Immune and non-immune responses to hepatitis C virus infection

The host innate and adaptive immune systems are involved in nearly every step of hepatitis C virus(HCV) infection. In patients,the outcome is determined by a series of complex host-virus interactions,whether it is a natural infection or results from clinical intervention. Strong and persistent CD8+ and CD4+ T-cell responses are critical in HCV clearance,as well as cytokineinduced factors that can directly inhibit virus replication. Newly available direct-acting antivirals(DAAs) are very effective in viral clearance in patients. DAA treatment may further result in the down-regulation of programmed death-1,leading to rapid restoration of HCV-specific CD8+ T cell functions. In this review,we focus on recent studies that address the host responses critical for viral clearance and disease resolution. Additional discussion is devoted to the prophylactic vaccine development as well as to current efforts aimed at understanding the host innate responses against HCV infection. Current theories on how the ubiquitin system and interferon-stimulated genes may affect HCV replication are also discussed.

Measuring ammonia(NH_(3))volatilization from urea-fertilized soils is crucial for evaluation of practices that reduce gaseous nitrogen(N)losses in *** small area of chambers used for NH_(3)volatilization measurements compared with the size of field plots may cause significant errors if inadequate sampling strategies are *** aims were:i)to investigate the effect of using multiple open chambers on the variability in the measurement of NH_(3)volatilization in urea-amended field plots and ii)to define the critical period of NH_(3)-N losses during which the concentration of sampling effort is capable of reducing *** use of only one chamber covering 0.015%of the plot(51.84 m^(2))generates a value of NH_(3)-N loss within an expected margin of error of 30%around the true *** reduce the error margin by half(15%),3–7 chambers were required with a mean of 5 chambers per *** the sampling efforts in the first two weeks after urea application,which is usually the most critical period of N losses and associated errors,represents an efficient strategy to lessen uncertainty in the measurements of NH_(3)*** strategy enhances the power of detection of NH_(3)-N loss abatement in field experiments using chambers.

Interactions between a Root Knot Nematode (Meloidogyne exigua) and Arbuscular Mycorrhizae in Coffee Plant Development (Coffea arabica)

This paper focuses on parasitic root knot nematodes (Meloidogyne exigua) and how to decrease their pathogenic effect on coffee plants (Coffea arabica), by examining the behaviour of and the interactions between nematodes, coffee plant and arbuscular mycorrhizae (AM). The experiment was carried out at the seedling stage, with six (6) treatments (plants with M. exigua, plants with arbuscular mycorrhizae, plants with both organisms, and the same time, first mycorrhizae plants, then nematodes were inoculated and vice versa). After 5 months the measured variables were: dry biomass (roots and shoot), nematode knots caused by M. exigua in root, nematode juvenile (j2) found in 100.0 g of soil, and mycorrhizal percentage. Plant nutrients (P and N) contents were analysed. Significant differences were found in all the variables, but concentration N content in plants. Plants with mycorrhizae and plants with mycorrhizae and then inoculated with nematodes have the same behaviour. Control plants and plants with nematode and then inoculated with mycorrhizae behave similarly. It is thought that arbuscular mycorrhizae are formed before the nematode infestation, allowing coffee plants to regain the energy lost by the parasitic interaction. AM may help coffee plants with lignifications of the plant cell wall cuticle. As the cuticle thickens it is more difficult for nematodes to penetrate and enter into plant roots. Therefore, arbuscular mycorrhizae help coffee plants to uptake and transport nutrients, improving its nutritional status and stabilizing nematode attacks. It is suggested that symbiotic interactions help neutralize parasitic interactions.

Fatal Outcome after Delayed Pipeline Embolization Device Migration for the Treatment of a Giant Superior Cerebellar Artery Aneurysm: Technical Note for Complication Avoidance

Purpose: Flow diversion is a relatively novel technique to treat some intracranial aneurysms. With new techniques, unusual complications can occur. We described a case of Pipeline Embolization Device (PED) migration, strategy for its recognition, and a technical point to prevent its occurrence. Publication of these rare events is important to make physicians aware of potential complications. Methods: A patient with a previously coiled giant superior cerebellar aneurysm presented with brainstem compression symptoms. Imaging verified progressive aneurysm growth. A decision was made to treat the aneurysm with PED. Results: Development of new neurologic symptoms prompted a computed tomography that showed hydrocephalus. An angiogram also showed recanalization of the aneurysm secondary to upward migration/retraction of the flow diverter. A ventriculo-peritoneal shunt was implemented and planning for placement of a second flow diverter was made. Unfortunately, the patient expired while waiting for the endovacular intervention. Conclusions: PED migration may occur even after correct placement of the device. Early recognition of this complication is essential. When clinical changes occur, it is paramount that this uncommon, but potentially deadly, complication is suspected. Appropriate sizing and deployment technique are important for the long-term stability of the device.

Occurrence of non-pathogenic nitrogen-fixing bacteria living within the plant tissue attracts considerable attention. The A15 strain was isolated from rice paddies in China (You and Zhou 1989, Can. j. Microbiol. 35:403-408). It can fix nitrogen under microaerobic conditions at the free-living state and it colonize rice endophytically. A15 is used as a plant growth promoting inoculant of rice in China.

