Lung oncogenesis relies on intracellular cysteine to overcome oxidative *** tumor types,including non-small cell lung cancer(NSCLC),upregulate the system x-c cystine/glutamate antiporter(xCT)through overexpression of the cystine transporter SLC7A11,thus sustaining intracellular cysteine levels to support glutathione *** factor erythroid 2-related factor 2(NRF2)serves as a master regulator of oxidative stress resistance by regulating SLC7A11,whereas Kelch-like ECH-associated protein(KEAP1)acts as a cytoplasmic repressor of the oxidative responsive transcription factor *** in KEAP1/NRF2 and p53 induce SLC7A11 activation in *** cystine is crucial in supplying the intracellular cysteine levels necessary to combat oxidative *** in cystine availability lead to iron-dependent lipid peroxidation,thus resulting in a type of cell death called *** inhibitors of xCT(either SLC7A11 or GPX4)induce ferroptosis of NSCLC cells and other tumor *** cystine uptake is impaired,the intracellular cysteine pool can be sustained by the transsulfuration pathway,which is catalyzed by cystathionine-B-synthase(CBS)and cystathionine g-lyase(CSE).The involvement of exogenous cysteine/cystine and the transsulfuration pathway in the cysteine pool and downstream metabolites results in compromised CD8^(+)T cell function and evasion of immunotherapy,diminishing immune response and potentially reducing the effectiveness of immunotherapeutic *** is a previously unrecognized form of regulated cell *** NSCLCs driven by EGFR,ALK,or KRAS,selective inhibitors induce pyroptotic cell death as well as *** targeted therapy,the mitochondrial intrinsic apoptotic pathway is activated,thus leading to the cleavage and activation of ***,gasdermin E is activated,thus leading to permeabilization of the cytoplasmic membrane and cell-lytic pyroptosis(indicated by characte

Chemical proﬁles of non-invasive biological material,such as feces,have great potential to study elusive animals or those with low population ***,we use a metabolomic approach to evaluate Neotropical mustelids as a biological model to describe the diversity of the metabolites present in fecal samples,as well as to evaluate the potential of chemical proﬁles for taxonomic *** collected fecal samples from captive individuals of 5 species of mustelids occurring in Brazil and analyzed them by liquid chromatography coupled to high-resolution mass *** 200 compounds have been annotated;“bile acids,alcohols and derivatives”was the most expressive class in the metabolome of all the *** successfully discriminated 3 taxonomic groups:1—tayra(Eira barbara);2—otters(Lontra longicaudis and Pteronura brasiliensis;1);and 3—grisons(Galictis vittata and Galictis cuja).Several compounds seemed to be associated with food intake and the digestive process,while others were found for theﬁrst time in Neotropical *** concluded that mustelids show high metabolome diversity and that species-speciﬁc identiﬁcation through metabolomic proﬁles is possible,thus contributing to the development and implementation of additional non-invasive approaches in the study of mustelids.

Dear Editor,Colorectal cancer(CRC)is the third most common cancer globally[1],even though a large proportion of those cancers would be preventable by *** those with a family history(FH)of CRC,it is commonly recommended to start screening at younger ages,e.g.,at age 40 or 10 years younger than the age at diagnosis of the youngest affected first-degree relative[2–5].Even within the group of those with a FH,risk of CRC is not homogeneous and depends on factors such as the number of affected relatives and the age at which the relatives were diagnosed with CRC[6,7],lifestyles,and genetic background ***,these metrics except genetic factors may change over lifetime,which limits their use for defining starting ages of *** aim of this study was to evaluate whether a polygenic risk score(PRS),which combines information from CRC-related risk variants[8]and genetically determined sex(2 constant and established CRC risk factors),could effectively contribute to enhanced risk *** also aimed to determine if it could assist in defining risk-adapted starting ages for CRC screening,even in individuals with a FH of *** assessment was performed using the wellestablished metric of risk advancement periods(RAPs)[9],which measures the impact of an exposure on the relation of age to disease.

