Objective:to study the canine echinococcosis by coproantigen ELISA ***:During the present investigation experimental infection was established using evaginated worms of Echinococcus granulosus(***).to check cross reactivity two pups were infected with taenia hydatigena(***).In order to detect the presence of antigen,hyperimmune sera were raised against excretory-secretory products of adult worms *** granulosus. Faecal sample collected either from experimentally infected pups or from other sources were heated at 70℃to detect heat stable soluble ***:Pups harbouring less than 104 worms showed negative *** collected from 14 days onwards from experimentally infected animals harbouring more than 104 worms showed positive *** maximum positive samples were detected in samples collected from in and around slaughter house and the least number of samples were detected positive maintained by dog ***:the affinity purified IgG exhibited promising results for detection of canine echinococcosis by indirect ELISA.

AIM: to investigate inpatient length of stay(LOS), complication rates, and readmission rates for sacral fracture patients based on operative ***: All patients who presented to a large tertiary care center with isolated sacral fractures in an 11-year period were included in a retrospective chart review. Operative approach(open reduction internal fixation vs percutaneous) was noted, as well as age, gender, race, and American Society of Anesthesiologists' score. Complications included infection, nonunion and malunion, deep venous thrombosis, and hardware problems; 90-d readmissions were broken down into infection, surgical revision of the sacral fracture, and medical complications. LOS was collected for the initial admission and readmission visits if applicable. Fisher's exact and non-parametric t-tests(Mann-Whitney U tests) were employed to compare LOS, complications, and readmissions between open and percutaneous ***: Ninety-four patients with isolated sacral fractures were identified: 31(30.4%) who underwentopen reduction and internal fixation(ORIF) vs 63(67.0%) who underwent percutaneous fixation. there was a significant difference in LOS based on operative approach: 9.1 d for ORIF patients vs 6.1 d for percutaneous patients(P = 0.043), amounting to a difference in cost of $13590. ten patients in the study developed complications, with no significant difference in complication rates or reasons for complications between the two groups(19.4% for ORIF patients vs 6.3% for percutaneous patients). Eight patients were readmitted, with no significant difference in readmission rates or reasons for readmission between the two groups(9.5% percutaneous vs 6.5% ORIF).CONCLUSION: there is a significant difference in LOS based on operative approach for sacral fracture patients. Given similar complications and readmission rates, we recommend a percutaneous approach.

AIM to assess nutritional recovery,particularly regarding feeding jejunostomy tube(FJt)utilization,following upper gastrointestinal resection for malignancy. MEtHODS A retrospective review was performed of a prospectively-maintained database of adult patients who underwent esophagectomy or gastrectomy(subtotal or total)for cancer with curative intent,from January 2001 to June 2014. Patient demographics,the approach to esophagectomy,the extent of gastrectomy,FJt placement and utilization at discharge,administration of parenteral nutrition(PN),and complications were evaluated. All patients were followed for at least ninety days or until *** the 287 patients underwent upper GI resection,comprised of 182 esophagectomy(n=107 transhiatal,58.7%; n=56 Ivor-Lewis,30.7%)and 105 gastrectomy [n=63 subtotal(SG),60.0%; n=42 total(tG),40.0%]. 181 of 182 esophagectomy patients underwent FJt,compared with 47 of 105 gastrectomy patients(99.5% vs 44.8%,P t had undergone tG(n=39,92.9% vs n=8 SG,12.9%,P th of stay was similar between esophagectomy and gastrectomy groups(14.7 d vs 17.1 d,P=0.076). Upon discharge,87 esophagectomy patients(48.1%)were taking enteral feeds,with 53(29.3%)fully and 34(18.8%)partially dependent. Meanwhile,20 of 39 tG patients(51.3%)were either fully(n=3,7.7%)or partially(n=17,43.6%)dependent on tube feeds,compared with 5 of 8 SG patients(10.6%),all of whom were partially dependent. Gastrectomy patients were significantly less likely to be fully dependent on tube feeds at discharge compared to esophagectomy patients(6.4% vs 29.3%,P=0.0006). PN was administered despite FJt placement more often following gastrectomy than esophagectomy(n=11,23.4% vs n=7,3.9%,P=0.0001). FJt-specific complications requiring reoperation within 30 d of resection occurred more commonly in the gastrectomy group(n=6),all after tG,compared to 1 esophagectomy patient(12.8% vs 0.6%,P=0.0003). In this article we discuss the challenges of delivering a high quality transition care. A good understanding of the adolescent needs with good communication between transition care physicians and the patient is essential for good continuity of care. Despite availability of several guidelines, one model doesn't fit all and any transition service development should be determined by the local need and available healthcare facilities.

