Bivariate whole-genome linkage scan for bone geometry and total body fat mass

To quantify the genetic correlations between total body fat mass （TBFM） and femoral neck geometric parameters （FNGPs） and, if pos- sible, to detect the specific genomic regions shared by them, bivariate genetic analysis and bivariate whole-genome linkage scan were carried out in a large Caucasian population. All the phenotypes studied were significantly controlled by genetic factors （P 〈 0.001） with the heritabilities ranging from 0.45 to 0.68. Significantly genetic correlations were found between TBFM and CSA （cross-section area）, W （sub-periosteal diameter）, Z （section modulus） and CT （cortical thickness） except between TBFM and BR （buckling ratio）. The peak bivariate LOD scores were 3.23 （20q12）, 2.47 （20p11）, 3.19 （6q27）, 1.68 （20p12）, and 2.47 （7q11） for the five pairs of TBFM and BR, CSA, CT, W, and Z in the entire sample, respectively. Gender-specific bivariate linkage evidences were also found for the five pairs. 6p25 had complete pleiotropic effects on the variations of TBFM ＆ Z in the female sub-population, and 6q27 and 17q11 had coincident link- ages for TBFM ＆ CSA and TBFM ＆ Z in the entire population. We identified moderate genetic correlations and several shared genomic regions between TBFM and FNGPs in a large Caucasian population.

Tunable photocycle kinetics of a hybrid bacteriorhodopsin/quantum dot system

The inclusion of inorganic nanoparticles in biological environments has led to the creation of hybrid nanosystems that are employed in a variety of applications. One such system includes quantum dots (QDs) coupled with the photoactive protein, bacteriorhodopsin (BR), which has been explored in developing enhanced photovoltaic devices. In this work, we have discovered that the kinetics of the BR photocycle can be manipulated using CdSe/CdS (core/shell) QDs. The photocycle lifetime of protein samples with varying QD amounts were monitored using time-resolved absorption spectroscopy. Concentration-dependent elongations of the bR and M state lifetimes were observed in the kinetic traces, thus suggesting that excitonic coupling occurs between BR and QDs. We propose that the pairing of BR with QDs has the potential to be utilized in protein-based computing applications, specifically for real-time holographic processors, which depend on the temporal dynamics of the bR and M photointermediates.

Stochastic walk-on-spheres(WOS)algorithms for solving the linearized Poisson-Boltzmann equation(LPBE)provide several attractive features not available in traditional deterministic solvers:Gaussian error bars can be computed easily,the algorithm is readily parallelized and requires minimal memory and multiple solvent environments can be accounted for by reweighting ***,previouslyreported computational times of these Monte Carlo methods were not competitive with existing deterministic numerical *** present paper demonstrates a series of numerical optimizations that collectively make the computational time of these Monte Carlo LPBE solvers competitive with deterministic *** optimization techniques used are to ensure that each atom’s contribution to the variance of the electrostatic solvation free energy is the same,to optimize the bias-generating parameters in the algorithm and to use an epsilon-approximate rather than exact nearest-neighbor search when determining the size of the next step in the Brownian motion when outside the molecule.

目的呼吸道上皮细胞在固有免疫和炎症反应中都扮演重要角色。本文旨在研究强效吸入型糖皮质激素丙酸氟替卡松(FP)对双链RNA(dsRNA,Toll样受体3的配体)刺激的呼吸道粘膜上皮细胞BEAS-2B产生的某些固有免疫相关蛋白和炎性细胞因子的作用。方法FP(10-7mol.L-1)作用于BEAS-2B 2 h后,加入dsRNA 25 mg.L-1继续培养18 h,提取细胞RNA和收集细胞培养上清液。实时定量PCR测定炎性细胞因子IL-8、gm-CSF、IL-1β和固有免疫蛋白血清淀粉样蛋白(SAA)、补体B因子、分泌性白细胞蛋白酶抑制物(SLPI)的mRNA含量;ELISA分析上清液中IL-8、gm-CSF、SAA蛋白水平。结果BEAS-2B细胞在dsRNA刺激后,IL-8、gm-CSF、IL-1β、SAA、补体B因子、SLPI的mRNA均增高。FP预处理的细胞,在dsRNA刺激后,SAA、补体B因子、SLPI的mRNA水平不降低,IL-8、gm-CSF、IL-1β的mRNA水平明显降低。IL-8、gm-CSF、SAA的ELISA分析证实了mRNA的试验结果。结论FP在强效抗炎的同时,并不损伤呼吸道上皮局部的固有免疫。

Evaluation of cardiolipin nanodisks as lipid replacement therapy for Barth syndrome

