BACKGROUND The prevalence of cardiovascular diseases,especially heart failure,continues to rise *** heart failure,increasing levels of circulating atrial natriuretic peptide(ANP)and brain natriuretic peptide(BNP)are associated with a worsening of heart failure and a poor *** To test whether a high concentration of BNP would inhibit relaxation to *** Pulmonary arteries were dissected from disease-free areas of lung resection,as well as pulmonary artery rings of internal diameter 2.5–3.5 mm and 2 mm long,were *** artery rings were mounted in a multiwire myograph,and a basal tension of 1.61gf was *** equilibration for 60 min,rings were pre-constricted with 11.21μmol/L PGF2α(EC80),and concentration response curves were constructed to vasodilators by cumulative addition to the myograph *** Although both ANP and BNP were found to vasodilate the pulmonary vessels,ANP is more potent than ***50 of ANP and BNP were 8.96±0.21 and 7.54±0.18,respectively,and the maximum efficacy(Emax)for ANP and BNP was-2.03 gf and-0.24 gf,*** addition of BNP,the Emax of ANP reduced from-0.96gf to-0.675gf(P=0.28).CONCLUSION BNP could be acting as a partial agonist in small human pulmonary arteries,and inhibits relaxation to *** levels of circulating BNP could be responsible for the worsening of decompensated heart *** finding could also explain the disappointing results seen in clinical trials of ANP and BNP analogues for the treatment of heart failure.

Current available treatment options for erectile dysfunction （ED） are effective but not without failure and/or side effects. Although the development of phosphodiesterase type 5 （PDE5） inhibitors （i.e. sildenafil, tadalafil and vardenafil） has revolutionized the treatment of ED, these oral medications require on-demand access and are not as effective in treating ED related to diabetic, post-prostatectomy and severe veno-occlusive disease states. Improvement in the treatment of ED is dependent on understanding the regulation of human corporal smooth muscle tone and on the identification of relevant molecular targets. Future ED therapies might consider the application of molecular technologies such as gene therapy. As a potential therapeutic tool, gene therapy might provide an effective and specific means for altering intracavernous pressure ＂on demand＂ without affecting resting penile function. However, the safety of gene therapy remains a major hurdle to overcome before being accepted as a mainstream treatment for ED. Gene therapy aims to cure the underlying conditions in ED, including fibrosis. Furthermore, gene therapy might help prolong the efficacy of the PDE5 inhibitors by improving penile nitric oxide bioactivity. It is feasible to apply gene therapy to the penis because of its location and accessibility, low penile circulatory flow in the flaccid state and the presence of endothelial lined （lacunar） spaces. This review provides a brief insight of the current role of gene therapy in the management of ED.

Background: There is great interest in developing blood-based biomarkers for Alzheimer’s disease (AD);however, there is no consensus as to what blood fraction is most appropriate for analyzing particular markers. The current study provides empirical evidence regarding how blood-based proteins vary depending on whether they are assayed in serum or plasma. Methods: Weanalyzed concentrations of 100 proteins in matched samples of serum and plasma from 39 Caucasian AD participants from the Texas Alzheimer’s Research and Care Consortium bymultiplex immunoassay. Results: Concentrations of 40 proteins were highly correlated (r2≥ 0.75) between plasma and serum while the remaining proteins were moderately to weakly correlated (r2< 0.75). Discussion: Whether plasma vs. serum is assayed can have a large impact on the observed concentration of some proteins, including several proteins that are of great interest to AD pathophysiology. The current findings may explain the significant discrepancies often times reported in the AD biomarker field.

[1]往好了说,山谷就像牢房,禁锢人的想象,但是山谷、严冬、病弱凑在一起,就真在这氛围中营造出一个牢房,把人禁锢得寸步难行。道路确已扫清无障,至少一条路蜿蜒通向山上,但是来此求健康的人却无法离径而行,不能纵横原野,不能沿溪徜徉,不能在林中随兴漫步。行走是寻常乏味的。若体力允许,他可以向周围四五个方向前行,仅此而已,从来不能偏离为他设下的路线,每次都是从同一个角度看同一片树林雪野。几月下来,这本身已让人无法忍受,再加上层层叠叠的雪几乎毫无细节、色调单一,更难让人忍受。