Objective：to investigate the effects of E7080 and N5-（1-iminoethyl）-L-ornithine dihydrochloride （L-NIO）on colorectal cancer alone and in ***：Ht29 colorectal cancer cell line from Sap Institute was ***-time cell analysis （xCELLigence system） was performed to determine the effects of E7080 and L-NIO on colorectal cell *** apoptosis was determined with Annexin v staining,and the effect of agents on angiogenesis was determined with chorioallantoic membrane （CAM） ***：We found that E7080 has a strong antiproliferative effect with an half maximum inhibition of concentration （IC50） value of 5.60×10-8 mol/*** it has been observed that E7080 showed antiangiogenic and apoptotic effects on Ht29 colorectal cancer *** scores of E7080 were 1.2,t.0 and 0.6 for 100,10 and 1 nmol/L E7080 concentrations,***,apoptosis has been detected in 71％ of Ht29 colorectal cancer cells after administration of 100 nmol/L E7080 which may indicate strong apoptotic *** administration of L-NIO alone did not show any effect,but the combination of E7080 with L-NIO increased the antiproliferative,antiangiogenic and apoptotic effects of ***：results of this study indicate that E7080 may be a good choice in treatment of colorectal *** the increased effects of E7080 when combined with L-NIO raise the possibility to use a lower dose of E7080 and therefore avoid/minimize the side effects observed with E7080.

Crohn’s disease(CD)is an inflammatory bowel disease characterized by immunemediated flares affecting any region of the intestine alternating with remission *** CD,the ileum is frequently affected and about one third of patients presents with a pure ileal ***,the ileal type of CD presents epidemiological specificities like a younger age at onset and often a strong link with smoking and genetic susceptibility *** of these genes are associated with Paneth cell dysfunction,a cell type found in the intestinal crypts of the ***,a Western-type diet is associated in epidemiological studies with CD onset and increasing evidence shows that diet can modulate the composition of bile acids and gut microbiota,which in turn modulates the susceptibility of the ileum to ***,the interplay between environmental factors and the histological and anatomical features of the ileum is thought to explain the specific transcriptome profile observed in CD ***,both immune response and cellular healing processes harbour differences between ileal and non-ileal *** together,these findings advocate for a dedicated therapeutic approach to managing ileal ***,interventional pharmacological studies have failed to clearly demonstrate distinct response profiles according to disease ***,the high rate of stricturing disease in ileal CD requires the identification of new therapeutic targets to significantly change the natural history of this debilitating disease.

AIM to characterize the role of apolipoprotein B100(apoB 100) in hepatitis C viral(HCv) infection. MEtHODS In this study, we utilize a gene editing tool, transcription activator-like effector nucleases(tALENs), to generate human hepatoma cells with a stable genetic deletion of APOB to assess of apoB in HCv. Using infectious cell culture-competent HCv, viral pseudoparticles, replicon models, and lipidomic analysis we determined the contribution of apoB to each step of the viral lifecycle. We further studied the effect of mipomersen, an FDAapproved antisense inhibitor of apoB 100, on HCv using in vitro cell-culture competent HCv and determined itsimpact on viral infectivity with the tCID50 method. reSULtS We found that apo B100 is indispensable for HCv infection. Using the JFH-1 fully infectious cell-culture competent virus in Huh 7 hepatoma cells with tALENmediated gene deletion of apoB(APOB KO), we found a significant reduction in HCv RNA and protein levels following infection. Pseudoparticle and replicon models demonstrated that apo B did not play a role in HCv entry or replication. However, the virus produced by APOB KO cells had significantly diminished infectivity as measured by the tCID-50 method compared to wildtype virus. Lipidomic analysis demonstrated that these virions have a fundamentally altered lipidome, with complete depletion of cholesterol esters. We further demonstrate that inhibition of apoB using mipomersen, an FDA-approved anti-sense oligonucleotide, results in a potent anti-HCv effect and significantly reduces the infectivity of the virus. CONCLUSION Apo B is required for the generation of fully infectious HCv virions, and inhibition of apo B with mipomersen blocks HCv. targeting lipid metabolic pathways to impair viral infectivity represents a novel host targeted strategy to inhibit HCv.

