Arsenic trioxide induces regulatory functions of plasmacytoid dendritic cells through interferon-α inhibition

Arsenic trioxide(As2O3)is recently found to have therapeutic potential in systemic sclerosis(SSc),a life-threatening multi-system fibrosing autoimmune disease with type I interferon(IfN-I)*** activated plasmacytoid dendritic cells(pDCs)are responsible for IfN-I secretion and are closely related with fibrosis establishment in *** this study,we showed that high concentrations of As2O3 induced apoptosis of pDCs via mitochondrial pathway with increased BAX/BCL-2 ratio,while independent of reactive oxygen species ***,at clinical relevant concentrations,As2O3 preferentially inhibited IfN-αsecretion as compared to other cytokines such as TNf-α,probably due to potent down-regulation of the total protein and mRNA expression,as well as phosphorylation of the interferon regulatory factor7(IRf7).In addition,As2O3 induced a suppressive phenotype,and in combination with cytokine inhibition,it down-regulated pDCs’capacity to induce CD4+T cell proliferation,Thl/Th22 polarization,and B cell differentiation towards ***,chronically activated pDCs from SSc patients were not resistant to the selective IfN-αinhibition,and regulatory phenotype induced by ***,our data suggest that As2O3 could target pDCs and exert its treatment efficacy in SSc,and more autoimmune disorders with IfN-I signature.

Successful liver allograft inflow reconstruction with the right gastroepiploic vein

portal vein thrombosis is a common complica- tion in cirrhotic patients. When portal vein thrombectomy is not a suitable option, a large collateral vessel can be used for allograft venous inflow reconstruction. We describe an unusu- al case of successful portal revascularization using the right gastroepiploic vein. The patient underwent a cadaveric or- thotropic liver transplantation with end-to-end anastomosis of the portal vein to the right gastroepiploic vein. Six months after liver transplantation the patient is well with good liver function. The use of the right gastroepiploic vein for allograft venous reconstruction is feasible and safe, with a great advan- tage of avoiding the need of venous jump graft.

An uneasy alliance:a nesting association between aggressive ants and equally fierce social wasps

Although the Neotropical territorially dominant arboreal ant Azteca chartifex forel is very aggressive towards any intruder,its populous colonies tolerate the close presence of the fierce polistine wasp Polybia rejecta(f.).In french Guiana,83.33%of the 48 *** nests recorded were found side by side with those of *** nesting association results in mutual protection from predators(i.e.,the wasps protected from army ants;the ants protected from birds).We conducted field studies,laboratorybased behavioral experiments and chemical analyses to elucidate the mechanisms allowing the persistence of this *** to differences in the cuticular profiles of the two species,we eliminated the possibility of chemical ***,analyses of the carton nests did not reveal traces of marking on the *** ant forager flows were not perturbed by extracts from the wasps’Dufour’s and venom glands,we rejected any hypothetical action of repulsive ***,we noted that the wasps"scraped"the surface of the upper part of their nest envelope using their mandibles,likely removing the ants'scent trails,and an experiment showed that ant foragers were perturbed by the removal of their scent *** leads us to use the term"erasure hypothesis."Thus,this nesting association persists thanks to a relative tolerance by the ants towards wasp presence and the behavior of the wasps that allows them to"contain"their associated ants through the elimination of their scent trails,direct attacks,"wing-buzzing"behavior and ejecting the ants.

