RNF26 up-regulates PD-l1 to regulate the cancer immune response in ccRCC

Background: Clear cell renal cell carcinoma (ccRCC) stands as the most prevalent form of kidney cancer, accounting for a significant proportion of malignancies affecting the kidneys. ccRCC is well known as a type of tumour with immunogenicity. Immune checkpoint inhibitors (ICIs) aim to enhance the anticancer immune response in ccRCC by blocking programmed cell death 1 ligand 1/programmed death 1 (PD-l1/PD-1) pathways. In a previous study, we showed that RING finger protein 26 (RNF26) degrades chromobox 7 (CBX7) to activate the tumor necrosis factor (TNF) in ccRCC. Methods: We analyzed The Cancer Genome Atlas (TCGA) database using the R package ESTIMATE and found that RNF26 was significantly associated with ccRCC immune infiltration. The relationship between RNF26 and the PD1 checkpoint signaling pathway was detected by enrichment analysis. In addition, the molecular mechanism of RNF26 up-regulation of PD-l1 was detected by transcriptome sequencing, RT-qPCR, and Western Blot in ccRCC cell lineages 786-O and A498 cells. The transplantation tumor experiments in C57Bl/6 mice were used to test the efficacy of anti-PD1 and knockdown of RNF26 in vivo. Results: We showed that RNF26 suppressed the immune response to ccRCC. Next, we revealed that RNF26 activated the PD-1 checkpoint pathway to suppress the immune response to ccRCC, possibly via the CBX7/PD-l1 axis. Conclusion: The suggestion derived from our results is that targeting RNF26 holds the potential to amplify the efficacy of anticancer immunotherapies in the treatment of ccRCC.

Polymer: Non-fullerene acceptor heterojunction-based phototransistor for short-wave infrared photodetection

It remains full of challenge for extending short-wave infrared(SWIR)spectral response and weak-light detection in the context of broad spectral responses for *** this work,a novel poly(2,5-bis(4-hexyldodecyl)-2,5-dihydro-3,6-di-2-thienyl-pyrrolo[3,4-c]pyrrole-1,4-dione-alt-thiophene)(PDPPT3-HDO):COTIC-4F organic bulk-heterojunction is prepared as active layer for bulk heterojunction ***3-HDO serves as a hole transport material,while COTIC-4F enhances the absorption of SWIR light to 1020 *** a result,smooth and connected PDPPT3-HDO film is fabricated by blade coating method and exhibits high hole mobility up to 2.34 cm^(2)·V^(-1)·s^(-1) with a current on/off ratio of 4.72×10^(5) in organic thin film ***3-HDO:COTIC-4F heterojunction phototransistors exhibit high responsivity of 2680 A·W^(-1) to 900 nm and 815 A·W^(-1) to 1020 nm,with fast response time(rise time~20 ms and fall time~100 ms).The photosensitivity of the heterojunction phototransistor improves as the mass ratio of non-fullerene acceptors increases,resulting in an approximately two orders of magnitude enhancement compared to the bare polymer ***,the phototransistor exhibits decent responsivity even under ultra-weak light power of 43μW·cm^(-2) to 1020 *** work represents a highly effective and general strategy for fabricating efficient and sensitive SWIR light photodetectors.

A flexible multiscale algorithm based on an improved smoothed particle hydrodynamics method for complex viscoelastic flows

Viscoelastic flows play an important role in numerous engineering fields,and the multiscale algorithms for simulating viscoelastic flows have received significant attention in order to deepen our understanding of the nonlinear dynamic behaviors of viscoelastic ***,traditional grid-based multiscale methods are confined to simple viscoelastic flows with short relaxation time,and there is a lack of uniform multiscale scheme available for coupling different solvers in the simulations of viscoelastic *** this paper,a universal multiscale method coupling an improved smoothed particle hydrodynamics(SPH)and multiscale universal interface(MUI)library is presented for viscoelastic *** proposed multiscale method builds on an improved SPH method and leverages the MUI library to facilitate the exchange of information among different solvers in the overlapping *** test the capability and flexibility of the presented multiscale method to deal with complex viscoelastic flows by solving different multiscale problems of viscoelastic *** the first example,the simulation of a viscoelastic Poiseuille flow is carried out by two coupled improved SPH methods with different spatial *** effects of exchanging different physical quantities on the numerical results in both the upper and lower domains are also investigated as well as the absolute errors in the overlapping *** the second example,the complex Wannier flow with different Weissenberg numbers is further simulated by two improved SPH methods and coupling the improved SPH method and the dissipative particle dynamics(DPD)*** numerical results show that the physical quantities for viscoelastic flows obtained by the presented multiscale method are in consistence with those obtained by a single solver in the overlapping ***,transferring different physical quantities has an important effect on the numerical results.