A Simulation Approach Including Underresolved Scales for Two-Component Fluid Flows in Multiscale Porous Structures

.In this study,we develop computational models and a methodology for accurate multicomponent flow simulation in underresolved multiscale porous structures[1].It is generally impractical to fully resolve the flow in porous structures with large length-scale differences due to the tremendously high computational *** flow contributions from underresolved scales should be taken into account with proper physics modeling and simulation *** precomputed physical properties such as the absolute permeability,K_(0),the capillary pressure-saturation curve,and the relative permeability,K_(r),in typical resolved porous structures,the local fluid force is determined and applied to simulations in the underresolved regions,which are represented by porous *** this way,accurate flow simulations in multiscale porous structures become *** evaluate the accuracy and robustness of this method,a set of benchmark test cases are simulated for both single-component and two-component flows in artificially constructed multiscale porous *** comparisons with analytic solutions and results with much finer resolution resolving the porous structures,the simulated results are ***,in all cases,the results successfully show high accuracy with proper input of K_(0),capillary pressure,and K_(r).Specifically,imbibition patterns,entry pressure,residual component patterns,and absolute/relative permeability are accurately captured with this approach.

Artificial neural network with a cross-validation approach to blast-induced ground vibration propagation modeling

Given their technical and economic advantages,the application of explosive substances to rock mass excavation is widely ***,because of serious environmental restraints,there has been an increasing need to use complex tools to control environmental effects due to blast-induced ground *** the present study,an artificial neural network(ANN)with k-fold cross-validation was applied to a dataset containing 1114 observations that was obtained from published results;furthermore,quantitative and qualitative parameters were considered for ground vibration amplitude *** best ANN model obtained has a maximum coefficient of determination of 0.840 and a mean absolute error of 5.59 and it comprises 17 input parameters,12 neurons in a one-layer hidden layer,and a sigmoid transfer *** with the traditional models,the model obtained using the proposed methodology demonstrated better generalization ***,the proposed methodology offers an ANN model with higher prediction ability.

Slurry ice as an alternative cooling medium for fish harvesting and transportation:Study of the effect on seabass flesh quality and shelf life

The objective of the study was to investigate the efficiency of slurry ice during harvesting and transportation of European sea bass(Dicentrarchus labrax)to retain flesh quality and extend shelf life,compared with conventional flake *** was slaughtered and transported in different mixtures of slurry ice and conventional flake ice(C:slaughtered and transported in 100%flake ice-Control samples,SC:slaughtered in 100%slurry ice and transported in 100%flake ice,S50:slaughtered and transported in 50%slurry ice-50%flake ice,S100:slaughtered and transported in 100%slurry ice)and subsequently stored under controlled isothermal conditions at 0℃for shelf life modelling and flesh quality evaluation(proteolytic enzymes).The replacement of conventional flake ice with slurry ice as a slaughtering method led to improved quality stability during subsequent refrigerated storage and shelf life extension,in terms of microbial growth,flesh quality and sensory degradation of *** on microbial growth,the shelf life of C samples was found to be 19 days,whereas the shelf life of S50/S100 and SC was 21 and 25 days,respectively,showing that the replacement of flake ice with slurry ice resulted in 2–6 days shelf life extension of whole sea bass stored at 0℃.The use of slurry ice at slaughter and flake ice in transportation was accompanied by low activities and late peaks of all four enzymes that is expected to lead to delayed proteolytic degradation and extended freshness.

Spermatogenesis involves extremely marked cellular, genetic and chromatin changes resulting in the generation of the highly specialized sperm cell. Proteomics allows the identification of the proteins that compose the spermatogenic cells and the study of their function. The recent developments in mass spectrometry （MS） have markedly increased the throughput to identify and to study the sperm proteins. Catalogs of thousands of testis and spermatozoan proteins in human and different model species are becoming available, setting up the basis for subsequent research, diagnostic applications and possibly the future development of specific treatments. The present review intends to summarize the key genetic and chromatin changes at the different stages of spermatogenesis and in the mature sperm cell and to comment on the presently available proteomic studies.

Given a significative class F of commutative rings, we study the precise conditions under which a commutative ring R has an S-envelope. A full answer is obtained when F is the class of fields, semisimple commutative rings or integral domains. When .F is the class of Noetherian rings, we give a full answer when the Krull dimension of R is zero and when the envelope is required to be epimorphic. The general problem is reduced to identifying the class of non-Noetherian rings having a monomorphic Noetherian envelope, which we conjecture is the empty class.
