Expression of hIF-1α and Its Target Genes in the Nanorana parkeri heart：Implications for high Altitude Adaptation

hypoxia-inducible factor 1 alpha（hIF-1α） and its target genes vascular endothelial growth factor（VEGF） and transferrins（TF） play an important role in native endothermic animals＇ adaptation to the high altitude environments. For ectothermic animals – especially frogs – it remains undetermined whether hIF-1α and its target genes（VEGF and TF） play an important role in high altitude adaptation, too. In this study, we compared the gene sequences and expression of hIF-1α and its target genes（VEGF and TF） between three Nanorana parkeri populations from different altitudes（3008 m a.s.l., 3440 m a.s.l. and 4312 m a.s.l.）. We observed that the c DNA sequences of hIF-1A exhibited high sequence similarity（99.38%） among the three altitudinally separated populations; but with increasing altitude, the expression of hIF-1A and its target genes（VEGF and TF） increased significantly. These results indicate that hIF-1α plays an important role in N. parkeri adaptation to the high altitude, similar to its role in endothermic animals.

For species that have a broad geographic distribution, adaptive variation may be attributable to gene expression plasticity. Nanorana parkeri is an anuran endemic to the southern Tibetan Plateau where it has an extensive altitudinal range（2850 to 5100 m asl）. Low oxygen concentration is one of the main environmental characteristics of the Tibetan Plateau. hypoxia-inducible factor α subunits（hIF-1α and hIF-2α, encoded by Endothelial PAs domain protein 1（EPAs1）） and associated genes（e.g., vascular endothelial growth factor（VEGF） and Erythropoietin（EPO）） play crucial roles in maintaining oxygen homeostasis. In this study, we compared the expression of hIF-1A, VEGF, EPAs1 and EPO mRNA between two populations of N. parkeri： one population inhabiting the native high altitudes, and the second living in, and being acclimated to, the lower plains（70 m asl）. The expression of hIF-1A, VEGF and EPAs1 mRNA in the high altitude population were significantly higher than in the acclimated population, whereas there was no significant difference for EPO between two groups. Our results indicated that gene expression plasticity may make significant contributions to local adaptation of species that have broad altitudinal distributions. In addition, we deepen our understanding of the adaptive potential of this species by evaluating the experiments in the scope of its evolutionary history.

Introgression,the incorporation of genetic material from one(sub)species into the gene pool of another by means of hybridization and backcrossing,is a common phenomenon in birds and can provide important insights into the speciation process.In the last decade,the toolkit for studying introgression has expanded together with the development of molecular markers.In this review,we explore how genomic data,the most recent step in this methodological progress,impacts different aspects in the study of avian introgression.First,the detection of hybrids and backcrosses has improved dramatically.The most widely used software package is sTRUCTURE.Phylogenetic discordance(i.***.different loci resulting in discordant gene trees) is another means for the detection of introgression,although it should be regarded as a starting point for further analyses,not as a definitive proof of introgression.specifically,disentangling introgression from other biological processes,such as incomplete lineage sorting,remains a challenging endeavour,although new techniques,such as the D-statistic,are being developed.In addition,phylogenetics might require a shift from trees to networks.second,the study of hybrid zones by means of geographical or genomic cline analysis has led to important insights into the complex interplay between hybridization and speciation.however,because each hybrid zone study is just a single snapshot of a complex and continuously changing interaction,hybrid zones should be studied across different temporal and/or spatial scales.A third powerful tool is the genome scan.The debate on which evolutionary processes underlie the genomic landscape is still ongoing,as is the question whether loci involved in reproductive isolation cluster together in ‘islands of speciation' or whether they are scattered throughout the genome.Exploring genomic landscapes across the avian tree of life will be an exciting field for further research.Finally,the findings from these different methods should be incorporated into specific speciation scena

Necrotic enteritis and antibiotic-free production of broiler chickens:Challenges in testing and using alternative products

The global trend towards raising broiler chickens without the use of in-feed antibiotics(IFAs) means that there is an ongoing need to develop alternative treatments capable of delivering the benefits that IFAs previously provided. IFAs supported the productivity performance of chickens and played a key role in maintaining their health. Necrotic enteritis(NE) is an important disease of broilers that affects health,productivity, and welfare, and was previously well controlled by IFAs. however, with the reduction in IFA use, NE is resurgent in some countries. Vaccines and various feed additives, including pre-, pro-, and postbiotics, phytobiotics, fatty acids, and phage therapies have been introduced as alternative methods of NE control. While some of these feed additives have specific activity against the NE pathogen, Clostridium perfringens, most have the more general goal of reinforcing gut health. Extensive reviews of the effects of many of these feed additives on gut health have been published recently. hence, rather than cover previously well reviewed areas of research this review focuses on the challenges and pitfalls in undertaking experimental assessment of alternative NE treatments and translating laboratory research to real world commercial production settings. The review is based on the author's particular experience,reading, thoughts, and analysis of the available information and inevitably presents a particular understanding that is likely to be at odds with others thinking on these issues. It is put forward to stimulate thinking and discussion on the issues covered.

Extracellular vesicles and angiotensin-converting enzyme 2 in COVID-19 disease

Extracellular vesicles(EVs)are membranous vesicular structures released from almost all eukaryotic cell types under different physiological or pathological conditions.Growing evidence demonstrates that EVs can serve as mediators of intercellular communication between donor and recipient cells or microorganism-infected and noninfected cells.Coronavirus disease 2019(COVID-19)disease is caused by infection of the severe acute respiratory syndrome coronavirus 2(sARs-CoV-2)of host cells in the respiratory system and various extra-pulmonary tissue/organs,resulting in complications of multiple organ systems.As the cell surface receptor,angiotensin-converting enzyme 2(ACE2)mediates cellular entry of sARs-CoV-2 into the host cells in patients with COVID-19.Recent studies have found that ACE2 can be released with EVs,which have been shown to interfere with the entry of the virus into host cells and thus may be involved in COVID-19 pathophysiology.In addition,ACE2,neprilysin(NEP),and thimet oligopeptidase(TOP)are the key enzymes that regulate angiotensin metabolism by converting angiotensin II or angiotensin I to angiotensin 1-7,the latter of which has protective effects in counterbalancing the harmful effects of angiotensin II in COVID-19 disease.This review summarizes the recent research progress regarding EV-associated ACE2,NEP,and TOP and the perspectives of their potential involvement in the pathophysiology of COVID-19 disease.

Mitigating preterm encephalopathy continues to be one of the greatest challenges in perinatal medicine.Preterm encephalopathy is associated with high mortality,serious morbidity,and significant socio-economic impacts on the individuals,their families,and public health sectors and welfare systems that last a lifetime.The cost of disability associated with preterm brain injury continues to rise.Prevention of this injury,and disability,would significantly reduce this socioeconomic burden.

Alzheimer's disease(AD) is a progressive neurodegenerative disease histologically characterized by the presence of extraneuronal plaques,mainly formed by the 42-aminoacid isoform of amyloid-(Aβ_(1-42)),and by intraneuronal neurofibrillary to ngles,mainly formed by the tau protein and its hyperphosphorylated isoforms(p-tau).AD is the most common cause of dementia,with an estimated lifetime risk of about 1 in 10 for men and 1 in 5 for women.