Optimal structural design for a certain near-space composite propeller of airship using adaptive region division blending model

Near-space airship is a frontier and hotspot in current military research and development,and the near-space composite propeller is the key technology for its *** order to obtain higher aerodynamic efficiency at an altitude of 22 km,a certain near-space composite propeller is designed as a long and slender aerodynamic shape with a 10 m diameter,which brings many challenges to the composite structure *** initial design is obtained by the composite structure variable stiffness design method using based on fixed region division blending ***,it weighs 23.142 kg,exceeding the required 20 *** order to meet the structural design requirements of the propeller,a variable stiffness design method using the adaptive region division blending model is proposed in this *** with the methods using the fixed region division blending model,this method optimizes region division,stacking thickness and stacking sequence in a single level,considering the coupling effect among *** a more refined region division,this method can provide a more optimal design for composite tapered ***,to improve the efficiency of optimization subjected to manufacturing constraints,a hierarchical penalty function is proposed to quickly filter out the solutions that do not meet manufacturing *** above methods combined with a Genetic Algorithm(GA)using specific encoding are adopted to optimize the near-space composite *** optimal design of the structure weighs 18.831 kg,with all manufacturing constraints and all structural response constraints being *** with the initial design,the optimal design has a more refined region division,and achieves a weight reduction of 18.6%.This demonstrates that a refined region division can significantly improve the mechanical performance of the composite tapered structure.

Study on the mechanism and reaction characteristics of metal-supported phosphogypsum as oxygen carrier in a chemical looping gasification application

This study aimed to explore the chemical looping gasification(ClG)reaction characteristics of the metal-supported composite phosphogypsum(PG)oxygen carriers(OCs)and the thermodynamic *** FactSage 7.1 thermodynamic simulation was used to explore the oxygen release and H_(2)S removal *** experimental results showed that the syngas yield of ClG with PG-CuFe_(2)O_(4)was more than that with PG-Fe_(2)O_(3)20/CuO40 or PG-Fe_(2)O_(3)30/CuO30 OC at 1023 K when the water vapor content was ***,the maximum syngas yield of the CO selectivity was 70.3% and of the CO_(2)selectivity was 23.8%.The H_(2)/CO value was 0.78,and the highest carbon conversion efficiency was 91.9% in PG-CuFe_(2)O_(4)at the gasification temperature of 1073 *** metal-supported PG composite oxygen carrier was proved not only as an oxygen carrier to participate in the preparation of syngas but also as a catalyst to catalyze coal gasification ***,both the experimental results and FactSage 7.1 thermodynamic analysis revealed that the trapping mechanism of H_(2)S by composite OCs was as follows:CuO first lost lattice oxygen as an oxygen carrier to generate Cu_(2)O,which,in turn,reacted with H_(2)S to generate Cu_(2)*** study provided efficient guidance and reference for OC design in ClG.

Aerial refueling scheduling of multi-receiver and multi-tanker under spatial-temporal constraints for forest firefighting

Forest fires pose a significant threat to human life and property,so the utilization of unmanned aircraft systems provides new ways for forest *** the constrained load capacities of these aircraft,aerial refueling becomes crucial to extend their operational time and *** order to address the complexities of firefighting missions involving multi-receiver and multi-tanker deployed from various airports,first,a fuel consumption calculation model for aerial refueling scheduling is established based on the receiver ***,two distinct methods,including an integrated one and a decomposed one,are designed to address the challenges of establishing refueling airspace and allocating tasks for *** methods aim to optimize total fuel consumption of the receivers and tankers within the aerial refueling scheduling *** optimization problem is established as nonlinear optimization models along with *** integrated method seamlessly combines refueling rendezvous point scheduling and tanker task allocation into unified *** has a complete solution space and excels in optimizing total fuel *** decomposed method,through the separation of rendezvous point scheduling and task allocation,achieves a reduced computational ***,this comes at the cost of sacrificing optimality by excluding specific feasible ***,numerical simulations are carried out to verify the feasibility and effectiveness of the proposed *** simulations yield insights crucial for the practical engineering application of both the integrated and decomposed methods in real-world *** comprehensive approach aims to enhance the efficiency of forest firefighting operations,mitigating the risks posed by forest fires to human life and property.