AIM: to investigate the effects of dextrans of various molecular weights (Mw) during a 12 h cold storage time-course on energetics, histology and mucosal infiltration of fluorescein isothiocyanate (FItC)-dextran. MEtHODS: Rodent intestines were isolated and received a standard University of Wisconsin vascular flush followed by intraluminal administration of a nutrientrich preservation solution containing dextrans of varying Mw: Group D1, 73 kdal; Group D2, 276 kdal; Group D3, 534 kdal; Group D4, 1185 kdal; Group D5, 2400 kdal. reSULtS: Using FItC-labeled dextrans, fluorescent micrographs demonstrated varying degrees of mucosal infiltration; lower Mw (groups D1-D3: 73-534 kdal) dextrans penetrated the mucosa as early as 2 h, whereas the largest dextran (D5: 2400 kdal) remained captive within the lumen and exhibited no permeability even after 12 h. After 12 h, median injury grades ranged from 6.5 to 7.5 in groups D1-D4 (73-1185 kdal) representing injury of the regenerative cryptal regions and submucosa; this was in contrast to group D5 (2400 kdal) which exhibited villus denudation (with intact crypts) corresponding to a median injury grade of 4 (P tissue energetics reflected a strong positive correlation between Mw and adenosine triphosphate (r 2 = 0.809), total adenylates (r 2 = 0.865) and energy charge (r 2 = 0.667).CONCLUSION: Our data indicate that dextrans of Mw > 2400 kdal act as true impermeant agents during 12 h ischemic storage when incorporated into an intraluminal preservation solution.

Aim:Ferroptosis is a non-apoptotic form of cell death caused by lethal lipid *** small molecule ferroptosis inducers(FINs)have been reported,yet little information is available regarding their interaction with the AtP-binding cassette(ABC)transporters P-glycoprotein(P-gp,ABCB1)and *** thus sought to characterize the interactions of FINs with P-gp and ABCG2,which may provide information regarding oral bioavailability and brain penetration and predict drug-drug ***:Cytotoxicity assays with ferroptosis-sensitive A673 cells transfected to express P-gp or ABCG2 were used to determine the ability of the transporters to confer resistance to FINs;confirmatory studies were performed in OvCAR8 and NCI/ADR-reS *** ability of FINs to inhibit P-gp or ABCG2 was determined using the fluorescent substrates rhodamine 123 or purpuin-18,***:P-gp overexpression conferred resistance to FIN56 and the erastin derivatives imidazole ketone erastin and piperazine erastin.P-gp-mediated resistance to imidazole ketone erastin and piperazine erastin was also reversed in UO-31 renal cancer cells by CRISPR-mediated knockout of *** FINs ML-162,GPX inhibitor 26a,and PACMA31 at 10µM were able to increase intracellular rhodamine 123 fluorescence over 10-fold in P-gp-expressing MDR-19 *** inhibitor 26a was able to increase intracellular purpurin-18 fluorescence over 4-fold in ABCG2-expressing R-5 ***:Expression of P-gp may reduce the efficacy of these FINs in cancers that express the transporter and may prevent access to sanctuary sites such as the *** ability of some FINs to inhibit P-gp and ABCG2 suggests potential drug-drug interactions.
