The curren. classification.system for the recogn.tion.of taxon.mic ran.s amon. fun.i, especially at high-ran.in. level, is subjective. With the developmen. of molecular approaches an. the availability of fossil calibration.data, the use of divergen.e times as a un.versally stan.ardized criterion.for ran.in. taxa has n.w become possible. We can.therefore date the origin.of Ascomycota lin.ages by usin. molecular clock methods an. establish the divergen.e times for the orders an. families of Dothideomycetes. We chose Dothideomycetes, the largest class of the phylum Ascomycota, which con.ain. 32 orders, to establish ages at which poin.s orders have split;an. Pleosporales, the largest order of Dothideomycetes con.ain. 55 families,to establish family divergen.e times. We have assembled a multi-gen. data set（LSU, SSU, TEF1 an. RPB2）from 391 taxa represen.in. most family groups of Dothideomycetes an. utilized fossil calibration.poin.s solely from within.the ascomycetes an. a Bayesian.approach to establish divergen.e times of Dothideomycetes lin.ages. Two separated datasets were an.lysed: i) 272 taxa represen.in. 32 orders of Dothideomycetes were in.luded for the order level an.lysis, an. ii) 191 taxa represen.in. 55 families of Pleosporales were in.luded for the family level an.lysis. Our results in.icate that divergen.e times（crown.age） for most orders（20 out of 32, or 63%） are between.100 an. 220 Mya, while divergen.e times for most families（39 out of 55, or 71%）) are between.20 an. 100 Mya. We believe that divergen.e times can.provide addition.l eviden.e to support establishmen. of higher level taxa, such as families, orders an. classes. Takin. advan.age of this added approach, we can.strive towards establishin. a stan.ardized taxon.mic system both within.an. outside Fun.i. In.this study we foun. that molecular datin. coupled with phylogen.tic in.eren.es provides n. support for the taxon.mic status of two curren.ly recogn.zed orders, n.mely Bezerromycetales an. 8羟基喹啉/甲醛/间苯二酚和8羟基喹啉/甲醛/对苯二酚树脂已被合成,并研究了它们的物理化学特性。对间歇萃取和树脂柱色谱预富集铜的条件进行了最佳化。为了测定n.?)级铜,采用装有树脂的微型柱预富集,反萃取和原子吸收光谱法,并配置了流动-注射分析(FIA)管线。对于间歇预富集铜,PH大约为2.5—3是最佳的,而在流动-注射分析系统,能使用的PH范围较宽(约2—3.5)。Cu(Ⅱ)与n.(Ⅱ)或Cu(Ⅱ)与Pb(Ⅱ)二元混合物的分离,没有出现任何交叉污染。为了定量提取铜,在流动-注射分析系统,2cm长的柱及2mL/min.流速是足够的;50μL0.1M盐酸可以定量淋洗铜。

目的:证实印度草药方n./CAL/06的甲醇提取物对卵巢切除大鼠的抗骨质疏松作用。方法:雌性Sprague-Drawley大鼠在无菌状态下行双侧卵巢切除术后被分为5组(n.10)。检测n./CAL/06的甲醇提取物(200和400mg/kg口服)的抗骨质疏松作用并以雷洛昔芬(5.4mg/kg口服)作为标准对照药,治疗时间为90d。实验前后测量各组大鼠的体质量、卵巢质量、骨质量、骨矿物质含量、骨强度,血清及尿液中的钙、磷含量及血清碱性磷酸酶含量。电镜下观察大鼠股骨外径、长度及厚度。结果:双侧卵巢切除术后,大鼠骨质量、骨矿物质含量及骨强度均有下降;电镜观察到卵巢切除大鼠股骨骨质疏松、多孔、破碎,且骨长度及厚度均有所下降。与假手术组相比,卵巢切除大鼠的血清钙、磷及碱性磷酸酶含量均有所升高,体质量及脂肪含量升高,子宫质量降低。n./CAL/06的甲醇提取物(200和400mg/kg口服)治疗90d能够剂量依赖性地改变大鼠因卵巢切除所引起的上述各项指标变化,并将骨质量、骨矿物质含量、骨强度,血清钙、磷及碱性磷酸酶含量,体质量及脂肪含量恢复至正常水平。此外,经n./CAL/06的甲醇提取物(200和400mg/kg口服)治疗的大鼠与卵巢切除模型组大鼠相比,电镜下股骨骨质孔形成减少,骨密度增加。结论:本实验的结果证实了n./CAL/06的甲醇提取物具有很好的抗骨质疏松作用,可用于骨质疏松症的治疗。
