Immunogenicity and efficacy of recombinant 78 kDa antigen of Leishmania donovani formulated in various adjuvants against murine visceral leishmaniasis

Objective: To analyze the protective efficacy of recombinant 78 kDa antigen of Leishmania donovani in combination with two adjuvants, that is, cationic liposomes or MPL-A against visceral leishmaniasis in BALB/c mice. Methods: The genomic DNA of promastigotes was isolated and 583 bp of T cell epitopes of gene encoding 78 k Da was amplified using specific primers. The amplified gene was cloned into p ET28 c, transformed into Escherichia coli BL21(DE3) and got expressed after IPTG induction. The recombinant protein was then purified using Ni-NTA and named r78. Three groups of mice were immunized with 10 μg of r78 plus MPL-A, r78 encapsulated in positively charged liposomes and control animals immunized with PBS. Two booster doses were given with the respective vaccine at an interval of 2 weeks each. Mice were challenged with 1×107 Leishmania promastigotes and sacrificed on different post infection/challenge days. Results: Immunization with r78 along with MPL-A and liposomeencapsulated r78 brought a significant reduction in parasite load. In comparison to the infected controls, the parasite load declined by 96.2% in mice immunized with r78 plus MPL-A and 97.23% in animals immunized with liposome-encapsulated r78. The immunized animals also exhibited profound DTH response. The serum antibody responses increased from 15 to 90 days post infection/challenge. Immunized animals showed greater IgG2 a levels and lesser Ig G1 levels in comparison to the infected controls. The splenocytes from immunized mice were cultured, stimulated with r78 and analyzed for cytokine profile. The levels of IL-2 and IFN-γ were greater in immunized animals as compared to control mice. Conclusions: The study proves that r78 in combination with suitable adjuvants is a potential vaccine candidate and may be instrumental in control of visceral leishmaniasis.

Anthropogenic activities are likely to cause global warming. Global warming will affect marine productivityand thus food availability for marine organisms. Both the seawater temperature and food availability affect benthicforaminifera, the most abundant marine calcareous microorganisms. Therefore, a laboratory culture experiment wasconducted on Cymbaloporetta plana to understand the coupled effect of temperature(25°C, 27°C, 30°C) and food(102±5,203±9, 305±14, 407±18, 508±23 diatom cells). At all temperatures, growth increased with increase in food. The specimenssubjected to 27°C temperature attained the maximum growth and those subjected to 25°C were the smallest. All specimensfed with 508±23 cells reproduced, 0 irrespective of temperature. Of all the specimens without food, 40% at 27°C and 20% at25°C reproduced, whereas specimens subjected to 30°C did not reproduce. The 100% rate of reproduction at 27°C suggeststhat it is the optimum temperature for growth and reproduction. All the specimens at 30°C and without additional food,died, whereas at 25°C and 27°C, the mortality rate was 80% and 60%, respectively. The limited growth and reproductionwith decreasing food clearly indicates that food availability also controls the growth and reproduction of shallow waterbenthic foraminifer C. plana. Therefore, warming coupled with decreasing productivity is likely to severely affect thegrowth and abundance of shallow water benthic foraminifera.

Development of cost-effective, highly reproducible non-conventional fabrication techniques for anisotropic metal nanostructures is essential to realizing potential applications of plasmonic devices, photonic devices, and surface enhanced Raman scattering （SERS） phenomenon based sensors. This report highlights the fabrication of nanotriangle arrays via nanoimprinting to overcome difficulties in creating large-area SERS active substrates with uniform, reproducible Raman signals. Electron beam lithography of anisotropic nanostructures, formation of arrays of nanotriangles in silicon and the transfer of triangular shapes to polymethylmethacrylate （PMMA） sheets via nanoimprinting have not been reported elsewhere. The reuse of silicon masters offers potential for production of low cost SERS substrates. The SERS activity and reproducibility of nanotriangles are illustrated and a consistent average enhancement factor of up to -2.9 × 1011, which is the highest value reported for a patterned SERS substrate, is achieved.

The N-terminal EF-hand calcium-binding proteins 1–3(NECAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one EF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NECAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NECAB2 interacts with predominantly striatal G-protein-coupled receptors, while NECAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NECAB proteins exhibit distinct subcellular localizations: NECAB1 is found in the nucleus and cytosol, NECAB2 resides in endosomes and the plasma membrane, and NECAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NECAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NECAB proteins by controlling the availability of protein–protein interaction domains situated between the EF hands and the antibiotic biosynthesis monooxygenase domain. A highly reproducible and efficient in vitro shoot regeneration system was developed in a potential medicinal plant, Albizia lebbeck using root explants. Root explants from 15 day-old-aseptic seedlings were cultured on Murashige and Skoog （MS） medium supplemented with different concentrations （0.5, 2.5, 5.0, 7.5 and 10.0 μM） of 6-Benzyladenine （BA）, kinetin （kn）, 2-Isopentenyl adenine （2-iP） singly as well as in combination with α-Naphthalene acetic acid （NAA）（0.1, 0.5, 1.0, 1.5 and 2.0 μM）. The highest rate of shoot multiplication （16.0 ± 1.87 for the average shoot number and 5.16 ± 0.38 cm for shoot length） was achieved on MS medium supplemented with 7.5 μM BA and 0.5 μM NAA. The effects of medium type, medium strength, pH and subculture on shoot induction and proliferation were also tested. An average of 21.6±2.87 shoots per explants could be obtained following this protocol. Rooting was achieved on microshoots using half strength MS medium with 2.0 μM Indole-3-butyric acid （IBA） after four weeks of culture. The in vitro raised healthy plantlets were successfully established in earthen pots containing garden soil and grown in greenhouse with 〉80% survival rate.

采用在炭纤维束的内部浸润熔融沥青的传统方法制备得到的炭/炭(C/C)复合材料难以实现细旦化水平,这是因为单束丝中含有数千根丝,经浸渍后沥青附着,从而引起沥青局部渗出,以及因不均匀沥青稳定性而导致微观结构不均匀。本文通过稳定的中间相沥青和分散的短切炭纤维混合、热压和炭化过程制备出细旦化水平的C/C复合材料。探讨了工艺参数对中间相沥青残炭率的影响。通过残炭率/工艺时间、残炭率!(表观密度/实密度)来优化工艺参数。结果表明,通过此法得到了细旦化水平的中间相沥青与炭纤维复合材料。中间相沥青的残炭率随加热升温速率、沥青/炭纤维质量比的增加而降低;随热压压力而增加。在高压力和高质量比下,发生沥青局部渗出。通过调控得到最佳的C/C复合材料制备工艺参数为:热压压力15 MPa,升温速率0. 2℃/min及沥青/炭纤维质量比1∶1。
