Essential amino acid ratios and mTOR affect lipogenic gene networks and miRNA expression in bovine mammary epithelial cells

Background： The objective of this study was to study how changing the ratio of Lys to Thr, Lys to His, and Lys to Val affects the expression of lipogenic genes and microRNA （miRNA） in bovine mammary epithelial cells. Results： Triplicate cultures with the respective ＂optimal＂ amino acid （AA） ratio （OPAA = Lys：Met 2.9：1; Thr：Phe 1.05：1; Lys：Thr 1.8：1; Lys：His 2.38：1; Lys：Val 1.23：1） plus rapamycin （OPAARMC; positive control）, OPAA, Lys：Thr 2.1：1 （LT2.1）, Lys：Thr 1.3：1 （LT1.3）, Lys：His 3.05：1 （LH3.0）, or Lys：Val 1.62：1 （LV1.6） were incubated in lactogenic medium for 12 h. The expression of 15 lipogenic genes and 7 miRNA were evaluated. Responses to LT2.1, LT1.3, LH3.0, and LV1.6 relative to the control （OPAARMC） included up-regulated expression ofACSS2, FABP3, ACACA, FASN, SCD, LPIN1, INSIG1, SREBF1, PPARD, and NR1H3 （commonly known as LXR-a）. Furthermore, LV1.6 up-regulated expression of ACSL1, DGAT1, and RXRA and down-regulated PPARG expression. Although no effect of OPAA on expression of PPARG was observed, compared with the control, OPAA up-regulated expression of the PPAR targets ACSS2, FABP3, ACACA, FASN, SCD, LPIN1, INSIG1, and SREBF1. Compared with the control, the expression of the anti-lipogenic MIR27AB was down-regulated by OPAA, LT2.1, LT1.3 and LH3.0. In contrast, compared with the control, the expression of the pro-lipogenic MIR21 was up-regulated by LT2.1, LT1.3, LH3.0, and LV1.6. Conclusions： The observed up-regulation of lipogenic gene networks and the changes in expression of key miRNA involved in the control of lipogenic balance are indicative of a potentially important role of EAA ratios and mTOR signaling in the regulation of milk fat synthesis.

Role of bile acids, prostag landins and COX inhibitors in chronic esophagitis in a mouse model

AIM. To develop a new experimental model of esophagitis that serves a complementary tool to clinical investigation in an insight into the mechanism of the damage to the esophagus mucosa by aggressive factors, and role of COX inhibitors in this process. METHODS： The study was conducted in 56 male mice. Animals were divided into seven groups：（1） perfused with HCI, （2） perfused with HCI and physiologic concentration of pepsin （HCI/P）, （3） perfused with similar HCI/P solution enriched with conjugated bile acids （glycho- and tauro-sodium salts） designated esophageal infusion catheter under the general anesthesia, （4） perfused as in group 2 treated with indometacin, （5） perfused as in group 2 treated with NS-398, （6） perfused as in group 3 treated with indometacin, and （7） perfused as in group 3 treated with NS-398. The esophagus was divided into 3 parts： upper, middle and lower. The PGE2 concentration was measured in all parts of esophagus using ria method. Esophagus of sacrificed animals was macroscopically evaluated using a low power dissecting microscope （20x）. Specimeris, representing the most frequently seen changes were fixed, stained with H＆E and assessed microscopically using the damage score, and inflammatory score. RESULTS： The macroscopic changes were significantly severer in HCI/P than those in HCI animals （77%） and in HCI/P/BA group （43%）. In HCI/P NS-398 group we noticed significantly less changes than those in not treated group （42%） and in analogical group treated with indometacine （45%）. In HCI/P/BA INDO group we observed significantly severer changes than that in not treated group （52%）. We noticed less changes in HCI/P NS-398 than that in group with indometacine （46%）. In HCI/P/BA NS-398 group we had less changes than that in indometacin group （34%）. The microscopic changes observed in HCI/P/BA INDO group were severer than that in not treated group （48%）. Direct oxidation of methane at low temperature using Pt/C,Pd/C,Pt/C-ATO and Pd/C-ATO electrocatalysts prepared by sodium borohydride reduction process

