Tumor organoids to study gastroesophageal cancer: a primer

Gastroesophageal cancers are leading causes of cancer *** attempts at adopting molecularly based treatment approaches have been slow and ineffective even though we begin to identify specific targetable gene mutations and *** is dear that we should no longer treat all gastroesophageal cancers as a homogeneous disease,which is what we do when we use nonspecific ***,we currently cannot monitor successful gene/pathway targeting,nor understand how/when tumors develop resistance,nor predict which patients will derive maximal *** improve outcomes,we must precisely detail the heterogeneity of these tumors to then individualize cancer therapy as well as develop novel avenues to study and predict treatment effects in individual *** this end,patient-derived organoids,in which tumor cells from individual patients are grown in a Petri dish,are a new versatile system that allows for timely expandability,detailed molecular characterization,and genetic manipulation with the promise of enabling predictive assessment of treatment *** this review,we will explore the development and basic techniques for organoid generation,and discuss the current and potential future applications of this exciting technology to study the basic science of carcinogenesis and to predict/guide cancer patient care in the clinics.

For β∈ R,the authors consider the evolution system in the unknown variables u and ααttu+αxxxxu+αxxtα-(β+αxu L22)αxxu=f,αttα-αxxα-αxxtα-αxxtu=0 describing the dynamics of type III thermoelastic extensible beams,where the dissipation is entirely contributed by the second equation ruling the evolution of the thermal displacement α.Under natural boundary conditions,the existence of the global attractor of optimal regularity for the related dynamical system acting on the phase space of weak energy solutions is established.

Gene editing in plants:progress and challenges

The clustered regularly interspaced short palindromic repeat(CRISPR)-associated protein 9(Cas9)genome editing system is a powerful tool for targeted gene modifications in a wide range of species,including plants. Over the last few years, this system has revolutionized the way scientists perform genetic studies and crop breeding, due to its simplicity, flexibility, consistency and high efficiency. Considerable progress has been made in optimizing CRISPR/Cas9 systems in plants, particularly for targeted gene mutagenesis. However, there are still a number of important challenges ahead, including methods for the efficient delivery of CRISPR and other editing tools to most plants, and more effective strategies for sequence knock-ins and replacements. We provide our viewpoint on the goals, potential concerns and future challenges for the development and application of plant genome editing tools.

Scarcity of cadaveric liver donors compared with the large number of patients with liver disease has been considered the main obstacle to use liver transplantation(LT)to treat hepatic malignancies and for the development of transplant *** is known that we are experiencing a true paradigm shift in the fundamental problem of transplantology,which is none other than the imbalance between donation and graft implants(1).Because the most common indication of LT in most countries[hepatitis C virus(HCV)cirrhosis]has started to decrease in the waiting lists(2,3).One might think that,for the first time in many years in Western countries,we could find a higher supply of organs than recipients for them.

Bisphenols are a group of organic compounds used frequently in industries to make plastics,coatings,and *** compounds recently gained attention due to their potential endocrine-disrupting *** A(BPA)is the most studied compound among the bisphenol *** countries have banned the use of bisphenol A(BPA)because of its toxic effects,which have led to the invention of other bisphenol analogs,such as bisphenol F,bisphenol B,bisphenol P,and bisphenol *** studies found that bisphenol analogs may also pose endocrine-disrupting effects like BPA or can be even more ***,they may exhibit genotoxicity,cytotoxicity,and reproductive *** concentrations of bisphenol analogs have been detected in the abiotic environment and human *** some regions,the detected quantity is similar to or higher than ***,a significant research gap exists concerning the precise elucidation of the mechanism of action,adverse physiological effects,and ramifications for human health associated with bisphenol ***,a compelling need arises for more research and the accumulation of empirical data to address the consequences of bisphenol analog exposure and ensure public health *** findings resulting from initiatives in biomonitoring and varied explorations into the detrimental effects of bisphenol analogs accentuate the pressing requirement for a more exhaustive *** manuscript provides a comprehensive review of the prevalence and toxicity of bisphenol analogs,along with existing knowledge *** underscores the need to address these gaps for effective regulatory and public health strategies,contributing to a more informed approach to managing the potential risks posed by these analogs.

The advent of clustered regularly interspaced short palindromic repeat(CRISPR) has had a profound impact on plant biology, and crop improvement. In this review, we summarize the state-of-the-art development of CRISPR technologies and their applications in plants, from the initial introduction of random small indel(insertion or deletion) mutations at target genomic loci to precision editing such as base editing, prime editing and gene targeting. We describe advances in the use of class 2, types II, V, and VI systems for gene disruption as well as for precise sequence alterations, gene transcription, and epigenome control.

Methotrexate induced sprue-like syndrome

A 52 year-old male patient diagnosed of ankylosing spon-dylitis presented with an iron deficiency anemia after a ten-month treatment of methotrexate. He did not respond to treatment with oral iron not a proton pump inhibitor and an upper endoscopy was performed. The histological study of the duodenal biopsies showed villus atrophy. After removing the methotrexate, administrating intra-muscular iron and undertaking a gluten-free diet, the histological and analytical alterations progressively resolved.