Effects of aquatic exercise on depression and anxiety in ischemic stroke subjects

Background: Stroke ranks third and is the leading cause of permanent disability in western countries. Furthermore there are no treatments for the psychosocial effects of this pathology. Purpose: Analyse the effect of an aquatic exercise program in depression and trace and state anxiety in subjects who suffered an ischemic stroke. Methods: Two groups were analyzed: experimental group (EG) n = 15, 50.3 ± 9.1 years;control group (CG) n = 13, 52.5 ± 7.7 years. EG underwent a 12-week aquatic exercise program. Both groups were evaluated in pre and post- treatment using Beck Depression Inventory (BDI) and Trace State Anxiety Inventory (IDATE). Wilcoxon signed-rank and Mann-Whitney tests were used to compare moments and groups, respec- tively. Results: The BDI scores of aquatic activities were: pre-treatment, 17.4 ± 7.7 and 16.9 ± 8.6 for the EG and CG, respectively;post-treat- ment, 13.2 + 7.1 and 16.4 + 7.9 for the EG and CG, respectively. The IDATE scores for anxiety trace in strength training were: pre-treatment, 43.2 + 12.5 and 42.9 + 12.2 for the EG and CG, respec- tively;post-treatment, 39.7 + 7.1 and 42.6 + 12.1 for the EG and CG, respectively. The IDATE scores for anxiety state in strength training were: pre-treatment, 46.9 + 7.6 and 47.4 + 8.1 for the EG and CG, respectively;post-treatment, 44.4 + 7.9 and 47.5 + 8.0 for the EG and CG, respectively. Significant differences were found in pre and post-treatment values in the EG and between groups in the depression and trace and state anxiety levels in post-treatment (p Conclusions: Aquatic physical activity contributes to an improvement of the levels of depression and anxiety in people who suffered a stroke.

End stage-renal-disease (esRD) is associated with dyslipidemia and premature atherosclerosis. The study evaluates the effect of hemodialysis (HD) on HDL-remodeling between pre- and post-HD. Was conducted a cross-sectional study with 50 esRD male patients, undergoing HD at Ana Neri Hospital, Salvador, Brazil. All individuals were on HD for at least 3 months, into a three sessions protocol for 3 - 4 hours per week, with a polysulfone low-flow basic-dialyzing-membrane and unfractionated-heparin. HDL Phospholipid-incorporation was measured by 14C-PL-scintillation-counting, expressed as %14C-PL/mL/hour. Paraoxonase (PON-1) activity was measured by spectrophotometry using paraoxon as substrate. Cardiovascular risk ratios and atherogenic index of plasma were calculated. Total cholesterol, HDL-C and non-HDL-C increased at post-HD on all age groups, but without triglycerides (TG) changes. TG/HDL-C decreased in 30 - 39 and 40 - 49 year (y) at post-HD (p 0.05). LDL-C/apoB increased in >60 y, after HD (p 60 (p 0.01). On the other hand, non-HDL-C/HDL-C reduced in 40 - 49 and >60 y, at post-HD (p 0.05). The linear-correlation between %14C-PL-incorporation and non-HDL-C/HDL-C was negative in 30 - 39 y, both at pre-HD (r = -0.90;p = 0.002) and post-HD (r = -0.78;p = 0.022). Linear-correlation between PON-1 and %14C-PL-incorporation was positive in >60 y, both at pre- (r = 0.63;p = 0.029) and post-HD (r = 0.65;p = 0.022). PON-1 activity increased at pre- (59 ± 30) and post-HD (73 ± 38) in 50 - 59 y (p 0.05). The %14C-PL-incorporation was reduced in >60 y (p 0.05), when compared to pre- and post-HD. esRD patients undergoing HD shows important changes on lipid-profile, PON-1-activity, cardiac risk ratios and HDL-remodeling. These results demonstrate the influence of HD with a polysulfone low-flow basic-dialyzing-membrane and unfractionated-heparin on lipoprotein metabolism.

目的:比较适利达与类似药物Drenatan在原发性开角型青光眼患者中关于降低眼内压的单药治疗疗效。方法:该回顾性研究包括62例患者(119眼)。初诊时,正在使用适利达的患者转为使用类似药物拉坦前列素(Drenatan)。12wk后的复诊阶段进行全面的眼科检查,包括裂隙灯、Goldmann压平眼压计、立体眼底、前房角镜、超声角膜测厚仪以及自动视野检查。结果:使用Drenatan之前和之后平均眼内压分别为12.30±2.02mmH g和12.38±2.05mmH g(P=0.559)。R=0.987显示两者显著相关。结论:适利达和Drenatan在降低眼内压方面具有相似功效。该发现对于降低公共卫生成本,维护其药物疗效和安全性政策具有重要意义。

Genetic Diversity of the Endemic Xantus’ Hummingbird Using 16 Novel Polymorphic Microsatellite Loci, and Their Cross Amplification between Six Related Species

A set of 16 microsatellite loci was developed and characterized for the Xantus’ hummingbird (Hylocharis xantusii) by using 454 next-generation sequencing. Twenty-five H. xantusii samples from one population were genotyped;all loci were polymorphic, with the number of alleles ranging from three to ten. The mean observed heterozygosity was 0.681 for all loci. No significant linkage disequilibrium was detected, but five loci (Hxan05, 06, 09, 13 and 14) showed deviation from Hardy-Weinberg equilibrium. These microsatellite loci are the first to be characterized for H. xantusii. A moderate to high level of cross-species amplification was observed across the six hummingbird species (31% - 87.5%), with the best cross amplification results observed in the closest related species (H. leucotis, Cynanthus latirostris, Calypte costae). The availability of these molecular tools allows assessing questions integrating population genetics, ecology, conservation, and evolutionary history for H. xantusii and for other phyogenetically related species.
