Derived from the bacterial adaptive immune system,CRISPR technology has revolutionized conventional genetic engineering methods and unprecedentedly facilitated strain *** this review,we outline the fundamental CRISPR tools that have been employed for strain *** tools include CRISPR editing,CRISPR interference,CRISPR activation and protein *** further characterize the CRISPR technology,we present current applications of these tools in microbial systems,including model-and non-model industrial ***,we point out the major challenges of the CRISPR tools when utilized for multiplex genome editing and sophisticated expression *** address these challenges,we came up with strategies that place emphasis on the amelioration of DNA repair efficiency through CRISPR-Cas9-assisted ***,multiple promising research directions were proposed,mainly focusing on CRISPR-based construction of microbial ecosystems toward high production of desired chemicals.

本文以"气"之过程宇宙论为诠释语境,对儒家人格观进行了较为清晰的透视。通过参较唐君毅、李泽厚等人的相关看法,本文指出,只有遵循儒家伦理学自身的运作方式方能达到对儒家"成人"之先决条件的细微体察。在"人—类"与"成人"的概念之间存有差异:前者强调,人具有抽象性与生物性的同一本质,个人虽如彼此独立之原子,然而均领受同样之本质,故能成为同"类"的一员。而儒家的"成人"概念,始于独特而多样性的家庭关系。此一家庭关系,膏泽深厚,洵属既伴随人之初生、却又可无尽延展的生命资源,浸润于斯,相互陶养,人格便终可抵于成型。与此不同,西方的"人—类"概念,具有根本的个人主义蕴涵,所造就的是其性格特征可以数字精确算计的"某个人",他们通过形成习惯来获得道德品质。儒家的"成人",是在关系的脉络之中,通过有效而又有品味涵养的个性行为来寻求独特而卓越的品格。个性并非先天给予,而是经由陶养构成人格的诸重关系而后天养成。

A Frisch-Newton Algorithm for Sparse Quantile Regression

Recent experience has shown that interior-point methods using a log barrierapproach are far superior to classical simplex methods for computing solutions to large parametricquantile regression problems. In many large empirical applications, the design matrix has a verysparse structure. A typical example is the classical fixed-effect model for panel data where theparametric dimension of the model can be quite large, but the number of non-zero elements is quitesmall. Adopting recent developments in sparse linear algebra we introduce a modified version of theFrisch-Newton algorithm for quantile regression described in Portnoy and Koenker[28]. The newalgorithm substantially reduces the storage (memory) requirements and increases computational *** modified algorithm also facilitates the development of nonparametric quantile regressionmethods. The pseudo design matrices employed in nonparametric quantile regression smoothing areinherently sparse in both the fidelity and roughness penalty components. Exploiting the sparsestructure of these problems opens up a whole range of new possibilities for multivariate smoothingon large data sets via ANOVA-type decomposition and partial linear models.

With more than 60 percent of China’s 1.34 billion population,about 50 percent of its most productive arable land,and the world’s second largest and rapidly growing national GDP exposed to flooding of various kinds,China has an intractable flood *** potential impacts of climate change and continued intensification of floodplain development driven by rapid industrialization and urbanization,it is very likely that China will see a continued increase of exposure to floods this *** overview article outlines and discusses fundamental dilemmas,plausible pathways,and key options for managing future flood risks in China in the context of rapid socioeconomic transition and climate *** dilemmas are the embedded difficult trade-off choices,from balancing economic development with flood vulnerability reduction,to coordination and cooperation among increasingly diverse actors and across *** plausible pathways,this article argues that a resilience strategy for managing flood risk is *** would require human adjustment to flood,not by aiming for full protection and control but by adjusting our use of floodplains,integrating and experimenting with a wide range of flood risk management options,so that a dynamic balance is maintained between exposure and coping capacity and flood risk is contained at an acceptable *** variability and uncertainty lies at the heart of such a flood resilience centered *** the flood toll cannot be had without trade-offs in economic development,food production,and agricultural productivity.

Lifelong education was developed in Europe in the 1970's. A learning society arises when principles of adult education are properly deployed. Vertical integration concerns learning throughout the lifespan. Horizontal integration requires education to occur in informal and nonformal as well as familiar formal settings. Democratization demands the dismantling of barriers that impede access to education and involvement of learners in the design and management of their own education. Chinese citizens have always learned from a broad array (of not just educational settings). The initiative to build 61 "learning cities" demonstrates a genius for adapting western ideas. China has already transcended some limitations of European ideas about lifelong education. However, because of ageism, the obsession with formal education and need to navigate within the contours of the party-state, building a learning society faces special challenges. In China, as elsewhere, universities are not leading these initiatives. They need to become more flexible and open.

Details are given herein of the development and application of a three-dimensional layer integrated numerical model to predict geo-morphological changes in estuarine waters. An Alternating Direction Implicit finite difference algorithm has been used for solving the governing differential equations, which include the conservation of mass and momentum for the hydrodynamics, the transport equation for suspended sediment fluxes and the sediment mass conservation equation for bed level changes. The model includes a modified term for the initiation of motion, which takes account of the unsteady nature of the flow. Model predictions have been compared with results of other model studies reported in the literature and also laboratory measurements, with the refined criteria for the initiation of motion for tidal currents giving improved results. Finally, the model has been applied to predict bed level changes due to suspended sediment fluxes in the Humber Estuary, UK.

Increasingly countries are seeking to reduce emission of greenhouse gases from the agricultural industries,and livestock production in particular,as part of their climate change *** many reviews update progress in mitigation research,a quantitative assessment of the efficacy and performance-consequences of nutritional strategies to mitigate enteric methane(CH_(4))emissions from ruminants has been lacking.A meta-analysis was conducted based on 108 refereed papers from recent animal studies(2000-2020)to report effects on CH4 production,CH_(4) yield and CH_(4) emission intensity from 8 dietary *** interventions(oils,microalgae,nitrate,ionophores,protozoal control,phytochemicals,essential oils and 3-nitrooxypropanol).Of these,macroalgae and 3-nitrooxypropanol showed greatest efficacy in reducing CH_(4) yield(g CH_(4)/kg of dry matter intake)at the doses *** confidence intervals derived for the mitigation efficacies could be applied to estimate the potential to reduce national livestock emissions through the implementation of these dietary interventions.
