Genome mapping has promised much to tree fruit breeding during the last 10 ***,one of the greatest challenges remaining to tree fruit geneticists is the translation of trait loci and whole genome sequences into diagnostic genetic markers that are efficient and cost-effective for use by breeders,who must select genetically optimal parents and subsequently select genetically superior individuals among their *** take this translational step,we designed the apple International RosBREED SNP Consortium OpenArray v1.0(IRSCOA v1.0)assay using a set of 128 apple single nucleotide polymorphisms(SNPs)linked to fruit quality and pest and disease resistance trait *** Thermo Fisher Scientific OpenArray®technology enables multiplexed screening of SNP markers using a real-time PCR instrument with fluorescent probe-based Taqman®*** validated the apple IRSCOA v1.0 multitrait assay by screening 240 phenotyped individuals from the Plant&Food Research apple cultivar breeding *** set of individuals comprised commercial and heritage cultivars,elite selections,and families segregating for traits of importance to *** total,33 SNP markers of the IRSCOA v1.0 were validated for use in marker-assisted selection(MAS)for the scab resistances Rvi2/Vh2,Rvi4/Vh4,Rvi6/Vf,fire blight resistance MR5/RLP1,powdery mildew resistance Pl2,fruit firmness,skin colour,flavour intensity,and *** availability of this set of validated trait-associated SNP markers,which can be used individually on multiple genotyping platforms available to various apple breeding programmes or re-designed using the flanking sequences,represents a large translational genetics step from genomics to crop improvement of apple.

knowledge of the genotypes for the self-incompatibility locus(S-locus)in apple varieties and in genotypes being used as parents is critical for breeding and commercial *** present a high-throughput set of molecular markers for the identification of 13 common S-RNase alleles(S_(1),S_(2),S_(3),S_(5),S_(7),S_(8),S_(9),S_(10),S_(20),S_(2)3,S_(24),S_(25)and S_(28).This set is composed of seven allele-specific quantitative PCR-based High-Resolution Melting assays and four multi-allelic SSR *** of these markers was performed using 86 apple accessions,including cultivars with known S-genotypes and recent commercial varieties arising from the Plant&Food Research(PFR)cultivar breeding *** also characterized the S-genotypes of 183 genotypes representing some of the most valuable parents within PFR’s cultivar breeding *** results of this work demonstrate the practical usefulness of this marker set to provide accurate cross-compatibility information to optimise choice of pollenisers in commercial apple orchard design,and to identify compatible parents and guide parental selection when executing apple breeding programmes,to optimise fruit crop yield and quality.

Experimental investigation on slip conditions during thread rolling with flat dies

The process design of flat die rolling operations can be a tedious and costly process due to the challenges posed by small process *** this work,an effort is made to closely investigate and understand the process limits caused by excessive slippage of the *** measurements are carried out to capture the characteristics of excessive slippage and the influence of the stroke rate and the tribological system on this process *** differences in slippage were observed,depending on the tribological *** systems with polymer based lubricants showed excessive slippage at low stroke rates,systems using oil or no lubricant endured higher stroke rates before process limits were *** optical measurements revealed three major forms of slippage:successful rolling without slippage,failure due to excessive slippage at process start,and failure due to excessive slippage at around one workpiece rotation.

Regression analysis of squeezing-induced hybrid nanofluid flow in Darcy-Forchheimer porous medium

This article presents a mathematical model addressing a scenario involving a hybrid nanofluid flow between two infinite parallel plates. One plate remains stationary, while the other moves downward at a squeezing velocity. The space between these plates contains a Darcy-Forchheimer porous medium. A mixture of water-based fluid with gold(Au) and silicon dioxide(Si O2) nanoparticles is formulated. In contrast to the conventional Fourier's heat flux equation, this study employs the Cattaneo-Christov heat flux equation. A uniform magnetic field is applied perpendicular to the flow direction, invoking magnetohydrodynamic(MHD) effects. Further, the model accounts for Joule heating, which is the heat generated when an electric current passes through the fluid. The problem is solved via NDSolve in MATHEMATICA. Numerical and statistical analyses are conducted to provide insights into the behavior of the nanomaterials between the parallel plates with respect to the flow, energy transport, and skin *** findings of this study have potential applications in enhancing cooling systems and optimizing thermal management strategies. It is observed that the squeezing motion generates additional pressure gradients within the fluid, which enhances the flow rate but reduces the frictional drag. Consequently, the fluid is pushed more vigorously between the plates, increasing the flow velocity. As the fluid experiences higher flow rates due to the increased squeezing effect, it spends less time in the region between the plates. The thermal relaxation, however, abruptly changes the temperature, leading to a decrease in the temperature fluctuations.