Recent studies have demonstrated a new role for Klf10, a Krüppel-like transcription factor, in skeletal muscle, specifically relating to mitochondrial function. Thus, it was of interest to analyze additional tissues that are highly reliant on optimal mitochondrial function such as the cerebellum and to decipher the role of Klf10 in the functional and structural properties of this brain region. In vivo (magnetic resonance imaging and localized spectroscopy, behavior analysis) and in vitro (histology, spectroscopy analysis, enzymatic activity) techniques were applied to comprehensively assess the cerebellum of wild type (WT) and Klf10 knockout (KO) mice. Histology analysis and assessment of locomotion revealed no significant difference in Klf10 KO mice. Diffusion and texture results obtained using MRI revealed structural changes in KO mice characterized as defects in the organization of axons. These modifications may be explained by differences in the levels of specific metabolites (myo-inositol, lactate) within the KO cerebellum. Loss of Klf10 expression also led to changes in mitochondrial activity as reflected by a significant increase in the activity of citrate synthase, complexes I and IV. In summary, this study has provided evidence that Klf10 plays an important role in energy production and mitochondrial function in the cerebellum.

Hemodynamic and antifibrotic effects of a selective liver nitric oxide donor V-PYRRO/NO in bile duct ligated rats

AIM: To assess whether a liver specific nitric oxide (NO) donor (V-PYRRO/NO) would prevent the development of portal hypertension and liver fibrosis in rats with bile duct ligation (BDL). METHODS: Treatment (placebo or V-PYRRO/NO 0.53 μmol/kg per hour) was administered i.v. to rats 2 d before BDL (D-2) and maintained until the day of hemodynamic measurement (D26). Intra-hepatic NO level was estimated by measuring liver cGMP level. Effects of V-PYRRO/NO on liver fibrosis and lipid peroxidation were also assessed. RESULTS: Compared to placebo treatment, V-PYRRO/ NO improved splanchnic hemodynamics in BDL rats: portal pressure was significantly reduced by 27% (P flow by 74%, P = 0.007). Moreover, V-PYRRO/NO significantly prevented liver fibrosis development in BDL rats (by 30% in hepatic hydroxyproline content and 31% in the area of fibrosis, P ffect being probably due to a decrease in lipid peroxidation by 44% in the hepatic malondialdehyde level (P = 0.007). Interestingly, we observed a significant and expected increase in liver cGMP, without any systemic hemodynamic effects (mean arterial pressure, vascular systemic resistance and cardiac output) in both sham- operated and BDL rats treated with V-PYRRO/NO. This result is in accordance with studies on V-PYRRO/NO metabolism showing a specific release of NO in the *** in BDL rats improved liver fibrosis and splanchnic hemodynamics without any noxious systemic hemo- dynamic effects.

荻草谷网蚜Sitobion miscanthi(Takahashi)(在中国长时间学名误用为麦长管蚜Sitobion avenae fabricius)是我国小麦的主要害虫,为小麦蚜虫优势种。该蚜虫因繁殖速度快、发育历期短、适应能力强且能随着气流远距离迁飞等特性,对小麦安全生产构成严重威胁。目前关于蚜虫种群遗传结构的研究主要基于线粒体基因开展。然而,初级共生菌布赫纳氏菌Buchnera aphidicola几乎存在于所有蚜虫中,其在研究蚜虫种群遗传结构多样性方面的潜在作用报道很少。本研究基于前期收集的荻草谷网蚜6个不同地理种群(苏州、武汉、昆明、廊坊、泰安、银川),对其线粒体COⅠ基因与初级共生菌***的两个单拷贝基因gnd与trpA进行PCR扩增、测序和群体遗传结构分析。根据对3个遗传标记基因聚类分析,荻草谷网蚜6个不同地理种群分为3组,分别为银川种群、苏州种群和其他种群(COⅠ:f_(CT)=0.3642,Pf_(CT)=0.4033,Pf_(CT)=0.2229,P<0.05),3组间存在显著的遗传差异。银川种群和泰安种群间有相同的稀有单倍型H8(trpA),苏州种群和武汉种群也有相同的稀有单倍型H2与H23(COⅠ),3组之间仍存在基因交流。因此我们推测在4、5月份东南季风盛行的情况下,银川种群迁入了外来虫源。蚜虫线粒体基因和初级共生菌基因的遗传结构分析结果基本一致,表明共生菌基因在研究蚜虫种群遗传结构和多样性具有潜在应用意义。综上所述,基于线粒体和共生菌基因序列的荻草谷网蚜不同地理种群的遗传结构解析可为蚜虫的迁飞提供分子证据。

