Co-administration of rlpaB domain of shigella with rGroEL of s. Typhi enhances the immune responses and protective efficacy against shigella infection

shigella species cause severe bacillary dysentery in humans and are associated with high morbidity and mortality. The Invasion plasmid antigen （IpaB） protein, which is conserved across all shigella spp., induces macrophage cell death and is required to invade host cells. The present study evaluates the immunogenicity and protective efficacy of the recombinant （r） domain region of IpaB （rlpaB） of s. flexneri, rlpaB was administered either alone or was co-administered with the rGroEL （heat shock protein 60） protein from s. Typhi as an adjuvant in a mouse model of intranasal immunization. The IpaB domain region （37 kDa） of s. flexneriwas amplified from an invasion plasmid, cloned, expressed in BL21 Escherichia colicells and purified. Immunization with the rlpaB domain alone stimulated both humoral and cell-mediated immune responses. Furthermore, robust antibody （IgG, IgA） and T-cell responses were induced when the rlpaB domain was co-administered with rGroEL. Antibody isotyping revealed higher IgG 1 and IgG2a antibody titers and increased interferon-gamma （IFN-γ） secretion in the co-administered group. Immunization of mice with the rlpaB domain alone protected 60%-70% of the mice from lethal infection by s. flexneri, s. boydiiand s. sonnei, whereas co-administration with rGroEL increased the protective efficacy to 80%-85%. Organ burden and histopathological studies also revealed a significant reduction in lung infection in the co-immunized mice compared with mice immunized with the rlpaB domain alone. This study emphasizes that the co-administration of the rlpaB domain and rGroEL protein improves immune responses in mice and increases protective efficacy against shigella infection. This is also the first report to evaluate the potential of the GroEL （Hsp 60） protein of s. Typhi as an adjuvant molecule, thereby overcoming the need for commercial adjuvants.

study of Propagation of Ion Acoustic Waves in Argon Plasma

The properties of small amplitude acoustic waves (IAW) in unmagnetised plasma have been discussed in detail. An experimental set up to study the propagation of IAW in argon plasma has been descried. The speed of IAW under different conditions of discharge current and pressure has been measured from the time-of flight technique. From these measurements, electron temperatures have been calculated. The results have been compared with those obtained by single probe method, and were found to be in good agreement with each other so IAW speeds can be used to calculate plasma parameters.

Predictors of spontaneous resolution of umbilical hernia in children

Objective The aims were to describe the management of umbilical hernias,to define postoperative complications,and to identify the characteristics of patients that were more likely to have spontaneous resolution of their *** All patients referred for umbilical hernia at the Children’s Hospital of Eastern Ontario from January 1990 to April 2017 were examined via retrospective chart *** resolution of umbilical hernia was examined using binary logistic *** We included 2621 patients presenting with an umbilical hernia.A total of 1587(60.5%)patients underwent surgical repair at a median age of 3.6 years(IQR 2.3-5.4).surgical complications consisted of infection(n=3),bleeding(n=3),hematoma(n=3),and anesthesia-related complications(n=3).For every one-unit increase in defect size,the odds of a spontaneous resolution of the hernia were 5%lower while controlling for prematurity and the presence of comorbidities[adjusted odds ratio(aOR)=0.95;95%confidence intervel(CI)0.93 to 0.97].Premature babies were 80%less likely to experience hernia resolution compared with non-premature babies(aOR=0.20;95%CI 0.03 to 0.74).Conclusion The odds of spontaneous resolution were lower for premature babies and were negatively correlated with defect size.

Producing environmentally stable monolayers and few-layers of hafnium disulphide （Hfs2） with a high yield to reveal its unlocked electronic and optoelectronic applications is still a challenge. Hfs2 is a layered two-dimensional material of group-IV transition metal dichalcogenides. For the first time, we demonstrate a simple and cost-effective method to grow layered belt-like nanocrystals of Hfs2 with a notably large interlayer spacing followed by their chemical exfoliation. Various microscopic and spectroscopic techniques confirm that these as-grown crystals exfoliate into single or multiple layers in a few minutes using solvent assisted ultrasonification method in N-cyclohexyl-2- pyrrolidone. The exfoliated nanosheets of Hfs2 exhibit an indirect bandgap of 1.3 eV with high stability against surface degradation. Furthermore, we demonstrate that these nanosheets hold potential for electronic applications by fabricating a field-effect transistor based on few-layered Hfs2, exhibiting a field-effect mobility of 0.95 cm2/（V.s） with a high on/off current modulation ratio of 10,000 in ambient conditions. The method is scalable and has a potential significance for both academic and industrial purposes.

Genotypic Variation for High Temperature Tolerance in Relation to Carbon Partitioning and Grain sink Activity in Wheat

Effect of high temperature tolerance in relation to carbon partitioning and grain sink activity was studied in ten genotypes of wheat (viz C 306, PBW 343, PBW 502, PBW 509, PBW 521, PBW 522, PBW 550, Inqalab, WL 711 and sonalika) under normal (November, 25.6°C during grain filling) and late planting conditions (December, 29.4°C). Contents of reducing sugars first increased till actively metabolizing stage i.e. 14 days after anthesis (DAA) in parallel with the activity of acid invertase and thereafter declined. significant reduction in total free sugars and sucrose content was observed during grain development. Acid invertase activity got induced especially in PBW 343 and C 306 under late sown conditions, whereas sucrose synthase, total amylase activities got repressed. sucrose synthase was significantly high at all stages and it predominated over invertases. Neutral invertase and total amylase showed peak activity of one week earlier in the late sown crop as compared to normal sown crop. A premature cessation of starch deposition occurred in cvs PBW 521, PBW 522 under late sown conditions due to shortening of grain filling period. Our results suggest that decline in sugar content in spite of high sucrolytic enzymes may be correlated to more utilization of assimilates over production/translocation for grain sink activity under high temperature influences. Knowledge of the mechanisms that underlie carbohydrate partitioning in diverse genotypes could be exploited for generating germplasm that can tolerate high temperature.

Lemon oil (LO), also known as Citrus limonum is a highly volatile essential oil (EO) with potential therapeutic properties like anti-oxidative, anti-proliferative, anti-fungal and anti-cancerous. However, the efficacy of LO is limited due to its physiological factors such as high volatility, poor stability (particularly sensitive to sunlight) and quick degradability upon exposure. To overcome these challenges, we formulated lemon oil loaded nanoemulsion system (LO-NE) (oil-in-water), using aqueous titration method. The formulation comprised of lemon oil (LO), Tween 80 and ethanol as oil, surfactant and co-surfactant phases respectively. The existence zone of NE was established by constructing pseudo-ternary phase diagrams using different concentrations of LO, surfactant and co-surfactant (ssub>mixsub>). The quantitative estimation of LO was performed using a high throughput gas chromatography, revealing the presence of various compounds like Limonene, Alpha-Pinene and Linalyl acetate followed by the estimation of total phenolics and flavonoid content. The characterization of LO-NE indicated the particle size of 60 ± 2.5 nm along with the polydispersity index of 0.125 and zeta potential of −14.9 mV. The size range of the NE particles dispersed in the colloidal system was further verified by TEM micrograph which shows size range between 46.2 - 104.7 nm. All the anti-oxidant assays outcomes exhibited the higher activity of LO-NE in comparison to LO alone with lower ICsub>50sub> values. The release kinetics statistical data showed that LO-NE had a sustained release and followed the Higuchi’s model in comparison to burst release of LO alone. Lastly, the stability analysis of the optimised formulation (LO-NE) and LO was estimated through antioxidant assay and subjecting them for thermodynamic stability after 6 months.
