Enhanced axonal regeneration and functional recovery of the injured sciatic nerve in a rat model by lithium-loaded electrospun nanofibrous scaffolds

increasing evidence indicates that engineered nerve grafts have great potential for the regeneration of peripheral nerve injuries(PNis). While most studies have focused only on the topographical features of the grafts, we have considered both the biophysical and biochemical manipulations in our applied nanoscaffold. To achieve this, we fabricated an electrospun nanofibrous scaffold(ENS) containing polylactide nanofibers loaded with lithium(Li) ions, a Wnt/β-catenin signaling activator. in addition, we seeded human adipose-derived mesenchymal stem cells(h ADMSCs) onto this engineered scaffold to examine if their differentiation toward Schwann-like cells was induced. We further examined the efficacy of the scaffolds for nerve regeneration in vivo via grafting in a PNi rat model. Our results showed that Li-loaded ENSs gradually released Li within11 d, at concentrations ranging from 0.02 to(3.64 ± 0.10) mmol/L, and upregulated the expression of Wnt/β-catenin target genes(cyclin D1 and c-Myc) as well as those of Schwann cell markers(growth-associated protein 43(GAP43), S100 calcium binding protein B(S100B), glial fibrillary acidic protein(GFAP), and SRY-box transcription factor 10(SOX10)) in differentiated h ADMSCs. in the PNi rat model, implantation of Li-loaded ENSs with/without cells improved behavioral features such as sensory and motor functions as well as the electrophysiological characteristics of the injured nerve. This improved function was further validated by histological analysis of sciatic nerves grafted with Li-loaded ENSs, which showed no fibrous connective tissue but enhanced organized myelinated axons. The potential of Li-loaded ENSs in promoting Schwann cell differentiation of h ADMSCs and axonal regeneration of injured sciatic nerves suggests their potential for application in peripheral nerve tissue engineering.

An innovative Finite Element Geometric Modeling of Single-Layer Multi-Bead WAAMed Part

Finite element (FE) coupled thermal-mechanical analysis is widely used to predict the deformation and residualstress of wire arc additive manufacturing (WAAM) parts. in this study, an innovative single-layermulti-bead profilegeometric modeling method through the isosceles trapezoid function is proposed to build the FE model of theWAAMprocess. Firstly, a straight-line model for overlapping beads based on the parabola function was establishedto calculate the optimal center distance. Then, the isosceles trapezoid-based profile was employed to replace theparabola profiles of the parabola-based overlapping model to establish an innovative isosceles trapezoid-basedmulti-bead overlapping geometric model. The rationality of the isosceles trapezoid-based overlapping model wasconfirmed by comparing the geometric deviation and the heat dissipation performance index of the two overlappingmodels. in addition, the FE-coupled thermal-mechanical analysis, as well as a comparative experiment of thesingle-layer eight-bead deposition process show that the simulation results of the above two models agree with theexperimental results. At the same time, the proposed isosceles trapezoid-based overlappingmodels are all straightlineprofiles, which can be divided into high-quality FE elements. it can improve the modeling efficiency andshorten the simulation calculation time. The innovative modeling method proposed in this study can provide anefficient and high-precision geometricmodelingmethod forWAAMpart FE coupled thermal-mechanical analysis.

A significant research focus is placed on identifying animal species and areas at future risk to human-induced alterations of the environment and long-term changes in climatic ***,the extent to which exposure to extreme climatic events and intense human pressure can increase the risk of harmful impacts on species remains poorly *** on terrestrial mammals in China,one of the world’s megadiverse countries,we investigated patterns of contemporary exposure to floods and human pressures and determined their taxonomic representation and distribution across three major area-based conservation schemes,namely,national nature reserves(NNRs),priority areas for biodiversity conservation(PABCs),and key biodiversity areas(KBAs).Among the 440 species assessed with moderate or high exposure to floods,327(∼75%)also qualified as moderate or high in exposure to intense human *** species mainly belong to the orders Chiroptera,Eulipotyphla,and ***,there were 305,311,and 311 species with moderate or high exposure to flood and intense human pressure represented across NNRs,PABCs,and KBAs,*** findings support the prioritization of KBAs for expansion of site-based protection efforts such as NNRs in China,considering threats to species from exposure to adverse effects from both extreme climate and human pressure.
