Elesclomol (ELC) is an anticancer drug inducing mitochondria cytotoxicity through reactive oxygen ***,for the first time,we encapsulate the poorly water soluble ELC in monoolein-based cubosomes stabilized with Pluronic *** uptake and nanocarrier accumulation close to the mitochondria with sub-micrometer distance is identified via three-dimensional (3D) confocal microscopy and edge-to-edge compartment *** monitor the therapeutic effect of the ELC nanocarrier,we apply for the first time,label-free time-lapse multi-photon fluorescence lifetime imaging microscopy (mP-FLIm) to track NAD(P)H cofactors with sub-cellular resolution on live cells exposed to an anticancer *** in vitro cytotoxicity is verified when loading the pre-complexed ELC with copper (ELC-Cu).Importantly,for equivalent copper concentration,cubosomes loaded with ELC-Cu show higher cytotoxicity compared to the free *** novel nanocarrier shows promising features for systemic ELC-Cu administration,and furthermore we establish the mP-FLIm technique for the assessment of anticancer drug delivery systems.

There have been many chapters written about macrophage *** chapters generally focus on the role of macrophages in orchestrating immune responses by highlighting the T-cell-derived cytokines that shape these polarizing *** bias toward immunity is understandable,given the importance of macrophages to host ***,macrophages are ubiquitous and are involved in many different cellular processes,and describing them as immune cells is undoubtedly an *** disregards their important roles in development,tissue remodeling,wound healing,angiogenesis,and metabolism,to name just a few *** this chapter,we propose that macrophages function as transducers in the *** to Wikipedia,WA transducer is a device that converts energy from one form to another.^The word transducer is a term used to describe both the"sensor,which can interpret a wide range of energy forms,and the"actuator,which can switch voltages or currents to affect the *** are able to sense a seemingly endless variety of inputs from their environment and transduce these inputs into a variety of different response ***,rather than functioning as immune cells,they should be considered more broadly as cellular transducers that interpret microenvironmental changes and actuate vital tissue *** this chapter,we will describe some of the sensory stimuli that macrophages perceive and the responses they make to these stimuli to achieve their prime directive,which is the maintenance of homeostasis.

N-acetylatedα-synuclein(αSyn)has long been established as an intrinsically disordered protein associated with a dysfunctional role in Parkinson’s *** recent years,a physiologically relevant,higher order conformation has been identified as a helical tetramer that is tailored by buried hydrophobic interactions and is distinctively aggregation *** canonical mechanism by which the tetramer assembles remains *** novel biochemical approaches,computational methods,pioneering purification platforms,and powerful imaging techniques continue to develop,puzzling information that once sparked debate as to the veracity of the tetramer has now shed light upon this new counterpart inαSyn *** magnetic resonance and computational studies on multimericαSyn structure have revealed that the protein folding propensity is controlled by small energy barriers that enable large scale ***,familial mutations ablate tetramerization and reconfigure polymorphic *** this review,we will discuss the dynamic landscape ofαSyn quaternary structure with a focus on the tetrameric conformation.

recent developments integrating micromechanics and neural networks offer promising paths for rapid predictions of the response of heterogeneous materials with similar accuracy as direct numerical *** deep material network is one such approaches,featuring a multi-layer network and micromechanics building blocks trained on anisotropic linear elastic *** trained,the network acts as a reduced-order model,which can extrapolate the material’s behavior to more general constitutive laws,including nonlinear behaviors,without the need to be ***,current training methods initialize network parameters randomly,incurring inevitable training and calibration ***,we introduce a way to visualize the network parameters as an analogous unit cell and use this visualization to“quilt”patches of shallower networks to initialize deeper networks for a recursive training *** result is an improvement in the accuracy and calibration performance of the network and an intuitive visual representation of the network for better explainability.

Isoperimetric problems consist in minimizing or maximizing a cost functional subject to an integral *** this work, we present two fractional isoperimetric problems where the Lagrangian depends on a combined Caputo derivative of variable fractional order and we present a new variational problem subject to a holonomic constraint. We establish necessary optimality conditions in order to determine the minimizers of the fractional problems. The terminal point in the cost integral,as well as the terminal state, are considered to be free, and we obtain corresponding natural boundary conditions.

In this paper,we address the long-term generation and transmission expansion planning for power systems of regions with very high solar *** target the power systems that currently rely mainly on thermal generators and that aim to adopt high shares of renewable *** propose a stochastic programming model with expansion alternatives including transmission lines,solar power plants(photovoltaic and concentrated solar),wind farms,energy storage,and flexible combined cycle gas *** model represents the longterm uncertainty to characterize the demand growth,and the short-term uncertainty to characterize daily solar,wind,and demand *** use the Saudi Arabian power system to illustrate the functioning of the proposed model for several cases with different renewable integration *** results show that a strong dependence on solar power for high shares of renewable sources requires high generation capacity and storage to meet the night demand.

Nervous system emerges from complex signaling interactions where extrinsic (neurotransmitters and trophic factors, among others) and intrinsic factors (transcription factors) interplay in the developing tissue to control gene activity promoting chronic changes in cell genesis, migration, differentiation and death. The retinal microenvironment is regulated by a broad variety of chemicals, including endocannabinoids and nucleotides that modulate embryonic progenitor-neuron-müller glia signaling in very early developing or pathophysiological conditions.

microbial-induced calcium carbonate precipitation is a promising technology for self-healing concrete due to its capability to seal *** main goal of this study was to evaluate the effects of adding hydrogelencapsulated bacteria on the compressive strength and the self-healing efficiency of *** achieve this objective,12 sets of mortar samples were prepared,including three different mineral precursors(magnesium acetate,calcium lactate,and sodium lactate),at two concentrations(67.76 and 75.00 mm/L),and under two different biological conditions(with and without bacteria).In addition,a set of plain mortar samples was prepared to serve as a *** each sample set,three mortar cubes and three beams were prepared and subjected to compression and flexural strength *** the compression tests,it was found that the sample containing calcium lactate along with yeast extract and bacteria displayed the best *** for the flexural tests,once cracked,the beams were subjected to 28 d of wet/dry cycles(16 h of water immersion and 8 h of drying),where the bottom crack width was monitored(at 0,3,7,14,28 d of wet/dry cycles).Once the sample with the highest healing efficiency was identified(the one containing calcium lactate and hydrogel-encapsulated bacteria),the study was scaled up to concrete *** sets of concrete cylinders(consisting of three control samples and three samples with bacteria along with calcium lactate)were tested under compression in order to evaluate the effect of the bacteria-precursor combination on the concrete mechanical *** samples that yielded the greatest compressive strength were the ones containing calcium lactate and bacteria,displaying an improvement of 17%as compared to the control ***,a flexural strength recovery analysis was performed on the concrete specimens revealing that the control showed better flexural strength recovery than the bacteriacontaining variant(41.5%vs.26.1%)after
