Bac.ground: The Geriatric.Depression Sc.le (GDS) is widely used to assess depressive symptoms in c.inic.l and researc. settings. This study utilized a 4 fac.or solution for the 30-item GDS to explore differenc.s in the presentation of depressive symptoms in various types of c.gnitive impairment. Method: Retrospec.ive c.art review was c.nduc.ed on 254 c.nsec.tive c.ses of c.mmunity dwelling elderly newly diagnosed with mild Alzheimer’s Dementia (AD) n = 122, mild Vasc.lar Dementia (VaD) n = 71 or Amnestic.Mild c.gnitive Impairment (aMc.) n = 32 and Non-Amnestic.Mc. (nMc.) n = 29. Results: Analysis revealed no signific.nt differenc.s (p 05). No statistic.lly signific.nt differenc.s were found between VaD and nMc. or between the Mc. groups. c.nc.usions: Support is provided for the use of GDS subsc.les in a wide range of c.gnitively impaired elderly. This study suggests in mild dementia the number and type of depressive symptoms vary signific.ntly between AD and VaD. There are indic.tions that aMc. patients are similar in their symptom endorsement to AD and nMc. are similar to VaD whic. is c.nsistent with some of the notions regarding likely trajec.ories of the respec.ive Mc. groups.

Depression and Brain-derived Neurotrophic.Fac.or Levels in Alzheimer's Disease

Bac.ground: Depression is often viewed as a risk fac.or for the development of Alzheimer's disease (AD), however little is known regarding the underlying biologic.l mec.anisms linking these two diseases. Brain-derived neurotrophic.fac.or (BDNF) has been linked to both c.gnitive impairment and depression in past researc.;however few studies have ex-amined this relation in a sample of Alzheimer's patients. The present study sought to address this gap in the literature by examining the relation between serum BDNF levels and depression assessed by the Geriatric.Depression Sc.le (GDS) in a group of patients diagnosed with probable Alzheimer's disease. Methods: Partic.pants inc.uded 169 individuals diagnosed with Probable AD enrolled in the TARc.Longitudinal Researc. c.hort with available BDNF levels and GDS sc.res. The partic.pants were divided into Depressed (N = 20) and Not Depressed (N = 149) based on GDS sc.res. Re-sults: BDNF levels signific.ntly predic.ed level (High vs. Low) of depression (β = 0.066, SE = 0.031, p = 0.034). BDNF levels for the Depressed group were signific.ntly higher than those observed in the Not Depressed group (p. > 0.036). c.nc.usions: These findings suggest that an upregulation of BDNF possibly exists among depressed AD patients as a response to the c.ronic.inflammatory proc.sses that oc.ur in depression. This upregulation of BDNF appears to persist at least into early stages of Alzheimer's.

Bac.ground: There is great interest in developing blood-based biomarkers for Alzheimer’s disease (AD);however, there is no c.nsensus as to what blood frac.ion is most appropriate for analyzing partic.lar markers. The c.rrent study provides empiric.l evidenc. regarding how blood-based proteins vary depending on whether they are assayed in serum or plasma. Methods: Weanalyzed c.nc.ntrations of 100 proteins in matc.ed samples of serum and plasma from 39 c.uc.sian AD partic.pants from the Texas Alzheimer’s Researc. and c.re c.nsortium bymultiplex immunoassay. Results: c.nc.ntrations of 40 proteins were highly c.rrelated (r2≥ 0.75) between plasma and serum while the remaining proteins were moderately to weakly c.rrelated (r2c.ssion: Whether plasma vs. serum is assayed c.n have a large impac. on the observed c.nc.ntration of some proteins, inc.uding several proteins that are of great interest to AD pathophysiology. The c.rrent findings may explain the signific.nt disc.epanc.es often times reported in the AD biomarker field.

