Possible biological and translational significance of mast cells density in colorectal cancer

Mast cells (MCs), located ubiquitously near blood vessels, are descended from CD34+ hematopoietic stem cells. Initially, although their role has been well defined in hypersensitivity reactions, the discovery of their sharing in both innate and adaptive immunity has allowed to redefine their crucial interplay on the regulatory function between inflammatory and tumor cells through the release of mediators granule-associated (mainly tryptase and vascular endothelial growth factor). In particular, in several animal and human malignancies it has been well demonstrated that activated c-Kit receptor (c-KitR) and tryptase (an agonist of the proteinase-activated receptor-2) take pivotal part in tumor angiogenesis after the MCs activation, contributing to tumor cells invasion and metastasis. In this review, we focused on crucial MCs density (MCD) role in colorectal cancer (CRC) development and progression angiogenesis-mediated; then, we will analyze the principal studies that have focused on MCD as possible prognostic factor. Finally, we will consider a possible role of MCD as novel therapeutic target mainly by c-KitR tyrosine kinase inhibitors (imatinib, masitinib) and tryptase inhibitors (gabexate and nafamostat mesylate) with the aim to prevent CRC progression.

Faced with the scarcity of surface water accentuated by climate change, particularly in many arid and semi-arid countries, the quality of groundwater used for irrigation is a concern to agronomists and hydrogeologists. When these waters are of deep origin, they may have high mineralization and chemical compositions unsuitable for irrigation;in particular, they may alter soils and crops. It is therefore important to optimize the spatial estimation of the salinity of these waters and contribute to better knowledge of their quality, through an adapted and robust statistical and geostatistical approach. In the case of north-eastern Algeria, the objective of this study is to characterize the quality of deep waters and to test two interpolation methods (Inverse distance weight and ordinary Kriging) of their electrical conductivity (EC) as an indicator of their salinity and of the risk of damaging irrigated soils. 51 groundwater samples were taken in this region where there are many thermal springs, the water of which is used for irrigation and often is highly mineralized (EC between 0.6 and 26.6 dS/m). The geology is composed of karstic rocks crossed by large faults that allow deep water to rise. Based on major elements contents, analysis of the hydrochemical facies of these waters shows that the main facies are hyperchlorinated sodium (38%) and sulfated calcium (32%). The RSC (Residual Sodium Carbonate) and SAR (Irrigation water salt) indexes were used to assess the water quality. The results indicate that the majority of the sampled groundwater present a risk for soils irrigated with these waters (almost 1/3 presents a strong risk). Dexamethasone-loaded silicone matrices offer an interesting potential as innovative drug delivery systems,*** the treatment of inner ear diseases or for ***,very long drug release periods are targeted:several years/*** renders the development and optimization of novel drug products cumbersome:experimental feedback on the impact of the device design is obtained very slowly.A better understanding of the underlying mass transport mechanisms can help facilitating research in this field.A variety of silicone films were prepared in this study,loaded with amorphous or crystalline *** polymorphic drug forms were investigated,the film thickness was altered and the drug optionally partially/completely exchanged by much more water-soluble dexamethasone‘phosphate’.Drug release studies in artificial perilymph,scanning electron microscopy,optical microscopy,differential scanning calorimetry,X-ray diffraction and Raman imaging were used to elucidate the physical states of the drugs and polymer,and of the systems’structure as well as dynamic changes thereof upon exposure to the release *** particles were initially homogeneously distributed throughout the *** hydrophobicity of the matrix former very much limits the amounts of water penetrating into the system,resulting in only partial drug *** mobile drug molecules diffuse out into the surrounding environment,due to concentration ***,Raman imaging revealed that even very thin silicone layers(<20µm)can effectively trap the drug for prolonged periods of *** physical state of the drug(amorphous,crystalline)did not affect the resulting drug release kinetics to a noteworthy extent.

S-Nitrosothiols or thionitrites with the general formula RSNO are formally composed of the nitrosyl cation(NOt) and a thiolate(RSà), the base of the corresponding acids RSH. The smallest S-nitrosothiol is HSNO and derives from hydrogen sulfide(HSH, H_2S). The most common physiological S-nitrosothiols are derived from the amino acid L-cysteine(Cys SH). Thus, the simplest S-nitrosothiol is S-nitroso-L-cysteine(Cys SNO). Cys SNO is a spontaneous potent donor of nitric oxide(NO) which activates soluble guanylyl cyclase to form cyclic guanosine monophosphate(c GMP). This activation is associated with multiple biological actions that include relaxation of smooth muscle cells and inhibition of platelet *** NO, Cys SNO is a short-lived species and occurs physiologically at concentrations around 1 n M in human blood. Cys SNO can be formed from Cys SH and higher oxides of NO including nitrous acid(HONO)and its anhydride(N_2O_3). The most characteristic feature of RSNO is the S-transnitrosation reaction by which the NOtgroup is reversibly transferred to another thiolate. By this way numerous RSNO can be formed such as the low-molecular-mass S-nitroso-N-acetyl-L-cysteine(SNAC) and S-nitroso-glutathione(GSNO), and the high-molecular-mass S-nitrosol-L-cysteine hemoglobin(Hb Cys SNO) present in erythrocytes and S-nitrosol-L-cysteine albumin(Alb Cys SNO) present in plasma at concentrations of the order of 200 n M. All above mentioned RSNO exert NO-related biological activity, but they must be administered intravenously. This important drawback can be overcome by lipophilic charge-free ***, we prepared the ethyl ester of SNAC, the S-nitroso-N-acetyl-L-cysteine ethyl ester(SNACET), from synthetic N-acetyl-L-cysteine ethyl ester(NACET). Both NACET and SNACET have improved pharmacological features over N-acetyl-L-cysteine(NAC) and S-nitroso-N-acetyl-L-cysteine(SNAC), respectively,including higher oral bioavailability. The performance model proposed by this study represents an innovative approach to deal with performance assessment in ATM （air traffic management）. It is based on Bayesian networks methodology, which presents several advantages but also some drawbacks as highlighted along the paper. We illustrate the main steps required for building the model and present a number of interesting results. The contribution of the paper is two-fold：（1） It presents a new methodological approach to deal with a problem which is of strategic importance for ANSPs （air navigation service providers）; （2） It provides insights on the interdependencies between factors influencing performance. Both results are considered particularly important nowadays, due to the SES （Single European Sky） performance scheme and its related target setting process.
