High arsenic(As)and chromium(Cr)concentrations are currently receiving attention because of their negative effects on the environment and human *** inhabiting contaminated environments have developed resistance mechanisms against the toxicity of these ***,members of the bacterial genus Micrococcus have been isolated from different toxic metal-contaminated environments;however,knowledge concerning its resistance mechanisms to As and Cr toxicity remains *** luteus strains(An24,Mh,NE2TL6,and NE2TTS4)were isolated from the endosphere and soil of two heavy metal-contaminated sites in Mexico to identify differences in the resistance mechanisms by the *** *** strains were resistant to As(As^(3+)and As^(5+)),chromate,dichromate,cobalt,copper,nickel,and *** analysis indicated that the heavy metal-resistant strains(An24,Mh,NE2TL6,and NE2TTS4)could be assigned to the *** group and had more heavy metal-resistant genes(transporters,chaperones,and enzymes)compared to reference strains of the *** group,*** NCTC 2665^(T)and Micrococcus endophyticus JCM 16951^(T).The resistant bacteria were able to biotransform As^(3+)and As^(5+)through a carbon source-dependent *** biotransformation of As5+was potentially carried out in the cytoplasm through a thioredoxin-dependent pathway,which may be coupled with biosorption.A qualitative analysis of organic acids(OAs)identified a change in the OA profile of the metal-resistant strains that was As-or *** genomic and phenotypic findings suggest that the four *** group strains evaluated in the current study have developed resistance mechanisms that may enable their survival in contaminated sites.

Olive (Olea europaea L.) tree is one of the most extensive and important agricultural crop in Mediterranean countries due to its beneficial health and nutritional properties and its high economic value. Currently, olive tree constitutes the sixth most important cultivated plant in the world, spreading from the Mediterranean region of origin to new production areas such as Australia, South and North America and South Africa. However, the mobilization processes of storage materials i.e. reserve proteins during seed germination, which are largely involved in essential physiological process including plant growth and development, remain poorly understood. Morphometric and immunohistochemistry analyses of protein bodies contained in olive seed storage tissues, cotyledon and endosperm, were performed by using different microscopy techniques, including light (bright-field and fluorescence) microscopy and transmission electron microscopy. Furthermore, we used legumin-like proteins (11S-type globulins) as a molecular marker to study the mobilization of reserve proteins from PBs of cotyledons at germinating seedling stages by using immunofluorescence assays. Results demonstrated that cotyledon and endosperm are characterized by distinct PBs populations containing legumin-like proteins, distinctly discriminated by the number of PBs per cell and tissue, size, immunofluorescence and histochemical staining. These features reflect differential PBs biogenesis during development and maturation processes in olive seed tissues endosperm and cotyledon, in relation to proteins (polypeptides) final composition, SSPs processing and/or packaging during seed maturation. Adolescent obesity is a health problem worldwide. Drug therapy for obesity in children is only acceptable if done within the framework of a comprehensive weight loss, prevention of comorbidities, including health education, changes in lifestyles, diet and exercise. These pharmacological treatments indicate that mainly young patients did not respond to standard measures and developing complications. The role of bariatric surgery, with or without the use of drugs is subject to further research to elucidate its role among the various treatment options for these patients. Currently being studied a wide range of drugs to those included gastrointestinal hormones. Scientific information regarding the use of exenatide in obese patients without diabetes is still limited. The use of exenatide in adolescents with morbid obesity may represent a therapeutic complement invaluable, effective and free of complications, and prevents future bariatric surgery.

OBJECTIVES Elderly patients show a higher incidence of ischemic and bleeding events after percutaneous transluminal coron-ary intervention(PCI).We sought to investigate outcomes in elderly patients treated with antithrombotic strategy guided by bleeding and ischemic risks after revascularization with last generation everolimus-eluting stent(EES).METHODS Prospective multicenter registry including patients over 75 years revascularized with EES and antithrombotic ther-apy guided by clinical presentation,PCI complexity and PRECISE DAPT ***-primary safety endpoints were:(1)composite of cardiac death,myocardial infarction and stent thrombosis and;(2)bleeding(BARC 2-5).Primary efficacy endpoint was target lesion revascularization.A matched group of patients revascularized with current drug-eluting stents and no such tailored antith-rombotic therapy was used as *** Finally,1064 patients were included in SIERRA-75 cohort,80.8±4.2 years,36.6%women,71%acute coronary syn-dromes(ACS)and 53.6%complex ***-primary safety endpoint of major adverse cardiovascular events was met in 6.2%,co-primary safety endpoint of bleeding in 7.8%and primary efficacy endpoint of TKLR in 1.5%.The multivariable adjusted model showed no significant association of the prescribed short/long dual antiplatelet therapy(DAPT)durations with any endpoint suggesting a well tailored *** stent thrombosis reported in the subgroup with 1-3 months DAPT *** compared to control group,bleeding BARC 2-5 was significantly lower in SIERRA-75 group(7.4%vs.10.2%,P=0.04)as well as the net safety-efficacy endpoint(14.3%vs.18.5%,P=0.02).CONCLUSIONS In elderly population,the application of this risks-adjusted antithrombotic protocol after revascularization with last generation EES seems to be associated with an improved prognosis in terms of ischemic and bleeding outcomes.

AIM To develop a fast, low-cost diagnostic strategy to identify single point mutations in highly variable genomes such as hepatitis C virus(HCV).METHODS In patients with HCV infection, resistance-associated amino acid substitutions within the viral quasispecies prior to therapy can confer decreased susceptibility to direct-acting antiviral agents and lead to treatment failure and virological relapse. One such naturally occurring mutation is the Q80 K substitution in the HCV-NS3 protease gene, which confers resistance to PI inhibitors, particularly simeprevir. Low-cost, highly sensitive techniques enabling routine detection of these single point mutations would be useful to identify patients at a risk of treatment failure. Light Cycler methods, based on real-time PCR with sequencespecific probe hybridization, have been implemented in most diagnostic laboratories. However, this technique cannot identify single point mutations in highly variable genetic environments, such as the HCV genome. To circumvent this problem, we developed a new method to homogenize all nucleotides present in a region except the point mutation of interest. RESULTS Using nucleotide-specific probes Q, K, and R substitutions at position 80 were clearly identified at a sensitivity of 10%(mutations present at a frequency of at least 10% were detected). The technique was successfully applied to identify the Q80 K substitution in 240 HCV G1 serum samples, with performance comparable to that of direct Sanger sequencing, the current standard procedure for this purpose. The new method was then validated in a Catalonian population of 202 HCV G1-infected individuals. Q80 K was detected in 14.6% of G1 a patients and 0% of G1 b in our setting. CONCLUSION A fast, low-cost diagnostic strategy based on real-time PCR and fluorescence resonance energy transfer probe melting curve analysis has been successfully developed to identify single point mutations in highly variable genomes such as hepatitis C virus.
