Cadmium isotope fractionation reveals genetic variation in Cd uptake and translocation by theobroma cacao and role of natural resistanceassociated macrophage protein 5 and heavy metal AtPase-family transporters

In response to new european Union regulations,studies are underway to mitigate accumulation of toxic cadmium(Cd)in cacao(theobroma cacao,tc).this study advances such research with Cd isotope analyses of 19 genetically diverse cacao clones and yeast transformed to express cacao natural resistance-associated macrophage protein(NRAMP5)and heavy metal AtPases(HMAs).the plants were enriched in light Cd isotopes relative to the hydroponic solution withΔ^(114/110)Cd_(tot-sol)=−0.22±0.08‰.Leaves show a systematic enrichment of isotopically heavy Cd relative to total plants,in accord with closed-system isotope fractionation ofΔ^(114/110)C_(dseq-mob)=−0.13‰,by sequestering isotopically light Cd in roots/stems and mobilisation of remaining Cd to *** findings demonstrate that(i)transfer of Cd between roots and leaves is primarily unidirectional;(ii)different clones utilise similar pathways for Cd sequestration,which differ from those of other studied plants;(iii)clones differ in their efficiency of Cd *** yeast that expresses tcNRAMP5(*** natural resistance-associated macrophage gene)had isotopically lighter Cd than did *** suggests that NRAMP5 transporters constitute an important pathway for uptake of Cd by *** isotope signatures of transgenic yeast expressing HMA-family proteins suggest that they may contribute to Cd *** data are the first to record isotope fractionation induced by transporter proteins in vivo.

Improving gastrointestinal scoring systems for predicting short-term mortality in critically ill patients

Shen et al’s retrospective study aims to compare the utility of two separate scoring systems for predicting mortality attributable to gastrointestinal(GI)injury in critically ill patients[the GI Dysfunction Score(GIDS)and the Acute Gastroin-testinal Injury(AGI)grade].the authors note that this study is the first proposal that suggests an equivalence between the ability of both scores to predict mor-tality at 28 days from intensive care unit(ICU)*** et al retrospec-tively analysed an ICU cohort of patients utilising two physicians administering both the AGI grade and GIDS score,using electronic healthcare records and ICU *** these physicians disagreed about the scores,the final decision as to the scores was made by an associate chief physician,or chief *** note that the primary reason for the development of GIDS was to create a clear score for GI dysfunction,with minimal subjectivity or inter-operator *** subjectivity inherent to the older AGI grading system is what ultimately led to the development of GIDS in *** ensuring consensus between physicians administering the AGI,Shen et al have controlled for one of this grading systems biggest *** have concerns,however,that this does not represent the real-world challenges associated with applying the AGI compared to the newer GIDS,and wonder if this arbitration process had not been instituted,would the two scoring systems remain equivalent in terms of predicted mortality?

Free vibration and transient response of double curved beams connected by intermediate straight beams

this paper investigates the free vibration and transient response of interconnected structures including double curved beams and intermediate straight beams,which are all constructed from functionally graded porous(FGP) materials. the strain potential and kinetic energies of each beam along with the work done by the external force are calculated. Additionally, a higher-order beam element is introduced to derive stiffness and mass matrices, along with the force vector. the curved and straight beams are discretized, and their assembled stiffness, mass matrices, and force vectors, are *** conditions at the joints are used to derive the total matrices of the entire structure. Subsequently, the natural frequencies and transient response of the system are determined. the accuracy of the mathematical model and the self-developed computer program is validated through the comparison of the obtained results with those of the existing literature and commercial software ANSYS, demonstrating excellent ***, a comprehensive study is conducted to investigate the effects of various parameters on the free vibration and transient response of the considered structure.

Inappropriate use of antibiotics leads to microbial ***-dose antibio-tic prophylaxis prior to laparoscopic cholecystectomy is well known for reducing the risk of postoperative infection in high-risk patients despite some conflicting ***-risk patients are those who are older than 70 years,have diabetes mellitus,whose operation time exceeded 120 minutes,have acute cholecystitis,experienced iatrogenic intraoperative gallbladder perforation resulting in bile or gallstone spillage,suffered from obstructive jaundice,or were deemed *** gallbladder perforation,one dose of antibiotic prophylaxis is ***,guidelines are needed and must be strictly ***-lactic treatment is not needed for patients at low risk of developing sepsis fo-llowing elective laparoscopic cholecystectomy,although the opposite is ***,superficial surgical infections are related to low *** without risk factors have a very low risk of ***,the routine use of anti-biotic prophylaxis in elective laparoscopic cholecystectomy is not recommended.