The main objective of this paper was to characterize the voltammetric profiles of the Pt/C,Pt/C-ATO,Pd/C and Pd/CATO electrocatalysts and study their catalytic activities for methane oxidation in an acidic electrolyte at 25 ℃ and in a direct methane proton exchange membrane fuel cell at 80 ℃. The electrocatalysts prepared also were characterized by X-ray diffraction( XRD) and transmission electron microscopy( TEM). The diffractograms of the Pt/C and Pt/C-ATO electrocatalysts show four peaks associated with Pt face-centered cubic( fcc) structure,and the diffractograms of Pd/C and Pd/C-ATO show four peaks associated with Pd face-centered cubic( fcc) structure. For Pt/C-ATO and Pd/C-ATO,characteristic peaks of cassiterite( SnO_2) phase are observed,which are associated with Sb-doped SnO_2( ATO) used as supports for electrocatalysts. Cyclic voltammograms( CV) of all electrocatalysts after adsorption of methane show that there is a current increase during the anodic scan. However,this effect is more pronounced for Pt/C-ATO and Pd/C-ATO. This process is related to the oxidation of the adsorbed species through the bifunctional mechanism,where ATO provides oxygenated species for the oxidation of CO or HCO intermediates adsorbed in Pt or Pd sites. From in situ ATR-FTIR( Attenuated Total Reflectance-Fourier Transform Infrared) experiments for all electrocatalysts prepared the formation of HCO or CO intermediates are observed,which indicates the production of carbon dioxide. Polarization curves at 80 ℃in a direct methane fuel cell( DMEFC) show that Pd/C and Pt/C electroacatalysts have superior performance to Pd/C-ATO and Pt/C-ATO in methane oxidation.

In this talk I report on recent work related to the dynamical generation of baryonic resonances, some made up from pseudoscalar meson-baryon, others from vector meson-baryon and a third type from two meson-one baryon systems. We can establish a correspondence with known baryonic resonances, reinforcing conclusions previously drawn and bringing new light on the nature of some baryonic resonances of higher mass.

The content of endogenons vasopressin (AVP1-9) and its metabolites in rat's various brain areas and pituitary gland was investigated by the combination techniques of high pressure liquid chromatography （HPLC） and radioimmunoassay （ria）. In addition to the AVP1-9, several compounds with C-terminal AVP immunoreactivity were detected in hypothalamus, hippceampus, amygdala, medulla oblongata and septum. The C-terminal AVP1-9metabolites were virtually absent in the pituitary gland. In view of the highly potent and selective effects oa memory processes of AVP metabolites, some endogenous AVP metabolites in the brain may play an important role in the central nervous system （CNS） function.

Visual-Motor Perceptual and Reading Processes of Schoolchildren With Attention Deficit Hyperactivity Disorder

Objectives:Compare and relate the performance of schoolchildren with Attention Deficit Hyperactivity Disorder(ADHD)and with good academic performance in the visual-motor perceptual and reading ***:Twenty schoolchildren,of both genders,aged between nine and 11 years old,participated in this *** were divided into two groups:Group I(GI):comprising 10 students diagnosed with ADHD;and Group II(GII):10 students with good academic performance paired according to gender,age group,and educational level with *** students were submitted to the Developmental Test of Visual Perception(DTVP 3)and Reading Processes Assessment Protocol(PROLEC).Results:There was lower performance of GI compared to GII in the subtests of copying,eye-hand coordination,and figure-ground in the standard score of general visual perception and visual-motor *** presented lower performance in all reading processes except for the punctuation marks *** was a statistically significant difference in the comparison between GI and GII regarding the classification of reading processes.A relationship was identified between the reading and visual perception subtests in the GI students,demonstrating a relationship between the visual perceptual motor skills and the reading processes in the group of students with ***:Schoolchildren with ADHD performed less well than their peers with good academic performance in the visual-motor perceptual and reading skills;such that the worse their performance in the visual-motor perceptual processes,the worse their performance in the reading processes.

The potential of gene therapies for spinal cord injury repair:a systematic review and meta-analysis of pre-clinical studies