AIM: To investigate the conformity of management practices of gastrointestinal hemorrhage in cirrhotic patients with relevant guidelines. METHODS: A questionnaire on the management of digestive bleeding was completed for all consecutive cirrhotic patients admitted to 31 french hospitals. RESULTS: One hundred and twenty-six bleeding events were recorded. It was the first bleeding episode in 79 patients (63%), of whom 40 (51%) had a prior diagnosis of cirrhosis and 25 (32%) had previously undergone an endoscopy. The bleeding episode was a recurrence in 46 patients (37%). The median time between onset and admission was 4 h, but exceeded 12 h in 42% of cases. There was an agreement between centers forearly vasoactive drug administration (87% of cases), association with ligation (42%) more often than sclerosis (21%) at initial endoscopy, and antibiotic prophylaxis (64%). By contrast, prescription of beta-blockade alone or in combination (0 to 100%, P = 0.003) for secondary prophylaxis and lactulose (26% to 86%, P = 0.04), differed among centers. CONCLUSION: In french hospitals, management of bleeding related to portal hypertension in cirrhotic patients is generally in keeping with the consensus. Broad variability still remains concerning beta-blockade use for secondary prophylaxis. Screening for esophageal varices, the use of antibiotic prophylaxis and patients information need to be improved.

Gut immune response in the presence of hepatitis C virus infection

Hepatitis C virus （HCV） is an important etiologic agent of hepatitis and a major cause of chronic liver infection that often leads to cirrhosis, fibrosis and hepatocel-lular carcinoma. Although, HCV is a hepatotropic virus, there is strong evidence that HCV could replicate extra-hepatic in the gastrointestinal tissue which could serve as a reservoir for HCV. The outcome of HCV infection depends mainly on the host innate and adaptive im-mune responses. Innate immunity against HCV includes mainly nuclear factor cells and activation of IfN-related genes. There is an immunologic link between the gut and the liver through a population of T-cells that are ca-pable of homing to both the liver and gut via the portal circulation. However, little is known on the role of Gut immune response in HCV. In this review we discussed the immune regulation of Gut immune cells and its as-sociation with HCV pathogenesis, various outcomes of anti-HCV therapy, viral persistence and degree of liver infammation. Additionally, we investigated the relation-ship between Gut immune responses to HCV and IL28B genotypes, which were identifed as a strong predictor for HCV pathogenesis and treatment outcome after acute infection.

Over the past 2 to 3 decades,Chinese forests are estimated to act as a large carbon sink,yet the magnitude and spatial patterns of this sink differ considerably among *** 3 microwave(L-and X-band vegetation optical depth[VOD])and 3 optical(normalized difference vegetation index,leaf area index,and tree cover)remote-sensing vegetation products,this study compared the estimated live woody aboveground biomass carbon(AGC)dynamics over China between 2013 and *** results showed that tree cover has the highest spatial consistency with 3 published AGC maps(mean correlation value R=0.84),followed by L-VOD(R=0.83),which outperform the other *** AGC estimation model was proposed to combine all indices to estimate the annual AGC dynamics in China during 2013 to *** performance of the AGC estimation model was good(root mean square error=0.05 Pg C and R^(2)=0.90 with a mean relative uncertainty of 9.8% at pixel scale[0.25°]).Results of the AGC estimation model showed that carbon uptake by the forests in China was about+0.17 Pg C year^(-1) from 2013 to *** the regional level,provinces in southwest China including Guizhou(+22.35 Tg C year^(-1)),Sichuan(+14.49 Tg C year^(-1)),and Hunan(+11.42 Tg C year^(-1))provinces had the highest carbon sink rates during 2013 to *** of the carbon-sink regions have been afforested recently,implying that afforestation and ecological engineering projects have been effective means for carbon sequestration in these regions.