the interest in using the Datura stramonium plant is due to its natural products,which are used in many pharmaceutical *** objective of the current study was to assess the therapeutic and cytotoxic effects of the *** plant on two types of human cancer cell models(MCF7 and Ht29)in vitro.A soxhlet apparatus was used to obtain methanolic extract from dried plant *** recovered crude,after the solvent had evaporated,was then dispersed at varied concentrations of extract 100,50,20,and 0.0µg/mL and tested to see how the cells ***,the cancer-testis antigen(CtA)gene transcription in the two cell types exposed to the plant extract was examined using a semi-quantitative real-time polymerase chain *** chromatography–mass spectrometry(GC-MS)results produced the significant main metabolites Nonanoic acid,tropine N-Oxide,3,6-Ditigloyloxy-7-hydroxytropane,Hexadecanoic acid,2-Pentadecanone,6,10,14-trimethyl-,Carvenone,methyl ester,Phytol,Aposcopolamine,Hyoscyamine,4,8,12,16-tetramethylheptadecan-4-olide,Scopolamine,Alpha.-tocospiro A,1,2-Cycloheptanedione,3,3,7,7-tetramethyl-,dihydrazone,Campesterol,Stigmasterol,Gamma-Sitosterol and dl-.alpha.-*** results showed that the two types of cell lines impacted by *** extract,through untreated type 1 cells(MCF7)gave a highly significant transcription according to all applicable *** implemented analyses cleared the strong genetic impacts of Datura extract on cancer cells’***2LY and C2orf63 transcript accumulation were also significantly elevated when exposed to plant extract at a level of 50µg/mL in cell line type 2(Ht29),but tGIF2LY and P53 had the lowest relative expression at a level of 100µg/mL when treated the same cell line type.

Kaempferol improves glucose uptake in skeletal muscle via an AMPK-dependent mechanism

Insulin resistance is a hallmark of type-2 diabetes(t2D)*** skeletal muscle(SkM)is the major tissue for insulin-mediated glucose disposal,insulin resistance in SkM is considered a major risk factor for developing ***,the identifi cation of compounds that enhance the ability of SkM to take up glucose is a promising strategy for preventing *** previous work showed that kaempferol,a fl avonol present in many foods,improves insulin sensitivity in obese mice,however,the mechanism underlying this beneficial action remains ***,we show that kaempferol directly stimulates glucose uptake and prevents lipotoxicity-impaired glucose uptake in primary human *** stimulates Akt phosphorylation in a time-dependent manner in human SkM *** effect of kaempferol on glucose uptake was blunted by inhibition of glucose transporter 4,phosphoinositide 3-kinase(PI3K),or *** addition,kaempferol induced AMPK phosphorylation,and inhibition of AMPK prevented kaempferol-stimulated Akt *** vivo,kaempferol administration induced rapid glucose disposal accompanied with increased Akt and AMPK phosphorylation in SkM tissue of the *** together,these fi ndings suggest that kaempferol stimulates glucose uptake in SkM via an AMPK/Akt dependent mechanism,and it may be a viable therapeutic agent for insulin resistance.

Amount of polyhydramnios attributable to diabetes may be less than previously reported

AIM to evaluate the frequency and the quantity of polyhydramnios attributable to diabetes in *** the charts of patients with a four-quadrant amniotic fluid index(AFI) ≥ 20 cm and either a diagnosis of diabetes or a diabetes screening test during the index pregnancy were retrospectively reviewed. AFI was stratified into 5 categories and the frequency of diabetes was evaluated for each group. the frequency of polyhydramnios attributable to diabetes was compared to the frequency of polyhydramnios in the setting of fetal anomalies or no known cause. ReSULtS One thousand five hundred and forty-five patients were included in the study. eight point five percent(n = 131) had diabetes and no other cause for polyhydramnios. eleven point two percent(173) had antenatally diagnosed anomalies. For all categories of AFI except the largest(> 40.9 cm) the most common cause of polyhydramnios was idiopathic. In patients with diabetes the AFI was most likely to be between 26 cm and 35.9 *** the rate of polyhydramnios in this study is 8.5%. Patients with diabetes most commonly have mild polyhydramnios between 26 and 35.9 cm of fluid on a four-quadrant AFI.

Mitigation of atrazine,S-metolachlor,and diazinon using common emergent aquatic vegetation

By the year 2050, the population of the United States is expected to reach over 418 million, while the global population will reach 9.6 billion. to provide safe food and fiber, agriculture must balance pesticide usage against impacts on natural resources. Challenges arise when storms cause runoff to be transported to aquatic receiving systems. Vegetated systems such as drainage ditches and constructed wetlands have been proposed as management practices to alleviate pesticide runoff. twelve experimental mesocosms （1.3 ±0.71 ± 0.61 m） were filled with sediment and planted with a monoculture of one of three wetland plant species （typha latifolia, Leersia oryzoides, and Sparganium arnericanurn）. three mesocosms remained unvegetated to serve as controls. All mesocosms were amended with 9.2 ± 0.8 μg/L, 12 ± 0.4 μg/L, and 3.1 ± 0.2 μg/L of atrazine, metolachlor, and diazinon, respectively, over a 4 hr hydraulic retention time to simulate storm runoff. Following the 4 hr amendment, non-amended water was flushed through mesocosms for an additional 4 hr. Outflow water samples were taken hourly from pre-amendment through 8 hr, and again at 12, 24, 48, 72, and 168 hr post-amendment. L. oryzoides and t. latifolia had mean atrazine, metolachlor, and diazinon retentions from B1%-55% for the first 4 hr of the experiment. Aside from S. arnericanum and atrazine （25% retention）, unvegetated controls had the lowest pesticide retention （17%-28%） of all compared mesocosms. While native aquatic vegetation shows promise for mitigation of pesticide runoff, further studies increasing the hydraulic retention time for improved efficiency should be examined.
