annealing of primers for 40 sec at 55°C and extension for 1 min at 72°C. Results: in 36 out 52 (69%) kidney stones it was detected the presence of biofilm coating the mineral surface of the stone when examined by sEM, consisting of coccoid particles, isolated or clustered, with diameter of 500 nm or less. Eleven out 27 (41%) kidney stone cultures produced white-colored sediment on the bottom of the tubes after eight-week incubation, revealing tiny structures similar to those observed directly by sEM. These structures were similar in size and morphology to spherical apatite particles previously observed in human kidney stones and named as nanobacteria (NB). No PCR products were observed in the samples. Conclusion: We found a strong correlation between renal stones and calcifying nanoparticles (CNP) in this study and these results open a new insight on this area to explore the etiology of stone formation. Whether NB/CNP are truly microorganisms or self-propagating mineral compounds is still controversial and its contribution, if any, in apatite nucleation and crystal growth remains uncertain.

Excitotoxicity and neuronal cell death: Glutamate is the main excitatory neurotransmitter of the central nervous system and functionally involved in most brain activities, including brain development, synaptic plasticity, learning and memory. Exc让atory synaptic transmission is primarily mediated by ligana-gated ion channels, including a-amino-3-hydroxy-5-methylisoxazoie-4-propionic acid (AMPA), N-methyl-D-aspartate (NMDA) and kainate receptors. Activation of glutamate receptors, particularly NMDA receptors, usually leads to calcium influx, whicn can act as a second messenger for several processes to mediate synaptic activity and brain function. Nevertheless, excessive release of glutamate neurotransmitter may produce intracellular calcium overload, leading to a cascade of events mediating cytoskeleton damage accompanied w让h reactive oxygen species (ros) generation, mitochondrial dysfunction and ultimately neuronal cell death. These toxic effects of glutamate are known as excitotoxicity. Neuronal excitotoxicity has been linked to several acute and chronic brain diseases, such as stroke/ischemia, epilepsy and a range of neurodegenerative disorders, including Alzheimer's disease (AD), Huntingtons disease, amyotrophic lateral sclerosis (ALs) and Parkinsons disease (PD), contributing to the neuronal lost in different brain regions. Unfortunately, treating nervous system disorders with general glutamate receptor blockers has been associated with undesirable side effects, becoming increasingly necessary to unravel downstream effectors in the excitotoxicity-dependent cell death pathway in order to develop novel therapeutic strategies.

*** strongest locus which associated with type 2 diabetes(T2D)by the common variant rs7903146 is the transcription factor 7-like 2 gene(TCF7L2).We aimed to quantify the interaction of diet/lifestyle interventions and the genetic effect of TCF7L2 rs7903146 on glycemic traits,body weight,or waist circumference in overweight or obese adults in several randomized controlled trials(RCTs).*** October 2016 to May 2018,a large collaborative analysis was performed by pooling individualparticipant data from 7 *** RCTs reported changes in glycemic control and adiposity of the variant rs7903146 after dietary/lifestyle-related interventions in overweight or obese *** treatment interaction models which used the genetic effect encoded by the allele dose and common covariates were applicable to individual participant data in all *** the joint analysis,a total of 7 eligible RCTs were included(n=4,114).importantly,we observed a significant effect modification of diet/lifestyle-related interventions on the TCF7L2 variant rs7903146 and changes in fasting *** with the control group,diet/lifestyle interventions were related to lower fasting glucose by-3.06(95%Ci,-5.77 to-0.36)mg/dL(test for heterogeneity and overall effect:i^(2)=45:1%,pisk ***,regardless of genetic risk,diet/lifestyle interventions were associated with lower waist ***,there was no significant change for diet/lifestyle interventions in other glycemic control and adiposity traits per one copy of TCF7L2 risk *** findings suggest that carrying the TCF7L2 T risk allele may have a modestly greater benefit for specific diet/lifestyle interventions to improve the control of fasting glucose in overweight or obese adults.

Postprandial hypertriglyceridemia and hyperglycemia are considered risk factors for cardiovascular disease. Evidence suggests that postprandial hypertriglyceridemia and hyperglycemia induce endothelial dysfunction and inflammation through oxidative stress. statins and angiotensin type 1 receptor blockers have been shown to reduce oxidative stress and inflammation, improving endothelial function. Methods and Results-Twenty type 2 diabetic patients ate 3 different test meals: a high-fat meal, 75 g glucose alone, and a high-fat meal plus glucose. Glycemia, triglyceridemia, endothelial function, nitrotyrosine, C-reactive protein, intercellular adhesion molecule-1, and interleukin-6 were assayed during the tests. subsequently, diabetics took atorvastatin 40 mg/d, irbesartan 300 mg/d, both, or placebo for 1 week. The 3 tests were performed again between 5 and 7 days after the start of each treatment. High-fat load and glucose alone produced a decrease in endothelial function and increases in nitrotyrosine, C-reactive protein, intercellular adhesion molecule-1, and interleukin-6. These effects were more pronounced when high-fat load and glucose were combined. short-term atorvastatin and irbesartan treatments significantly counterbalanced these phenomena, and their combination was more effective than either therapy alone. Conclusions-This study confirms an independent and cumulative effect of postprandial hypertriglyceridemia and hyperglycemia on endothelial function and inflammation, suggesting oxidative stress as a common mediator of such an effect. short-term treatment with atorvastatin and irbesartan may counterbalance this phenomenon; the combination of the 2 compounds is most effective.
