In the present investigation the structural proteins associated with MAC-1 bacteriophage have been characterized using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE);tandem mass spectrometry of protein bands from SDS-PAGE gel;from the open reading frames (OrFs) deduced from MAC-1 genome sequence and amino acid sequence homology searches from the Uniprot database (up000002418). results have led to the identification of at least three structural proteins associated with MAC-1 phage genome. They are: capsid protein (~55,000-daltons);spike protein (~22,000-daltons) and a low molecular weight DNA binding protein (~4000-dal- tons). In addition, two other minor proteins were tentatively identified as replicative and scaffold proteins based on two to three unique peptides from mass spectrometry data. However, other proteins coded (OrFs) by phage genome remain to be identified.

For decades,clinicians have developed medications and therapies to alleviate the symptoms of Parkinson’s disease,but no treatment currently can slow or even stop the progression of this localized ***,sparked by the genetic revolution,stem cell reprogramming research and the advancing capabilities of personalization in medicine enable forward-thinking to unprecedented patient-specific modeling and cell therapies for Parkinson’s disease using induced pluripotent stem cells(iPSCs).In addition to modeling Parkinson’s disease more accurately than chemically-induced animal models,patient-specific stem cell lines can be created,elucidating the effects of genetic susceptibility and sub-populations’differing responses to in vitro *** cell therapy with iPSC lines provides ethical advantages because these stem cell lines do not require the sacrifice of human zygotes and genetically-specific drug trails can be tested in vitro without lasting damage to *** hopes of finally slowing the progression of Parkinson’s disease or re-establishing function,iPSC lines can ultimately be corrected with gene therapy and used as cell sources for neural transplantation for Parkinson’s *** relatively localized neural degeneration,similar to spinal column injury,Parkinson’s disease presents a better candidacy for cell therapy when compared to other diffuse degeneration found in Alzheimer’s or Huntington’s *** implantation of pluripotent cells poses the risk of an innate immune response and ***,therefore,must be taken to ensure cell line quality before *** cell quality can be quantified using a number of assays,a yielding a high percentage of therapeutically relevant dopaminergic neurons,minimal de novo genetic mutations,and standard chromosomal structure is of the utmost *** techniques focus on iPSCs because they can be matched with donors using human leukocyte antig

Prostate cancer （PCa） is the most common visceral malignancy in men with androgen deprivation therapy （ADT） the preferred therapy to suppress testosterone production and hence tumor growth. Despite its effectiveness in lowering testosterone, ADT is associated with side effects including loss of muscle mass, diminished muscle strength, decrements in physical performance, earlier fatigue and declining quality of life. This review reports a survey of the literature with a focus on changes in muscle strength, physical function and body composition, due to short-term and long-term ADT. Studies in these areas are sparse, especially well-controlled, prospective randomized trials. Cross-sectional and longitudinal data （up to 2 years） for men with PCa treated with ADT as well as patients with PCa not receiving ADT and age-matched healthy men are presented when available. Based on limited longitudinal data, the adverse effects of ADT on muscle function, physical performance and body composition occur shortly after the onset of ADT and tend to persist and worsen over time. Exercise training is a safe and effective intervention for mitigating these changes and initial guidelines for exercise program design for men with PCa have been published by the American College of Sports Medicine. Disparities in study duration, types of studies and other patient-specific variables such as time since diagnosis, cancer stage and comorbidities may all affect an understanding of the influence of ADT on health, physical performance and mortality.

Alpine Ecosystems of Northwest Yunnan,China:an Initial Assessment for Conservation

Implementing conservation actions on-the-ground is not a straightforward process,especially when faced with high scientific uncertainty due to limited available information. This is especially acute in regions of the world that harbor many unique species that have not been well studied,such as the alpine zone of the Hengduan Mountains of Northwest Yunnan (NWY),a global biodiversity hotspot and site of The Nature Conservancy’s Yunnan Great rivers Project. We conducted a quantitative,but rapid regional-level assessment of the alpine flora across NWY to provide a broad-based understanding of local and regional patterns of the alpine flora,the first large-scale analysis of alpine biodiversity patterns in this region. Multivariate analyses were used to classify the major plant community types and link community patterns to habitat variables. Our analysis indicated that most species had small distributions and/or small population sizes. Strong patterns emerged with higher diversity in the more northern mountains,but beta diversity was high,averaging only 10% among sites. The ordinations indicated that elevation and geographic location were the dominant environ-mental gradients underlying the differences in the species composition among communities. The high beta diversity across the alpine of these mountains implies that conservation strategies ultimately will require the protection of large numbers of species over a large geographical area. However,prioritiza-tion should be given to areas where potential payoffs are greatest. Sites with high species richness also have a greater number of endemic species,and,by focusing efforts on these sites,conservation investments would be maximized by protecting the greatest number of unique species.