Thermal rectification is a promising way to manipulate the heat flow,in which thermal phonons are spectrally and collectively *** phononic devices are mostly relying on monochromatic phonons,in this work we propose a phononic rectifier based on the carbon schwarzite host–guest *** using molecular dynamic simulations,we demonstrate that the phononic rectification only happens at a specific frequency of the hybridized mode for the host–guest system,due to its strong confinement ***,a significant rectification efficiency,~134%,is observed,which is larger than most of the previously observed *** study of length and temperature effects on the phononic rectification shows that the monochromaticity and frequency of the rectified thermal phonons depend on the intrinsic anharmonicity of the host–guest system and that the on-center rattling configuration with weak anharmonicity is *** study provides a new perspective on the rectification of thermal phonons,which would be important for controlling monochromatic thermal phonons in phononic devices.

A computational platform for considering the effects of aerodynamic and seismic load combination for utility scale horizontal axis wind turbines

The wide deployment of wind turbines in locations with high seismic hazard has led engineers to take into account a more comprehensive seismic design of such structures. Turbine specific guidelines usually use simplified methods and consider many assumptions to combine seismic demand with the other operational loads effecting the design of these structures. As the turbines increase in size and capacity, the interaction between seismic loads and aerodynamic loads becomes even more important. In response to the need for a computational tool that can perform coupled simulations of wind and seismic loads, a seismic module is developed for the FAST code and described in this research. This platform allows engineers working in this industry to directly consider interaction between seismic and other environmental loads for turbines. This paper details the practical application and theory of this platform and provides examples for the use of different capabilities. The platform is then used to show the suitable earthquake and operational load combination with the implicit consideration of aerodynamic damping by estimating appropriate load factors.

Effects of P301L-TAU on post-translational modifications of microtubules in human iPSC-derived cortical neurons and TAU transgenic mice

BACkGROUND Transfusion transmissible infections(TTIs)are illnesses spread through contaminated blood or blood *** India,screening for TTIs such as hepatitis B virus(HBV),hepatitis C virus(HCV),human immunodeficiency virus(HIV)-I/II,malaria,and syphilis is mandatory before blood ***,HCV,HBV,and HIV are the leading viruses causing mortality,affecting millions of people globally,including those with co-infections of HIV/HCV and HIV/*** highlight the impact of TTIs on life expectancy and health risks,such as liver cirrhosis,cancer,and other diseases in individuals with chronic ***,millions of blood donations take place annually,emphasizing the importance of maintaining blood *** To study the prevalence of TTIs,viz.,HBV,HCV,HIV I/II,syphilis,and malaria parasite(MP),among different blood donor *** The study assessed the prevalence of TTIs among different blood donor groups in Delhi,*** included total donors,in-house donors,total camp donors,institutional camp donors,and community camp *** for HIV,HBV,and HCV were done using enzyme-linked immunosorbent assay,while syphilis was tested with rapid plasma reagins and MP rapid card *** prevalence of HBV,HCV,HIV,and syphilis,expressed as *** in infection rates between the groups were analyzed usingχ²tests and P-values(less than 0.05).RESULTS The study evaluated TTIs among 42158 blood donors in *** overall cumulative frequency of TTIs in total blood donors was 2.071%,and the frequencies of HBV,HCV,HIV-I/II,venereal disease research laboratory,and MP were 1.048%,0.425%,0.221%,0.377%,and 0.0024%,***-house donors,representing 37656 donors,had the highest transfusion transmissible infection(TTI)prevalence at 2.167%.Among total camp donors(4502 donors),TTIs were identified in 1.266%of donors,while community camp donors(2439 donors)exhibited a prevalence of 1.558%.Institutional camp donors(2063 donors)had the l

Cell transplantation to replace retinal ganglion cells faces challenges-the Switchboard Dilemma

How coherence is governing diffuson heat transfer in amorphous solids

Thermal transport in amorphous materials has remained one of the fundamental questions in solid state physics while involving a very large field of *** a heat conduction theory incorporating coherence,we demonstrate that the strong phase correlation between local and non-propagating modes,commonly named diffusons in the terminology of amorphous systems,triggers the conduction of *** treating the thermal vibrations as collective excitations,the significant contribution of diffusons,predominantly relying on coherence,further reveals interesting temperature and length dependences of thermal *** propagation length of diffuson clusters is found to reach the micron,overpassing the one of *** explored wavelike behavior of diffusons uncovers the unsolved physical picture of mode correlation in prevailing models and further provides an interpretation of their ability to transport *** work introduces a framework for understanding thermal vibrations and transport in amorphous materials,as well as an unexpected insight into the wave nature of thermal vibrations.
