Benzo[a]pyrene(BaP),a ubiquitous pollutant,raises environmental health concerns due to induction of bone toxicity in the unexposed *** of F0 ancestor medaka(Oryzias latipes)to 1μg/L BaP for 21 days causes reduced vertebral bone thickness in the unexposed F3 male *** reveal the inherited modifications,osteoblast(OB)abundance and molecular signaling pathways of transgenerational BaP-induced bone thinning were *** analysis showed a reduction in OB *** of the miRNA and mRNA transcriptomes revealed the dysregulation of Wnt signaling(frzb/ola-miR-1–3p,sfrp5/ola-miR-96–5p/miR-455–5p)and bone morphogenetic protein(Bmp)signaling(bmp3/ola-miR-96–5p/miR-181b-5p/miR-199a-5p/miR-205–5p/miR-455–5p).Both pathways are major indicators of impaired bone formation,while the altered Rank signaling in osteoclasts(c-fos/miR-205–5p)suggests a potentially augmented bone ***,a typical BaP-responsive pathway,the Nrf2-mediated oxidative stress response(gst/ola-miR-181b-5p/miR-199a-5p/miR-205),was also ***,mRNA levels of epigenetic modification enzymes(e.g.,hdac6,hdac7,kdm5b)were found *** findings indicated that epigenetic factors(e.g.,miRNAs,histone modifications)may directly regulate the expression of genes associated with transgenerational BaP bone toxicity and warrants further *** identified candidate genes and miRNAs may serve as potential biomarkers for BaP-induced bone disease and as indicators of historic exposures in wild fish for conservation purposes.

Near-infrared Emissive 1,2-Dioxetane-based Chemiluminescent Probes

Chemiluminescence, a phenomenon emitting light from chemical reactions rather than photon absorption, has gained significant interest for applications in bioimaging and biosensing due to its high sensitivity and low background interference. Now there is a growing interest in near-infrared (NIR) chemiluminescent probes for improved tissue penetration and reduced autofluorescence. This review summarizes NIR emissive chemiluminescent probes based on 1,2-dioxetane and discusses their chemical structures and applications. Structure modification strategies for red-shifting wavelength and enhancing brightness include incorporating electron-withdrawing groups, designing chemiluminophore-fluorophore cassettes, and exploring alternative chemiluminescent scaffolds. This review aims to inspire the exploration of NIR chemiluminescent probes in disease detection and treatment.

A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATCC *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

Experimental Study on Elevated Fires in a Ceiling Vented Compartment

The impacts of elevation on fires in a ceiling vented compartment were investigated experimentally. The flame behavior of elevated fires was recorded. Various parameters including the fuel mass loss rate, the light extinction coefficient, the oxygen concentration and the gas temperature were measured. Results indicated that the variations of the flame behavior were consistent with that of the fuel mass loss rate. The fire location significantly impacted the light extinction coefficient, the oxygen concentration and the gas temperature, which all showed distinct stratification phenomena. For a higher elevated f＇we, the average fuel loss rate and the overall light extinction co- efficient were smaller, the oxygen concentration was higher and the gas temperature was lower. In addition, the smoke descending was slower. From the perspective of those parameters the fire was less hazardous if the fire was elevated higher, which was totally different from the elevated fires in closed compartments.

Spatial memory enhances the evacuation efficiency of virtual pedestrians under poor visibility condition

Spatial memory is a critical navigation support tool for disoriented evacuees during evacuation under adverse environ-mental conditions such as dark or smoky conditions. Owing to the complexity of memory, it is challenging to understand the effect of spatial memory on pedestrian evacuation quantitatively. In this study, we propose a simple method to quan- titatively represent the evacuee＇s spatial memory about the emergency exit, model the evacuation of pedestrians under the guidance of the spatial memory, and investigate the effect of the evacuee＇s spatial memory on the evacuation from theoretical and physical perspectives. The result shows that （i） a good memory can significantly assist the evacuation of pedestrians under poor visibility conditions, and the evacuation can always succeed when the degree of the memory exceeds a threshold （φ〉 0.5）; （ii） the effect of memory is superior to that of ＂follow-the-crowd＂ under the same environmental conditions; （iii） in the case of multiple exits, the difference in the degree of the memory between evacuees has a significant effect （the greater the difference, the faster the evacuation） for the evacuation under poor visibility conditions. Our study provides a new quantitative insight into the effect of spatial memory on crowd evacuation under poor visibility conditions.

Development of an agent-based indoor evacuation model for local fire risks analysis

Evidence suggests that in the event of a fire accident, a certain number of building occupants escape through smoke-filled environments. Consequently, evaluating the corresponding evacuation performance under such life-threatening conditions is important for advancing fire safety analyses. This study aimed to develop a fire-integrated evacuation model to consider the effects of spreading fire hazards (i.e., radiation, temperature, toxic gas, visibility) on evacuees in a room fire evacuation scenario. Furthermore, a novel quantitative approach was introduced to evaluate evacuees’ local fire risks and stress levels according to their egress paths. The escape characteristics at various stages of fire development were studied as well. The results demonstrate that evacuation performance varies considerably depending on the severity of evacuees’ confronted fire hazard conditions, which emphasizes the importance of minimizing the pre-evacuation time in fire evacuation emergencies.

Study on single-group dynamics in evacuation considering pre-defined and undeclared leadership

Grouping is a common phenomenon that occurs *** leader-follower relationship inside groups has often been qualitatively characterized in previous models using simple ***,a general method is lacking to quantitatively explain leadership in an evacuating *** understand the evolution of single-group dynamics throughout an evacuation,we developed an extended social force model integrated with a group force.A series of single-group evacuations from a room were *** information-theoretic method,transfer entropy(TE),was applied to detect predefined and undeclared leadership among *** results showed that the predefined leader was correctly detected by TE,suggesting its capability in measuring leadership based on time series of evacuees’movement information(e.g.,velocity and acceleration).When evacuees were grouped together,TE was higher than when they were *** presented a monotonically increasing cumulative influence curve over the investigated period,whereas followers showed a diminishing *** found that leadership emergence correlated with evacuees’spatial *** individual located in the foremost part of the group was most likely to become a leader of those in the rear,which concurred with the experimental *** observed how a large group split into smaller ones with undeclared leadership during *** observations were quantitatively verified by TE *** study provides novel insights into quantifying leadership and understanding single-group dynamics during evacuations.

The prediction of exit choice using cumulative prospect value

Exit choice is essential for pedestrian safety and evacuation efficiency during the context of an emergency. Cumulative prospect theory is a widespread realistic decision-making theory that can transform choice outcomes and probabilities into subjective terms and integrate them as a parameter of cumulative prospect value (CPV), which determines the decision. The main contribution of this paper is the use of the CPV to predict exit choice. Furthermore, the different decision-making rules including Max, exponential(Expo), and Ratio (i.e., the three variable choice functions) were summarized and examined. This study used a confusion matrix to compare the predicted results with experimental data. Consequently, the parameters of Accuracy and F1-score showed that the predictions from Max and Expo were significantly more realistic while the results from Ratio were much more robust.