In-depth understanding of transport behavior of sulfided nano zerovalent iron/reduced graphene oxide@guar gum slurry: Stability and mobility

Nanoscale zero-valent iron(NZVI),which has the advantages of small particle size,large specific surface area,and high reactivity,is often injected into contaminated aquifers in the form of ***,the prone to passivation and agglomeration as well as poor stability andmobility of NZVI limit the further application of this technology in ***,sulfided NZVI loaded on reduced graphene oxide(S-NZVI/rGO)and guar gum(GG)with shearthinning properties as stabilizers were used to synthesize S-NZVI/rGO@GG ***,TEM,and FT-IR confirmed that the dispersion and anti-passivation of NZVI were optimized in the coupled *** stability and mobility of the slurry were improved by increasing the GG concentration,enhancing the pH,and decreasing the ionic strength and the presence of Ca^(2+)ions,respectively.A modified advection-dispersion equation(ADE)was used to simulate the transport experiments considering the strain and physicochemical deposition/***,colloidal filtration theory(CFT)demonstrated that Brownian motion plays a dominant role in themigration of S-NZVI/rGO@GG slurry,and the maximummigration distance can be increased by appropriately increasing the injection ***-Derjaguin-landau-Verwey-Overbeek(XDlVO)theory showed that the excellent stability and migration of S-NZVI/rGO@GG slurry mainly came from the GG spatial *** study has important implications for the field injection of S-NZVI/rGO@GG *** to the injection parameters,the injection range of S-NZVI/rGO@GG slurry is effectively controlled,which lays the foundation for the promotion of application in actual fields.

A highly sensitive fluorescent HClO probe for discrimination between cancerous and normal cells/tissues

Elevated level of hypochlorous acid(HClO)is closely associated with cancer *** HClO level in cancer cells would provide important evidence in either early-stage cancer diagnostics or monitoring of its treatment *** this work,a new pyronine-based fluorescent probe for rapid and sensitive detection of HClO was developed by condensing meso-formyl pyronine(PyCHO)with 2-hydrazinopyridine to form meso-pyridylhydrazone-functionalized pyronine PyHP,PyHP is nonfluorescent due to the excited-state C=N isomerization nonradiative decay,whereas the HClO-triggered formation of meso-triazolopyridyl pyronine PyTP abolishes the C=N isomerization and thus greatly enhances the *** the probe,the cancer cells/tumor were distinguished with high-contrast from normal ones by laser confocal fluorescence imaging,and the tumor-to-normal(T/N)ratios obtained exceed the clinically acceptable threshold of ***,its capability of in vivo imaging tumor was also *** results indicate the potential of PyHP as an effective tool in the early clinical diagnosis of cancers.

Resource utilization of drinking water treatment aluminum sludge in green cementing materials: Hydration characteristics and hydration kinetics

As a byproduct of water treatment,drinking water treatment aluminum sludge(DWTAS)has challenges related to imperfect treatment and disposal,which has caused potential harm to human health and the *** this paper,heat treatment DWTAS as a supplement cementitious material was used to prepare a green cementing *** results show that the 800℃ is considered as the optimum heat treatment temperature for ***-800℃ is fully activated after thermal decomposition to form incompletely crystallized highly activeγ-Al2O3 and active SiO_(2).The addition of DWTAS promoted the formation of ettringite and C-(A)-S-H gel,which could make up for the low early compressive strength of cementing materials to a certain *** cured for 90 days,the compressive strength of the mortar with 30% DWTAS-800℃ reached 44.86 *** dynamic process was well simulated by Krstulovi′c-Dabi′c hydration kinetics *** study provided a methodology for the fabrication of environmentally friendly and cost-effective compound cementitiousmaterials and proposed a“waste-to-resource”strategy for the sustainable management of typical solid wastes.

Few-shot incremental radar target recognition framework based on scattering-topology properties

The continuous emergence of new targets in open scenarios leads to a substantial decrease in the performance of Inverse Synthetic Aperture Radar(ISAR)recognition ***,data scarcity further exacerbates the challenge of identifying new classes of ISAR *** this paper,a few-shot incremental target recognition framework based on Scattering-Topology Properties(STPIl)is ***,STPIl extracts scattering-topology properties of ISAR targets as recognition ***,the pseudo-incremental training strategy effectively alleviates the algorithm’s forgetting of old knowledge,and improves compatibility with new ***,a feature embedding network,with few parameters,is designed based on the graph neural *** embedding network is highly adaptable to changes in data ***,STPIl fully considers the joint distribution and marginal distribution in scattering features,and uses the Brownian distance metric module to make the scattering-topology features more *** results on both the simulation dataset and the public measured data indicate that STPIl can effectively balance new classes with old classes,and has superior performance to other advanced methods in the incremental recognition of targets.
