This paper presents a methodology based on computational intelligence techniques for classification of hydrological cycles that can infer the change in the physico-chemical parameters and metals from the water of a reservoir in the Amazon. The methodology initially consists in perform a pre-processing the data to select the most relevant variables of the samples. After that, we compared two different machine learning classifiers, namely SVM (support vector machine) and ANN (artificial neural network). The automatic model selection is made to choose the parameters of the classifiers. The results indicate that the support vector machine classifier using radial basis function or polynomial kernel exhibited superior results to ANN in terms of overall accuracy and robustness. The SVM classifier accuracy (89.1%) can be considered satisfactory, since there is a great variability of physico-chemical parameters and metals in the hydrological cycles and in the different ecosystems where are the sampling station.

Macrophage plasticity is critical for normal tissue repair following *** pathologic states such as diabetes,macrophage plasticity is impaired,and macrophages remain in a persistent proinflammatory state;however,the reasons for this are ***,using single-cell RNA sequencing of human diabetic wounds,we identified increased JMJD3 in diabetic wound macrophages,resulting in increased inflammatory gene ***,we report that in wound healing,JMJD3 directs early macrophage-mediated inflammation via JAK1,3/STAT3 ***,in the diabetic state,we found that IL-6,a cytokine increased in diabetic wound tissue at later time points post-injury,regulates JMJD3 expression in diabetic wound macrophages via the JAK1,3/STAT3 pathway and that this late increase in JMJD3 induces NFκB-mediated inflammatory gene transcription in wound macrophages via an H3K27me3 ***,RNA sequencing of wound macrophages isolated from mice with JMJD3-deficient myeloid cells(Jmjd3f/fLyz2Cre+)identified that the STING gene(Tmem173)is regulated by JMJD3 in wound *** limits inflammatory cytokine production by wound macrophages during ***,in diabetic mice,its role changes to limit wound repair and enhance *** finding is important since STING is associated with chronic inflammation,and we found STING to be elevated in human and murine diabetic wound macrophages at late time ***,we demonstrate that macrophage-specific,nanoparticle inhibition of JMJD3 in diabetic wounds significantly improves diabetic wound repair by decreasing inflammatory cytokines and *** together,this work highlights the central role of JMJD3 in tissue repair and identifies cell-specific targeting as a viable therapeutic strategy for nonhealing diabetic wounds.

Conventional clear-cut timber harvest is a widespread industrial practice across the Pacific Northwest;however,information regarding how these practices impact soil microbial community structure at the regional scale is *** evidence of consistent and substantial impact of harvest on soil microbial functional profiles across the region(despite a range of environmental conditions),the objective of this study was to determine the extent to which harvest also influences the structure of prokaryotic and fungal soil microbial communities,and how generalized these trends are throughout the geographic *** soil samples were collected one year before and after harvest across nine second-growth Douglas-fir forests in the Pacific *** community DNA was extracted from the soils,and high-throughput targeted gene sequencing of the 16 S r RNA gene for prokaryotes and the internal transcribed spacer(ITS)gene for fungi was *** diversity was consistently and significantly higher after harvest;it was moderately so for fungal communities(+14.6%),but only marginally so for prokaryotic communities(+2.0%).Similarly,on average,a greater proportion of the variation in the community structure of fungi(20.1%)at each site was associated with forest harvest compared to that of prokaryotes(13.2%).Overall,the greatest influence of timber harvest on soil microbial communities appeared to be a relative depletion of ectomycorrhizal fungi,with a concomitant enrichment of saprotrophic *** the short-term responses of soil microbial communities across the region,particularly those of tree root-associated symbionts,may aid our understanding of the role soil microbial communities play in ecological succession.

Survival in changing and challenging environments requires an organism to efficiently obtain and use its resources. Due to their sessile nature, it is particularly critical for plants to dynamically optimize their meta- bolism. In plant primary metabolism, metabolic fine-tuning involves feed-back mechanisms whereby the output of a pathway controls its input to generate a precise and robust response to environmental changes. By contrast, few studies have addressed the potential for feed-back regulation of secondary metabolism. In Arabidopsis, accumulation of the defense compounds glucosinolates has previously been linked to genetic variation in the glucosinolate biosynthetic gene AOP2. AOP2 expression can increase the transcript levels of two known regulators （MYB28 and MYB29） of the pathway, suggesting thatAOP2 plays a role in positive feed-back regulation controlling glucosinolate biosynthesis. We generated mutants affecting AOP2, MYB28/29, or both. Transcriptome analysis of these mutants identified a so far unrecognized link between AOP2 and jasmonic acid （JA） signaling independent of MYB28 and MYB29. Thus, AOP2 is part of a regulatory feed-back loop linking glucosinolate biosynthesis and JA signaling and thereby allows the glucosinolate pathway to influence JA sensitivity. The discovery of this regulatory feed-back loop provides insight into how plants optimize the use of resources for defensive metabolites.

Background: The availability of an anesthesiologist is often a limiting factor in the number of operations that can be performed by International Medical Surgical Response Teams (IMSuRT). Because emergency physicians (EPs) possess skills in airway control, management of moderate and deep sedation, and ventilator management, we propose that with proper training in general anesthesia, EPs can serve as anesthetists for IMSuRT with anesthesiologist supervision. Methods: During a 10-week period, a board-certified EP administered general anesthesia to 60 patients prior to a surgical medical mission trip. The breakdown of surgical cases was: 11 orthopedic, 2 genitourinary, 20 ear, nose, and throat, 8 obstetrics and gynecological, 13 general surgery, and 6 vascular. A simplified protocol for induction, maintenance, and emergence was adhered to for all cases. Results: Fourteen orthopedic cases using general anesthesia were performed in a one-week period in Haiti. These cases involved open reduction and internal fixation (ORIF), hemiarthoplasty, hardware removal, tendon transfer and external fixation of fractured bone. Conclusion: We demonstrate the feasibility of a model curriculum to train EPs in the basics of anesthesia. The EP can safely and effectively deliver general anesthesia for major cases on surgical medical mission trips under the auspices of an anesthesiologist in an austere environment.

Characterizing Wyoming Ranching Operations: Natural Resource Goals, Management Practices and Information Sources

Spanning 12 million hectares, Wyoming rangelands produce food and provide other vital ecosystem services. However, the decision-making process of the ranchers who steward these lands is complex and poorly understood. In cooperation with the Wyoming Stock Growers Association (WSGA)—a predominant agricultural organization in the state—we asked WSGA producer members about their goals, ranching operation characteristics, and management practices via a mail survey. A total of 307 ranchers (50%) responded to the survey. Livestock production and forage production were the top management goals, with ecosystem characteristics that support these goals (e.g., soil health, water quality) tied for second. Survey respondents’ ranches had a median size of 4220 hectares, but ranged up to 185,000 hectares;71% of operations included public land and 60% included private leased land. The majority of reporting operations grazed cow-calf pairs (91%), with a median of 260 pairs per ranch. Most survey respondents managed grazing by moving 1 - 5 herds of livestock (84%) among two or more pastures (92%) after three months of grazing or less (87%). Most operations (74%) included other resource use activities, with extractive recreation (e.g., hunting;55%), conventional energy development (23%), and other agricultural production (20%) most common. Survey respondents primarily got information about grazing management from other ranchers (97%), although they preferred to receive information through print publications (69%). Wyoming ranching operations are diverse, which may represent a challenge for policy makers designing programs and incentives to increase production of food and ecosystem services. However, efforts that focus on livestock and forage production and supporting ecosystem functions are likely to find synergies with ongoing management goals and strategies.
