Enantioseparation of Palonosetron Hydrochloride and Its Related Enantiomeric Impurities by computer Simulation and Validation

A rapid, simple and single stereo selective high-performance liquid chromatographic (HPLc) method was developed and validated for enantiomers of palonosetron hydrochloride (PALO) and its process related chiral impurities. A computer simulating software was used for the development of chiral method. The developed method was able to separate not only the enantiomers of palonosetron hydrochloride but also its process related chiral impurities within 12 min. The chromatographic separation was carried out by normal phase chromatography using a 3 μm column of cellulose based chiral stationary phase (chiralcel-OD 250mm × 4.6mm) with a mobile phase comprised of n-hexane: ethanol: methanol: heptafluoro butyric acid: diethyl amine (70:15:15:0.05:0.1, v/v) at a flow rate of 1.0 mL/min. The effects of additive concentration as well as nature of polar organic modifier, flow rate, and temperature on enantioselectivity were investigated. The limit of detection (LOD) and limit of quantification (LOQ) of the palonosetron isomers and its related chiral impurities were found to be in the range 0.06-0.10 μg/mL and 0.14 - 0.24 μg/mL respectively. The method showed excellent linearity (R2 > 0.998) over a range of 0.14 to 1.125 μg/mL. The percentage recovery of the isomers in bulk drug samples ranged from 87.0 to 116.0.

In the present work the dynamic characteristics of Multilayer Polyurethane foam glass/fiber composite sandwich panels have been determined through Experimental Investigations. Using a Multilayer Polyurethane foam sandwich panel with rectangular cross-section, the natural frequencies, mode shapes and the damping ratio of sandwich panels were evaluated. Three types of boundary conditions were simulated namely, c-F-F-F (clamped Free-Free-Free), c-F-c-F (clam-ped-Free-clamped-Free), c-c-c-c-(clamped-clamped-clamped-clamped). Experimental modal tests were conducted on sandwich panels with available polyurethane foam of densities 56 kg/m3, 82 kg/m3and 289 kg/*** traditional “strike method” has been used to measure vibration properties. The modal characteristics of the specimens have been obtained by studying their impulse response. Each specimen has been subjected to impulses through a hard tipped hammer which is provided with a force transducer and the response has been measured through the accelerometer. The impulse and the response are processed through a computer aided FFT Analyzing test system in order to extract the modal parameters. Finite Element modeling was carried out treating the facing and core as orthotropic with different elastic constants as recommended in the literature. The experimental results were validated with FEA and were found to be in good *** results obtained through modal test on multilayer sandwich panels are presented. They indicate a significant variation in the dynamic parameters in case of multilayer sandwich panels for the same core density as compared to a monolayer sandwich panel.

目的:探讨RNA甲基化酶WTAP在肝母细胞瘤细胞中的生物学功能。方法:收集2016年8月至2020年6月同济大学附属第十人民医院手术治疗的肝母细胞瘤患者的肿瘤组织和正常对照组织13对,采用Western blot检测组织中WTAP的表达,在肝母细胞瘤细胞系中转染siRNA敲减WTAP,慢病毒包装WTAP过表达载体WTAP过表达,使用q PcR和Western blot检测敲减和过表达效率,采用ccK8实验检测细胞增殖活性,克隆形成实验检测细胞克隆形成能力,流式细胞术检测细胞凋亡。裸鼠皮下成瘤实验检测细胞在体内的成瘤能力。结果:WTAP在肝母细胞瘤组织中高表达(P=0.0007);Huh6细胞(P=0.0002)和HepG2细胞(P=0.0030)中的WTAP表达水平与QSG-7701细胞相比显著升高,Huh6细胞(PcK组的凋亡细胞百分比(10.93±0.2603)%显著低于siWTAP-1组[(16.43±0.6333)%,P=0.0013]和siWTAP-2组[(20.27±0.7535)%,P=0.0003],HepG2细胞MOcK组的凋亡细胞百分比(4.733±0.1764)%显著低于siWTAP-1组[(14.33±0.2728)%,P<0.0001]和siWTAP-2组[(15.33±0.2728)%,P<0.0001]。WTAP稳定敲减细胞株形成的移植瘤的体积(701±82.31)mm^(3)显著小于对照组[(200.2±31.59)mm^(3),P=0.0001];WTAP稳定敲减细胞株形成的移植瘤的重量(0.6368±0.08391)g显著小于对照组[(0.1356±0.03329)g,P<0.0001];WTAP稳定敲减细胞株形成的移植瘤的Ki-67阳性细胞数(108±13.05)显著少于对照组[(406±14.73),P=0.0001]。结论:WTAP在肝母细胞瘤细胞中发挥着促进增殖抑制凋亡的重要生物学功能,可能是肝母细胞瘤诊疗的一个潜在靶点。