Taxonomic and metabolic shifts in the Coorong bacterial metagenome driven by salinity and external inputs

The Coorong estuary lies at the terminus of Australia's largest river system, the Murray-Darling; both are strongly influenced by human activities; including farming and extensive flow modification. Metagenomic approaches were used to determine the planktonic bacterial community composition and potential metabolic function at two extremes in the Coorong, the river mouth which exhibits marine-like salinity, and the hypersaline upper-reaches of the estuary. Significant shifts in taxa and metabolic function were seen between the two sites. The river mouth exhibited an increase in abundance of r hodobacteriaceae and Alteromonadaceae; families readily able to adapt to change in nutrient conditions; and the potentially pathogenic families B rucellaceae, Enterobacteriaceae and Vibrionaceae. Metabolisms over-represented include motility and chemotaxis, rNA metabolism and membrane transport, all of which are involved in actively searching for and obtaining nutrients. Also over-represented were metabolisms involved in population succession and stress response. An over-representation of taxa and metabolisms indicative of environmental change is reflective of anthropogenically af fected riverine input. In the hypersaline upper reaches of the estuary, the halophilic family Ectothiorhodospiraceae was over-represented, as were the families Flavobacteriaceae, Cytophagaceae and Nocardioidaceae, members of which are able to survive over a wide salinity range. Metabolisms over-represented here were reflective of increased bacterial growth, characteristic of hypersaline environments, and included DNA metabolism, nucleotide and nucleoside synthesis and cell cycle. Coorong metagenomes clustered taxonomically and metabolically with other planktonic metagenomes, but remained an outlier of this group with only 71% and 84% similarity, respectively. Acute on chronic liver failure(ACLF) is rarely the initial manifestation of a malignant process or precipitated by the initiation of anti-viral treatment with a nucleoside or nucleotide agent. We report an unusual case of ACLF temporally associated with initiation of Entecavir for treatment of chronic hepatitis B. Early Hodgkin's lymphoma(HL) was unmasked with initiation of the antiviral treatment which may have exacerbated ACLF. To the best of our knowledge, this has not been described in the literature. In reviewing our patients clinical course and liver autopsy, he developed a severe acute exacerbation of his chronic hepatitis B virus coinciding with the institution of antiviral therapy and the underlying HL perhaps modulating the overall degree of hepatic injury.

Distribution of antioxidants and phenolic compounds in flour milling fractions from hard red winter wheat

Mature wheat kernels contain three main parts:endosperm,bran,and *** milling results in multiple streams that are chemically different;however,the distribution of antioxidants and phenolic compounds has not been well documented in terms of conventional milling by-product *** this study,multiple analytical methods were used to investigate antioxidant activity and phenolic compound compositions of hard red winter wheat(whole ground wheat),the parts of a wheat kernel(bran,flour,germ),and wheat by-product streams(mill feed,red dog,shorts)for the first *** each mill stream,phenolic compounds(total,flavonoid,and anthocyanin contents)were determined and antioxidant activities were evaluated with 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical-scavenging activity,ferric reducing/antioxidant power(FrAP),and total antioxidant capacity *** differences(Pre observed in phenolic concentrations among fractions of bran,flour,and germ milled from the same kernels and noted that germ accounts for the majority of antioxidant properties,whereas bran contains a substantial portion of phenolic compounds and *** feed was high in phenolic content(5.29 mg FAE/g),total antioxidant capacity(866 mg/g),and antioxidant activity(up to 75% DPPH inhibition and 20.26μmol FeSO_(4)/g).The comprehensive information on distribution of antioxidants and phenolic compounds provides insights for future human consumption of commonly produced co-products from flour milling,and for selecting and using different milling fractions to make foods with improved nutritional properties.

The stadium effect:rodent damage patterns in rice fields explored using giving-up densities

rodents are globally important pre-harvest pests of *** Southeast Asia,rodent damage to growing rice crops is commonly concentrated towards the center of rice fields,away from the field edge,resulting in a clear pattern known as the“stadium effect.”To further understand this behavior of rodent pests and to develop recommendations for future research and management,we examined the relation between giving-up densities(GUDs)and damage *** Tanay,Luzon,Philippines,GUD trays containing pieces of coconut in a matrix of sand were placed at 4 different distances from the field edge to quantify the perceived risk of predation in a rice field pest,rattus *** were recorded during a dry and wet season crop at the reproductive and ripening stages of *** addition,assessments of active burrows,tracking tile activity and rodent damage to the rice crop,were conducted in the dry *** were significantly lower in the center of the rice fields than on the field edges,suggesting that rodent damage to rice is greater in the middle of rice fields due to a lower perceived predation ***,this perception of predation risk(or fear)increases towards the field edge and was greatest on the rice bund,where there was no vegetation *** discuss the implications for rodent management and rodent damage assessments in rice *** is the first documented use of GUDs in a rice agro-ecosystem in Asia;thus we identify the challenges and lessons learned through this process.