Barth syndrome（BTHS） is a mitochondrial disorder characterized by cardiomyopathy and skeletal muscle weakness. Disease results from mutations in the tafazzin（TAZ） gene, encoding a phospholipid transacylase. Defective tafazzin activity results in an aberrant cardiolipin（CL） profile. The feasibility of restoring the intracellular CL profile was tested by in vivo administration of exogenous CL in nanodisk（ND） delivery particles. Ninety mg/kg CL（as ND）was administered to doxycycline-inducible taz shRNA knockdown（KD） mice once a week. After 10 weeks of CLND treatment, the mice were sacrificed and tissues harvested. Liquid chromatography-mass spectrometry of extracted lipids revealed that CL-ND administration failed to alter the CL profile of taz KD or WT mice. Thus, although CL-ND were previously shown to be an effective means of delivering CL to cultured cells, this effect does not extend to an in vivo setting. We conclude that CL-ND administration is not a suitable therapy option for BTHS.

Twelve weeks of dance exergaming in overweight and obese adolescent girls:Transfer effects on physical activity,screen time,and self-efficacy

Background: Given the low levels of physical activity(PA) among adolescent girls in the US, there is a need to identify tools to motivate increased PA. Although there is limited evidence that adolescents transfer PA from one context to another, exergames(i.e., video games that require gross motor activity) may act as a gateway to promote overall PA outside game play. The purpose of this study was to examine potential transfer effects(i.e., influences on external behaviors and psychological constructs) of a 12-week exergaming intervention on adolescent girls' PA, screen time,and self-efficacy toward PA, as well as the intrinsic motivation of ***: Participants were 37 girls aged 14–18 years(65% African American, 35% white) who were overweight or obese(body mass index ≥ 85 th percentile) and were recruited from the community via school, physicians, news media, and social media websites. Adolescents were randomly assigned to a 12-week group exergaming intervention(thirty-six 60 min sessions of group-based dance exergaming in a research laboratory using Kinect for Xbox360(Microsoft Corporation, Redmond, WA, USA)) or to a no-treatment control group. Outcome variables included objectively measured PA(total) and self-reported leisure-time PA(discretionary time only) 1 week before vs. 1 week after the intervention; selected type and intensity of PA when placed in a gym setting for 30 min("cardio free choice"); screen time; self-efficacy toward PA; and intrinsic motivation toward ***: Attendance at the exergaming sessions was high(80%). Compared with the control group, the intervention group self-reported an increase in PA(p = 0.035) and fewer hours watching television or videos(p = 0.01) after the intervention, but there were no significant differences in sedentary, light, moderate, or vigorous PA measured by accelerometry. Dear Editor,Beige adipocytes,which increase energy expenditure by dissipating energy as heat,have gained attention as a therapeutic target for combating obesity.1 Adipocytes express many types of G-protein-coupled receptors(GPCRs),each of which has a unique preference for the Gs,Gi,Gq,and G12 *** the function of Gs-coupledβ-adrenergic receptors in beige adipocyte induction is well established,2 little is known about the function of G12-coupled GPCRs beyond its suppressive roles in white adipocyte maturation.3 In this study,we generated transgenic mice conditionally expressing a G12-coupled designer GPCR using a Cre-loxP system and investigated the potential effects of G12 signaling on adipocyte biology.

Fluorine-thiol displacement probes for acetaminophen's hepatotoxicity

Chemicals possessing reactive electrophiles can denature innate proteins leading to undesired toxicity,and the overdose-induced liver injury by drugs containing electrophiles has been one of the major causes of non-approval and withdraw by the US Food and Drug Administration(FDA).Elucidating the associated proteins could guide the future development of therapeutics to circumvent these drugs’toxicities,but was largely limited by the current probing tools due to the steric hindrance of chemical tags including the common“click chemistry”*** the widely used non-steroidal anti-inflammatory drug acetaminophen(APAP)as an example,we hereby designed and synthesized an APAP analogue using fluorine as a steric-free *** toxicity studies indicated our analogue has similar activity to the parent *** analogue was applied to the mouse hepatocellular proteome together with the corresponding desthiobiotinSH probe for subsequent fluorine-thiol displacement reactions(FTDRs).This set of probes has enabled the labeling and pull-down of hepatocellular target proteins of the APAP metabolite as validated by Western *** preliminary validation results supported the interaction of APAP with the thioredoxin protein,which is an important redox protein for normal liver *** results demonstrated that our probes confer minimal steric perturbation and mimic the compounds of interest,allowing for global profiling of interacting *** fluorine-thiol displacement probing system could emerge as a powerful tool to enable the investigation of drug-protein interactions in complex biological environments.