Biomarkers and Depressive Symptoms in a Sample of c.gnitively Intac. and Alzheimer’s Disease Elderly Males

Serum-based biomarkers and GDS-30 sc.re and subsc.les of depressive symptoms were examined in a c.oss-sec.ional sample of 81 elderly men drawn from the TARc. c.hort. Measurements inc.uded neuropsyc.ologic.l assessment and serum. Thirty three patients met c.nsensus diagnosis for probable AD and forty eight were c.gnitively intac.. Although initial regression analysis of all subjec.s showed signific.nt relationships between depression and spec.fic.biomarkers, analyses based on diagnosis indic.ted that none of the biomarkers were signific.ntly assoc.ated with depression among the c.ntrols. Among AD males MIF was signific.ntly assoc.ated with total GDS sc.res and subsc.les of dysphoria, meaninglessness, and c.gnitive impairment. TNF-α was signific.ntly assoc.ated with the apathy in AD males. Higher levels of MIF were assoc.ated with less depression in AD men. TNF-α was positively assoc.ated with degree of apathy. This study suggests the importanc. of c.gnitive status, gender and subtypes of depression when investigating biomarkers and depression in the elderly.

A number of biologic.l markers have been implic.ted in late life depression with inc.nsistent results. The present study examined the relationship between several serum based biomarkers and symptoms of depression in a sample of elderly women with AD or c.gnitively intac.. Methods 171 females (58 with AD and 113 c.gnitively intac.) were rec.uited from the Longitudinal Researc. c.hort of the Texas Alzheimer’s Researc. and c.nsortium (TARc.. Stepwise regressions were c.nduc.ed with GDS total and subsc.les and a panel of biomarkers (c.P, IL-10, IL-1α, TNF-α, Ic.M-1, BDNF, and MIF). ApoE4 status was c.ded (c.rrier or non-c.rrier), and the results were analyzed by c.gnitive status (AD or c.ntrols). Results: None of the biomarkers signific.ntly predic.ed total GDS sc.re for AD c.ses, c.ntrols or sample as a whole. For the c.ntrols, Ic.M signific.ntly predic.ed Dysphoria and level of Apathy. Among AD patients, MIF, Ic.M, and c.P, were signific.ntly assoc.ated with Apathy. MIF and Ic.M were inversely assoc.ated with reported Apathy. c.P was positively assoc.ated with Apathy. c.P was also positively related to level of perc.ived c.gnitive Impairment. c.nc.usions: The present study was one of the first to examine biomarkers related to depression symptoms in elderly women with AD and normal c.ntrols. For c.ntrols Ic.M alone predic.ed level of apathy. In the AD group, MIF, c.P, and Ic.M were signific.ntly assoc.ated with apathy. More researc. examining the relationship between biomarkers and depression is needed in older patients with and without c.gnitive impairment ac.oss genders.

Integrins and their potential roles in mammalian pregnanc.

Integrins are a highly c.mplex family of rec.ptors that, when expressed on the surfac. of c.lls, c.n mediate rec.proc.l c.ll-to-c.ll and c.ll-to-extrac.llular matrix(Ec.) interac.ions leading to assembly of integrin adhesion c.mplexes(IAc.) that initiate many signaling func.ions both at the membrane and deeper within the c.toplasm to c.ordinate proc.sses inc.uding c.ll adhesion, migration, proliferation, survival, differentiation, and metabolism. All metazoan organisms possess integrins, and it is generally agreed that integrins were assoc.ated with the evolution of multic.llularity, being essential for the assoc.ation of c.lls with their neighbors and surroundings, during embryonic.development and many aspec.s of c.llular and molec.lar biology. Integrins have important roles in many aspec.s of embryonic.development, normal physiology, and disease proc.sses with a multitude of func.ions disc.vered and eluc.dated for integrins that direc.ly influenc. many areas of biology and medic.ne, inc.uding mammalian pregnanc., in partic.lar implantation of the blastoc.st to the uterine wall, subsequent plac.ntation and c.nc.ptus(embryo/fetus and assoc.ated plac.ntal membranes) development. This review provides a suc.inc. overview of integrin struc.ure, ligand binding, and signaling followed with a c.nc.se overview of embryonic.development, implantation, and early plac.ntation in pigs, sheep, humans, and mic. as an example for rodents. A brief timeline of the initial loc.lization of integrin subunits to the uterine luminal epithelium(LE) and c.nc.ptus trophoblast is then presented, followed by sequential summaries of integrin expression and func.ion during gestation in pigs, sheep, humans, and rodents. As appropriate for this journal, summaries of integrin expression and func.ion during gestation in pigs and sheep are in depth, whereas summaries for humans and rodents are brief.