Objective:To evaluate the anti-proliferative and apoptogenic activity of ethyl acetate extract from the leaves of Memecylon edule(EtAc-LME) in MKN-74,NUGc gastric cancer cells and non cancerous gastric mucous cells(GES-1),and to explore the mechanism of EtAc-LME induced ***:The mechanism of EtAc-LME induced apoptosis was explored by analysing the activation of pro-caspases,PARP cleavage,expression of cytochrome-c (cyt-c) was determined by western blotting,mRNA expression of Bcl-2,Bax by RT-PcR,loss of mitochondrial potential using DiOc6 dye,annexin binding assay and its influence on cell cycle arrest by flow ***:The results indicated that EtAc-LME inhibited the gastric cancer cell growth in dose-dependent manner and cytotoxicity was more towards the gastric cancer cells(NUcc and MKN-74) compared to normal gastric cells(GES-1),suggesting more specific cytotoxicity to the malignant *** expression of cyt-c and subsequent activation of caspases-3 and down regulation of Bcl-2 and loss in mitochondrial potential in EtAc-LME treated MKN-74 and NUGc cells suggested that EtAc-LME induced apoptosis by mitochondrial dependent ***:The present findings suggest that ethyl acetate extract of Memecylon edule induces apoptosis selectively in gastric cancer cells emphasizing the importance of this traditional medicine for its potential in the treatment of gastric cancer.

The trends and periodicities in the annual and seasonal temperature time series at fifteen weather stations within Ontario Great Lakes Basins have been analyzed, for the period 1941-2005, using the statistical analyses (Fourier series analysis, t-test, and Mann-Kendall test). The stations were spatially divided into three regions: northwest (NW), southwest (SW), and southeast (SE) to evaluate spatial variability in temperature. The results of the study reveal that the annual maximum mean temperature showed increasing trend for NW, and mixed trends for SW and SE regions. The variability was found to be more for northern stations as compared to southern stations for annual extreme minimum temperature. In addition, the trend slope per 100 years for the average annual extreme minimum temperature increased within the range of -0.8°c (Stratford) to 15°c (Porcupine). The seasonal analysis demonstrated that extreme maximum temperature has an increasing trend and maximum mean temperature has a decreasing trend during summer and winter. The extreme minimum temperature for winter illustrated an increasing trend (90%) with 22% statistically significant for NW region. For the SW region, the trend is also increasing (80%) for most of the temperature variables and 25% of temperature data were significantly increased in the SW region. The SE region stations showed overall very clear increasing trends (95%) for all the temperature variables. The data also showed that 47% of data were statistically significant in the SE region. The analysis of variance accounted for by trend, significant periodicities, and random component show that the pattern is similar for the percent of variance accounted for periodicities, and random component contribute dominantly for the four temperature variables and frost free days (FFD) for all three regions. Overall, the study reveals that the extreme minimum temperature is increasing annually and seasonally, with statistically significant at many stations

In this investigation, an attempt has been made to develop Al6061-Frit particulate metal matrix composites through stir casting technique using metal molds and to study the corrosion behaviour. Pre heated frit particles were added to matrix as reinforcement. Al6061 containing 2 wt% to 8 wt % in steps of 2 wt % of frit particulate composites were prepared. corrosion tests were conducted by using Potentiostat model SEP238c where 3.5% Nacl solution was used as corrodent. The corrosion rate of metal matrix composites was lower than that of matrix material Al6061 under the corrosive atmosphere for both un-heat treated and heat treated conditions.

Aim: Breast reconstruction has several beneficial effects on psychosocial well-being and quality of life. The ultimate goal has always been to create the most natural breast mound. Thus in many centers, the unilateral pedicled transverse rectus abdominis myocutaneous (TRAM) flap remains the most common technique for breast reconstruction. Our objective was to retrospectively compare the outcomes of ipsilateral and contralateral pedicle TRAM flaps. Methods: The total of 110 patients underwent unilateral breast reconstruction with pedicle TRAM flap at cancer Institute of Tehran University of Medical Science from January 1996 to June 2011. Premorbid risk factors, postoperative outcomes and demographic data were assessed. The analysis of the recordings was done by SPSS 20. Results: Out of 110 patients who were included in the study, 87 had ipsilateral and 23 contralateral pedicle TRAM flaps. The incidence of flap complications that did not require surgical intervention was 19.7% in ipsilateral and 30.4% in contralateral pedicle TRAM flap. The incidence of flap loss requiring revision was significantly higher in contralateral group (P = 0.001). Major complications were noted in 11.5% of the ipsilateral pedicle TRAM patients and 26.1% of the contralateral group (P complications were noted in 17.2% of the ipsilateral pedicle TRAM patients and in 34.8% of the contralateral group (P contralateral pedicle TRAM flaps (7.66 days vs. 10.68 days, P = 0.83). H igher c omplications were e ncountered i n c ontralateral p edicle T RAM flaps compared to ipsilateral pedicle TRAM patients (39.1% vs. 19.5%, P = 0.001).
